    • Humans host complex microbial communities believed to contribute to health maintenance and, when in imbalance, to the development of diseases. Determining the microbial composition in patients and healthy controls may thus provide novel therapeutic targets. For this purpose, high-throughput, cost-effective methods for microbiota characterization are needed. We have employed 454-pyrosequencing of a hyper-variable region of the 16S rRNA gene in combination with sample-specific barcode sequences which enables parallel in-depth analysis of hundreds of samples with limited sample processing. In silico modeling demonstrated that the method correctly describes microbial communities down to phylotypes below the genus level. Here we applied the technique to analyze microbial communities in throat, stomach and fecal samples. Our results demonstrate the applicability of barcoded pyrosequencing as a high-throughput method for comparative microbial ecology.

    • The essential catalytic radical of Class-I ribonucleotide reductase is generated and delivered by protein R2, carrying a dinuclear metal cofactor. A new R2 subclass, R2c, prototyped by the Chlamydia trachomatis protein was recently discovered. This protein carries an oxygen-activating heterodinuclear Mn(II)/Fe(II) metal cofactor and generates a radical-equivalent Mn(IV)/Fe(III) oxidation state of the metal site, as opposed to the tyrosyl radical generated by other R2 subclasses. The metal arrangement of the heterodinuclear cofactor remains unknown. Is the metal positioning specific, and if so, where is which ion located? Here we use X-ray crystallography with anomalous scattering to show that the metal arrangement of this cofactor is specific with the manganese ion occupying metal position 1. This is the position proximal to the tyrosyl radical site in other R2 proteins and consistent with the assumption that the high-valent Mn(IV) species functions as a direct substitute for the tyrosyl radical.

  • Turbulence in the Vicinity of a Rotating Cylinder in a Quiescent Fluid

    • An experimental and numerical investigation of the turbulence field in the vicinity of a rotating cylinder in a quiescent fluid has been carried out. Radial distributions of mean velocity and non-vanishing Reynolds stress components have been measured using hot-wire technique, and corresponding components have been calculated with a low-Reynolds number second- moment closure model. From the measurements it can be concluded the turbulence field is established in the present set up at a Reynolds number which is roughly twenty times larger than earlier measurements have shown. Comparisons between calculations and measurements yield some deviations. However, a qualitatively good agreement is obtained. In the paper, different methods for the determination of the friction velocity are also described and discussed.

  • The greenhouse gas emissions of an electrified vehicle combined with renewable fuels: Life cycle assessment and policy implications

    • A life cycle assessment is presented for a current vehicle’s greenhouse gas impact when using a combination of electrification and renewable fuels. Three degrees of electrification are considered: a hybrid electric vehicle, a plug-in hybrid electric vehicle, and a battery-electric vehicle. These are combined with fuels with various degrees of renewable content, representing a fossil fuel, a first-generation biofuel and a second-generation biofuel. For charging, the 2020 European electricity mix is used and compared with an electricity mix of low greenhouse-gas intensity. Renewable fuels are found to have a greater potential to reduce the life-cycle greenhouse gas emissions than a low carbon electricity mix. The results are discussed in terms of the supply potential for renewable fuels on the fleet level. It is found that plug-in hybrid vehicles may enable the automotive sector to reach more ambitious climate goals than battery-electric vehicles. An assessment is also made of how the life cycle greenhouse gas emissions compare with the emissions as measured by current policy instruments. The discrepancies indicate that current climate policy instruments are inadequate for minimizing the automotive sector’s climate impact and suggestions for improvements are made.

    • The folate metabolism enzyme MTHFD2 (methylenetetrahydrofolate dehydrogenase/cyclohydrolase) is consistently overexpressed in cancer but its roles are not fully characterized, and current candidate inhibitors have limited potency for clinical development. In the present study, we demonstrate a role for MTHFD2 in DNA replication and genomic stability in cancer cells, and perform a drug screen to identify potent and selective nanomolar MTHFD2 inhibitors; protein cocrystal structures demonstrated binding to the active site of MTHFD2 and target engagement. MTHFD2 inhibitors reduced replication fork speed and induced replication stress followed by S-phase arrest and apoptosis of acute myeloid leukemia cells in vitro and in vivo, with a therapeutic window spanning four orders of magnitude compared with nontumorigenic cells. Mechanistically, MTHFD2 inhibitors prevented thymidine production leading to misincorporation of uracil into DNA and replication stress. Overall, these results demonstrate a functional link between MTHFD2-dependent cancer metabolism and replication stress that can be exploited therapeutically with this new class of inhibitors. Helleday and colleagues describe a nanomolar MTHFD2 inhibitor that causes replication stress and DNA damage accumulation in cancer cells via thymidine depletion, demonstrating a potential therapeutic strategy in AML tumors in vivo.

  • Sparing land for biodiversity at multiple spatial scales

    • A common approach to the conservation of farmland biodiversity and the promotion of multifunctional landscapes, particularly in landscapes containing only small remnants of non-crop habitats, has been to maintain landscape heterogeneity and reduce land-use intensity. In contrast, it has recently been shown that devoting specific areas of non-crop habitats to conservation, segregated from high-yielding farmland (“land sparing”), can more effectively conserve biodiversity than promoting low-yielding, less intensively managed farmland occupying larger areas (“land sharing”). In the present paper we suggest that the debate over the relative merits of land sparing or land sharing is partly blurred by the differing spatial scales at which it is suggested that land sparing should be applied. We argue that there is no single correct spatial scale for segregating biodiversity protection and commodity production in multifunctional landscapes. Instead we propose an alternative conceptual construct, which we call “multiple-scale land sparing,” targeting biodiversity and ecosystem services in transformed landscapes. We discuss how multiple-scale land sparing may overcome the apparent dichotomy between land sharing and land sparing and help to find acceptable compromises that conserve biodiversity and landscape multifunctionality.

  • Silica exposure increases the risk of stroke but not myocardial infarction : A retrospective cohort study

    • INTRODUCTION: Work-related exposure to silica is a global health hazard that causes diseases such as silicosis. Some studies have also reported that silica exposure is linked to elevated cardiovascular disease mortality. However, these diagnoses have not been investigated in detail and there have been few studies on morbidity. The aim of this study is to examine morbidity and mortality from different cardiovascular diseases among silica-exposed Swedish foundry workers.METHODS: Historical and contemporary measurements (1968-2006) of respiratory silica exposure were matched to job categories, individual foundries, and 4 time periods (1968-1979, 1980-1989, 1990-1999, 2000-2006) using a mixed model. Morbidity and mortality data for the studied cohorts were matched against the General Population Registry. Statistical analyses were performed with SPSS and STATA, and the data were stratified by age, gender, and year.RESULTS: Mortality from cardiovascular disease (SMR 1.3; 95% CI 1.2-1.4) and stroke (SMR 1.6, 95% CI 1.2-2.1) was significantly elevated among the studied population. The cohort also exhibited significantly elevated morbidity from stroke (SIR 1.34; 95% CI 1.2-1.5) but not myocardial infarction. The mean age at the time of first morbidity from stroke was 64 years, with 36% of the cases occurring before the age of 60.CONCLUSIONS: Swedish foundry workers exposed to respirable silica exhibit elevated morbidity and mortality from stroke, but not from myocardial infarction. Our results also suggest a relationship between silica exposure and morbidity from stroke at a younger age than the general population.

  • Gender and Programming : A Case Study

    • Our study highlights the effects of gender-related learning styles on a computer programming course at introductory level of engineering education. It was triggered by the observation of statistically relevant under-achievements among female students over the years. We try to identify concrete differences in motivation/learning styles between genders and prove that lack of previous relevant computer experience is not the only factor to blame. The paper analyzes the situation at a LTH course from the point of view of the conflicts outlined in the literature. Data from “before-starting” questionnaires and follow ups for subsequent evaluations expose significant gender differences. Analysis of the course materials and interviews with students reveals problems of constructive alignment and discouragements to the motivation of novice programmers. We investigated several pedagogical methods to adapt teaching and evaluation in order to increase all students’ competence and at the same time reduce the gap between genders. Our key recommendation is to make the separation between the teaching of algorithms and the teaching of the specific language syntax clearer. It is our belief that good teaching of engineering subjects will enhance learning for all students
