  • CUBN Is a Gene Locus for Albuminuria

    • Identification of genetic risk factors for albuminuria may alter strategies for early prevention of CKD progression, particularly among patients with diabetes. Little is known about the influence of common genetic variants on albuminuria in both general and diabetic populations. We performed a meta-analysis of data from 63,153 individuals of European ancestry with genotype information from genome-wide association studies (CKDGen Consortium) and from a large candidate gene study (CARe Consortium) to identify susceptibility loci for the quantitative trait urinary albumin-to-creatinine ratio (UACR) and the clinical diagnosis microalbuminuria. We identified an association between a missense variant (I2984V) in the CUBN gene, which encodes cubilin, and both UACR (P = 1.1 x 10(-11)) and microalbuminuria (P = 0.001). We observed similar associations among 6981 African Americans in the CARe Consortium. The associations between this variant and both UACR and microalbuminuria were significant in individuals of European ancestry regardless of diabetes status. Finally, this variant associated with a 41% increased risk for the development of persistent microalbuminuria during 20 years of follow-up among 1304 participants with type 1 diabetes in the prospective DCCT/EDIC Study. In summary, we identified a missense CUBN variant that associates with levels of albuminuria in both the general population and in individuals with diabetes.

  • A comparison between nailfold capillaroscopy patterns in adulthood in juvenile and adult-onset systemic sclerosis: A EUSTAR exploratory study

    • Objective: Qualitative capillaroscopy patterns in juvenile- and adult-onset systemic sclerosis (SSc) were studied in adulthood using data from the EULAR Scleroderma Trials and Research (EUSTAR) database. Methods: Data collected between June 2004 and April 2013 were examined with focus on capillaroscopy. In this retrospective exploratory study, series of patients with juvenile-onset SSc were matched with series of adult-onset SSc having the same gender and autoantibody profile. Results: 30 of 123 patients with juvenile-onset and 2108 of 7133 with adult-onset SSc had data on capillaroscopy. Juvenile-onset SSc showed scleroderma pattern more frequently than adult-onset SSc (93.3% and 88%). The OR was 2.44 and 95% Cl 0.57-10.41. An active scleroderma pattern was present in 58% of juvenile- and 61% of adult-onset SSc. The OR was 0.91 and 95% Cl 0.28-2.93. The late scleroderma pattern was present in 61% of juvenile- and 55.5% of adult-onset SSc. The OR was 1.06 and 95% Cl 0.34-3.56. Conclusion: This is the first exploratory study on the comparison of capillaroscopy between juvenile- and adult-onset SSc in adulthood. Juvenile-onset SSc had an increase prevalence of sderoderma pattern, but a similar distribution of the three patterns was suggested. Further studies are needed to define this issue.   • Nailfold capillaroscopy in systemic sclerosis: Data from the EULAR scleroderma trials and research (EUSTAR) database

    • Objective: The aims of this study were to obtain cross-sectional data on capillaroscopy in an international multi-center cohort of Systemic Sclerosis (SSc) and to investigate the frequency of the capillaroscopic patterns and their disease-phenotype associations. Methods: Data collected between June 2004 and October 2011 in the EULAR Scleroderma Trials and Research (EUSTAR) registry were examined. Patients' profiles based on clinical and laboratory data were obtained by cluster analysis and the association between profiles and capillaroscopy was investigated by multinomial logistic regression. Results: 62 of the 110 EUSTAR centers entered data on capillaroscopy in the EUSTAR database. 376 of the 2754 patients (13.65%) were classified as scleroderma pattern absent, but non-specific capillary abnormalities were noted in 55.48% of the cases. Four major patients' profiles were identified characterized by a progressive severity for skin involvement, as well as an increased number of systemic manifestations. The "early" and "active" scleroderma patterns were generally observed in patients with mild/moderate skin involvement and a low number of disease manifestations, while the "late" scleroderma pattern was found more frequently in the more severe forms of the disease. Conclusion: These data indicate the importance of capillaroscopy in SSc management and that capillaroscopic patterns are directly related to the extent of organ involvement. (C) 2013 Elsevier Inc.   • Genome-wide meta-analysis for Alzheimer's disease cerebrospinal fluid biomarkers.

    • Amyloid-beta 42 (Aβ42) and phosphorylated tau (pTau) levels in cerebrospinal fluid (CSF) reflect core features of the pathogenesis of Alzheimer's disease (AD) more directly than clinical diagnosis. Initiated by the European Alzheimer & Dementia Biobank (EADB), the largest collaborative effort on genetics underlying CSF biomarkers was established, including 31 cohorts with a total of 13,116 individuals (discovery n=8074; replication n=5042 individuals). Besides the APOE locus, novel associations with two other well-established AD risk loci were observed; CR1 was shown a locus for Aβ42 and BIN1 for pTau. GMNC and C16orf95 were further identified as loci for pTau, of which the latter is novel. Clustering methods exploring the influence of all known AD risk loci on the CSF protein levels, revealed 4 biological categories suggesting multiple Aβ42 and pTau related biological pathways involved in the etiology of AD. In functional follow-up analyses, GMNC and C16orf95 both associated with lateral ventricular volume, implying an overlap in genetic etiology for tau levels and brain ventricular volume.

    • This paper reviews the technical aspects related to the long-term field exposure practice in marine environments, based on the return of experiences of major marine exposure sites in world-wide scope. The long-term exposure practice helps both the research on durability mechanisms of structural concretes under real environments and the calibration of durability models to support the life-cycle management of concrete structures. The presentation of the field exposure data can be categorized into the information relevant to exposure sites, the data related to the exposed materials and specimens, the information of environmental actions, and the data related to the performance of materials. A standardized presentation of these data can help the efficiency of data sharing and exploitation. The exploitation of exposure data employs various models to represent the chloride ingress and the induced corrosion risk of the embedded steel bars. There are needs for models addressing the strong environment-material interactions, and simple yet reliable durability indicators for engineering use. The design and operation of exposure stations need the careful choice of exposure sites and specimens, the appropriate scheme for monitoring and inspection of exposed specimens, the systematic recording and management of exposure data, and the regular maintenance of exposure facilities. The support of exposure data for life-cycle management is demonstrated through the durability planning of a real project case. The good practice of long-term field exposure is summarized in the end.

    • This guideline prescribes the technical requirements for the design and operation of long-term exposure sites in marine environments. The technical content includes the design of exposure site, the monitoring of environmental conditions, the monitoring of exposed specimens, the data management and the maintenance of exposure facilities. The design of exposure site covers the choice of exposure sites, the exposure infrastructure and facilities, exposure specimens and their placing and protection. Then, the guideline defines the environmental factors to be monitored and the corresponding monitoring methods. For the exposed specimens, the guideline specifies the target performance to monitor, the sampling, the testing and the reporting of exposure results. The data management deals with such aspects as the data storage, data transmission and the data sharing. At the end, the guideline gives the principles for the maintenance of the exposure facilities. This guideline summarizes the best practice of long-term exposure in marine environments, and it is expected to support the normalization of exposure practice and to generate more added values from this practice.

  • Genetic associations at 53 loci highlight cell types and biological pathways relevant for kidney function

    • Reduced glomerular filtration rate defines chronic kidney disease and is associated with cardiovascular and all-cause mortality. We conducted a meta-analysis of genome-wide association studies for estimated glomerular filtration rate (eGFR), combining data across 133,413 individuals with replication in up to 42,166 individuals. We identify 24 new and confirm 29 previously identified loci. Of these 53 loci, 19 associate with eGFR among individuals with diabetes. Using bioinformatics, we show that identified genes at eGFR loci are enriched for expression in kidney tissues and in pathways relevant for kidney development and transmembrane transporter activity, kidney structure, and regulation of glucose metabolism. Chromatin state mapping and DNase I hypersensitivity analyses across adult tissues demonstrate preferential mapping of associated variants to regulatory regions in kidney but not extra-renal tissues. These findings suggest that genetic determinants of eGFR are mediated largely through direct effects within the kidney and highlight important cell types and biological pathways.

  • Model for the assessment of bond in corroded steel ribbed bars

    • The aim of the present paper is to further validate a model proposed for assessing bond strength in corroded and non-corroded steel bars. The model was obtained by applying multiple linear regression analysis to an initial database obtained from literature containing over 650 bond tests: 372 with corroded steel bars. In this paper, a second stage for further validating the model has been conducted with new database consisting of 131 new tests of bond with corroded steel bars resulting in a final database of 500 tests of bond with corroded reinforcing steel bars. In this model, bond strength is considered as an average stress on the nominal surface of a straight length of a bar over the bond length. The corrosion effect is considered using the cross-section loss (% Cor) considered as uniform corrosion along the bonded length of the bar. In addition to the cross-section loss, corrosion effect in bond is additionally considered with an additional variable that implicitly includes the effect of cracking depending on the range of cross section loss, bond condition and presence of transverse reinforcement. In the paper, the former formulation proposed in is compared with the formulation adapted with rounded coefficients for the former database, the new database and the combined database with different statistical criteria to compare the accuracy of the bond strength predictions obtained with the model. Finally, the predictions of the model show a good fitting with the experimental results with the new database and also have a low scatter. This is showing its utility in the safety assessment of bond strength in reinforced concrete members.

  • Power-Aware Virtual Network Function Placement and Routing using an Abstraction Technique

    • The Network Function Virtualization (NFV) is very promising for efficient provisioning of network services and is attracting a lot of attention. NFV can be implemented in commercial off-the-shelf servers or Physical Machines (PMs), and many network services can be offered as a sequence of Virtual Network Functions (VNFs), known as VNF chains. Furthermore, many existing network devices (e.g., switches) and collocated PMs are underutilized or over-provisioned, resulting in low power-efficiency. In order to achieve more energy efficient systems, this work aims at designing the placement of VNFs such that the total power consumption in network nodes and PMs is minimized, while meeting the delay and capacity requirements of the foreseen demands. Based on existing switch and PM power models, we formulate an Integer Linear Programming (ILP) model to find the optimal solution. We also propose a heuristic based on the concept of Blocking Islands (BI), and a baseline heuristic based on the Betweenness Centrality (BC) property of the graph. Both heuristics and the ILP solutions have been compared in terms of total power consumption, delay, demands acceptance rate, and computation time. Our simulation results suggest that BI-based heuristic is superior compared with the BC-based heuristic, and very close to the optimal solution obtained from the ILP in terms of total power consumption and demands acceptance rate. Compared to the ILP, the proposed BI-based heuristic is significantly faster and results in 22% lower end-to-end delay, with a penalty of consuming 6% more power in average.
