  • Nuclear Spent Fuel Parameter Determination using Multivariate Analysis of Fission Product Gamma Spectra

    • In this paper, we investigate the application of multivariate data analysis methods to the analysis of gamma spectroscopy measurements of spent nuclear fuel (SNF). Using a simulated irradiation and cooling of nuclear fuel over a wide range of cooling times (CT), total burnup at discharge (BU) and initial enrichments (IE) we investigate the possibilities of using a multivariate data analysis of the gamma ray emission signatures from the fuel to determine these fuel parameters. This is accomplished by training a multivariate analysis method on simulated data and then applying the method to simulated, but perturbed, data.We find that for SNF with CT less than about 20 years, a single gamma spectrum from a high resolution gamma spectrometer, such as a high-purity germanium spectrometer, allows for the determination of the above mentioned fuel parameters.Further, using measured gamma spectra from real SNF from Swedish pressurized light water reactors we were able to confirm the operator declared fuel parameters. In this case, a multivariate analysis trained on simulated data and applied to real data was used.

  • Fed-batch production assessment of a tetravalent bispecific antibody : A case study on piggyBac stably transfected HEK293 cells

    • The transition from preclinical biological drug development into clinical trials requires an efficient upscaling process. In this context, bispecific antibody drugs are particularly challenging due to their propensity to form aggregates and generally produce low titers. Here, the upscaling process for a tetravalent bispecific antibody expressed by a piggyBac transposon-mediated stable HEK293 cell pool has been evaluated. The project was performed as a case study at Testa Center, a non-GMP facility for scale-up testing of biologics in Sweden, and encompassed media adaptation strategies, fed-batch optimization and a novel antibody purification technology. The cell pool was adapted to different culture media for evaluation in terms of cell viability and titers compared to its original Expi293 Expression Medium. These parameters were assessed in both sequential stepwise adaption and direct media exchanges. By this, a more affordable medium was identified that did not require stepwise adaptation and with similar titers and viability as in the Expi293 Expression Medium. Fed-batch optimizations resulted in culture densities reaching up to 20 x 106 viable cells/mL with over 90 % viability 12 days postinoculum, and antibody titers three times higher than corresponding batch cultures. By implementing a novel high-speed protein A fiber technology (Fibro PrismA) with a capture residence time of only 7.5 s, 8 L of supernatant could be purified in 4.5 h without compromising the purity, structural integrity and function of the bispecific antibody. Results from this study related to medium adaptation and design of fed-batch protocols will be highly beneficial during the forthcoming scale-up of this therapeutic antibody.

  • Global blood miRNA profiling unravels early signatures of immunogenicity of Ebola vaccine rVSVΔG-ZEBOV-GP

    • The vectored Ebola vaccine rVSVΔG-ZEBOV-GP elicits protection against Ebola Virus Disease (EVD). In a study of forty-eight healthy adult volunteers who received either the rVSVΔG-ZEBOV-GP vaccine or placebo, we profiled intracellular microRNAs (miRNAs) from whole blood cells (WB) and circulating miRNAs from serum-derived extracellular vesicles (EV) at baseline and longitudinally following vaccination. Further, we identified early miRNA signatures associated with ZEBOV-specific IgG antibody responses at baseline and up to one year post-vaccination, and pinpointed target mRNA transcripts and pathways correlated to miRNAs whose expression was altered after vaccination by using systems biology approaches. Several miRNAs were differentially expressed (DE) and miRNA signatures predicted high or low IgG ZEBOV-specific antibody levels with high classification performance. The top miRNA discriminators were WB-miR-6810, EV-miR-7151-3p, and EV-miR-4426. An eight-miRNA antibody predictive signature was associated with immune-related target mRNAs and pathways. These findings provide valuable insights into early blood biomarkers associated with rVSVΔG-ZEBOV-GP vaccine-induced IgG antibody responses.

  • Beyond professional terms – the family metaphor in staff descriptions of their relations to young people in secure

    • One premise for the organisation of residential care for youthis that staff are expected to relate to each young personindividually, but also to the group of young people as a whole.The relational interplay between staff and placed youths insecure unit care is fundamentally based on asymmetry, withinteractions taking place in a context of confinement. The aimhere is to explore how staff working in secure institutionalcare for youths in Sweden understand and describe theirrelationships with youth in terms that extend beyondprofessionalism, and especially their use of the familymetaphor. Fifty-three interviews with staff were analysed in atwo-step qualitative analysis, which generated three themesthat highlighted staff narratives focusing on descriptions ofparenting, sibling relationships, and closeness without usingthe family metaphor. One conclusion is that despite an overallshift away from the family metaphor, in the direction offraming residential care in professional terms, the familyconcept seems to sit quite well even in an environment withambitions to provide professional care. The family metaphormay not be the cornerstone of care, but it is eminentlypresent.

  • Countertransference and therapeutic holding : Working with a perpetrator of domestic violence

    • In the context of the treatment of perpetrators of violence, psychodynamic discourse has often focused on individual aggression and other related emotions and mental phenomena, such as anger, rage, cruelty, sadism and brutality. Psychodynamic theory has been less explicit, however, about how to address an individual’s actual violent acts within the psychotherapeutic setting. This article aims to address this gap. It illustrates how psychoanalytic theory – most notably through Winnicott, Fonagy and Glasser – can be used to inform thinking about, and interventions with, men who use violence against their children and partners, using a clinical case that describes psychotherapeutic work with a perpetrator of domestic violence. The article concludes that the psychotherapist’s countertransference is an important tool in working with men who use violence, and highlights the importance of the therapist’s capacity for impulse regulation, emotional understanding, and handling of his or her own aggression.

  • Emotional adjustments to violent situations at secure units for adolescents : A staff perspective

    • Secure units for adolescents are emotion-filled places. This paper examines how secure unit staff are socialized to match emotions to violent situations in a dynamic social context according to different feeling rules. The analysis applies theories about emotional culture as well as feeling rules. The article builds on 53 semi-structural interviews with staff at three secure units for detained boys and girls run by the Swedish National Board of Institutional Care. The findings illustrate different ways in which staff adjust their feelings in violent situations by both breaking and following rules. Results are discussed with respect to three themes: (1) emotional adjustments tuned to co-workers; (2) emotional adjustments tuned to self; and (3) emotional adjustments tuned to youth behaviour. The results show the importance of taking emotions seriously in daily work, as this makes it possible to visualize how staff emotionally socialize into their organization. Due to a lack of education and professional identity among staff, the socialization process is highly relevant, as it is primarily perceptions of youth and co-workers that control and develop feeling rules for staff.

    • This article studies a special type of verbal coordination in Swedish, pseudocoordination, which involves two verbs or verb phrases typically combined with och ‘and’. Although it consists of two verbs with a conjunction between them, pseudocoordination in many ways behaves like a subordinating complex-predicate construction. In the literature, pseudocoordination is treated more or less explicitly as a unitary phenomenon, but previous diachronic studies of some types of pseudocoordination point in slightly different directions regarding the origin of pseudocoordinations with different first verbs. It has been claimed that they originate in (canonical) verbal coordinations, but it has also been claimed that they are originally hypotactic structures in which the conjunction is a covert infinitival marker (both are typically pronounced in the same way today). To the best of our knowledge, the gå och V ‘walk/go and V’ construction has not previously been studied both diachronically and corpus-based. By examining gå och V in historical corpora, we therefore add to the historical knowledge of pseudocoordination with gå in Swedish. The gå och V construction is particularly interesting since it has multiple meanings and since it, unlike the bulk of Swedish pseudocoordinations, also has subordination counterparts in Modern Swedish.

    • This article explores conditions for implementing shared decision-making (SDM) in coordinated individual care planning (CIP) with individuals with complex mental health needs. SDM in CIP are described as central, although such user centred collaboration still remains to be realised. Research underlines the need for a changed way of working, where user expertise is valued and a balance of power is promoted. The aim of the present study is to investigate the conditions for implementing SDM in connection with CIP for and with people with mental illness. To better understand the context and conditions that can promote such an implementation, altogether 15 participants were interviewed in three regions in Sweden within the scope of a stakeholder analysis. Both hindering and supporting factors were identified with respect to an implementation process, such as staff turnover, differences in work culture and committed leadership. Further focus should be directed specifically towards professionals working more closely with CIP and towards in-depth analysis of the construct of culture in terms of implementation processes.

  • Handling fear among staff : violence and emotion in secure units for adolescents

    • The Swedish National Board of Institutional Care (SiS) has reported a rise in violent incidents between staff and youth placed in secure units. This paper explores how secure unit staff narratively position themselves and youth when speaking about violence, and how staff describe the emotional impact violence has on them. The paper takes two theoretical starting points. First is the concept of emotional labour, including feeling rules, emphasizing that staff work with the emotions of others whilst also being expected to control their own. Second is the idea that the interaction among staff shapes various social representations and positionings. Five focus groups were conducted with staff (n = 27) who worked with both boys and girls at three different secure units in Sweden. The empirical data was first processed through narrative analysis and then by an interaction analysis. The results, presented as four excerpts within four themes, reveal collegial processes of narrative helping that steer group members to find emotional positions when talking about experienced violence, i.e., to find appropriate feeling rules. Furthermore, despite counter-narratives expressed by participants in the focus group, a representation of youth as violent persists. Finally, emotional labour seems to involve working with one’s own feelings and controlling the emotions of youth, not the opposite. The results suggest how important it is for staff to recognize youth from different templates and that violence can take various forms, and furthermore, that it is essential to make the emotion of fear visible in this context.
