  • Tailored for decision – Knowing your target group prior to adaptation

    • Life cycle thinking is but one perspective - if at all considered -, in everyday business decisions throughout the organization; in the selection of suppliers, in the strategy of new product ranges, and, in what information is chosen to be highlighted to the customers. Tradeoffs are constantly made between e.g. environment, quality, price and other company goals. Before any successful adaptation and visualization of life cycle information, it is important for internal life cycle experts to identify and understand how other functions of the company perceive and value life cycle information in their specific working situations.To get a better understanding of these internal users of life cycle information, life cycle experts in four multinational companies (Akzo Nobel, Vattenfall, Volvo Cars, Volvo Group) have joined forces with researchers in life cycle management and behavioral science to create a graphical map of how life cycle information is spread and used in different parts of an organization. The aim of the map is to be used as a basis for discussions and recommendations on how to tailor life cycle information in order to support decision making throughout a company.The map is constructed by combining a) inventories on how quantitative data seeks its way to internal users through databases, reports and KPIs, with b) qualitative interviews on goal framing and decision weights of e.g. environmental and economic information. As a result, the map illustrates both the “physical” flows of life cycle information and the “cognitive logics” of this information for different users (e.g. how values, attitudes and norms influence the target groups’ likelihood of including life cycle information in their decision processes).Based on the map, each company can identify and discuss who the main users of life cycle information are and what premises for life cycle thinking these users have: In what decision making situation is, or can, life cycle information be used? How is the information understood? What other sources of information and rationales for decisions are used in parallel to, or in conflict with, LCA-results?Initial analyses on the usefulness of the map point to a better understanding of how life cycle experts can tailor information for decisions in different parts of the company, as well as on its usefulness in illustrating to people outside of the environmental departments the widespread use of life cycle information that already exist in the company. The latter is not least important for creating an understanding in how the organization respond to ongoing external pressure to focus more on a life cycle approach, e.g. new requirements in ISO 14001, new EU Directives on public procurement and current EU work to establish a common LCA methodology.

  • LDO proteins and Vac8 form a vacuole-lipid droplet contact site to enable starvation-induced lipophagy in yeast

    • Lipid droplets (LDs) are fat storage organelles critical for energy and lipid metabolism. Upon nutrient exhaustion, cells consume LDs via gradual lipolysis or via lipophagy, the en bloc uptake of LDs into the vacuole. Here, we show that LDs dock to the vacuolar membrane via a contact site that is required for lipophagy in yeast. The LD-localized LDO proteins carry an intrinsically disordered region that directly binds vacuolar Vac8 to form vCLIP, the vacuolar-LD contact site. Nutrient limitation drives vCLIP formation, and its inactivation blocks lipophagy, resulting in impaired caloric restriction-induced longevity. We establish a functional link between lipophagy and microautophagy of the nucleus, both requiring Vac8 to form respective contact sites upon metabolic stress. In sum, we identify the tethering machinery of vCLIP and find that Vac8 provides a platform for multiple and competing contact sites associated with autophagy.

    • Background: High levels of nitric oxide (NO) are formed when nitrite in swallowed saliva meets acidic refluxates in the distal esophagus. In inflamed tissue with production of reactive-oxygen-species (ROS) it is reasonable to assume that luminal NO will react with particularly superoxide and form the extremely labile oxidising compound peroxynitrite. Such oxidative species have potential roles in all steps of carcinogenesis. The aim of the present study is to elucidate the presence of radical producing enzymes, and elucidate the histological changes and number of inflammatory cells in human esophageal mucosa. Material and Methods: Biopsies from healthy volunteers and patients with erosive reflux disease (ERD) were analysed for myeloperoxidase (MPO), inducible nitric oxide synthase (iNOS) and NADPH oxidase, well as the expression of inflammatory markers IL1β and IL6 by western blot technique. Expression of nitrotyrosine was used as an indicator of peroxynitrite activity, thus mirroring the foregoing radical formation. A histological evaluation of inflammation and a morphometric investigation were performed on mucosal specimens. Results: Expression of MPO and nitrotyrosine formation was up regulated in the ERD-patients compared healthy controls. No significant differences in expression of iNOS, NADPH oxidase, IL1β or IL6 were detected. No signs of active inflammation, defined as presence of inflammatory cells were observed, whereas histological signs of erosive mucosal disease were confirmed in the esophageal mucosa. Conclusion: There were clear signs of increased radical forming capacity in the epithelium despite absence of histological inflammation, in association to ERD. During acidified refluxes, epithelial ROS production in combination with luminal NO formation, may constitute aggravated factors in the carcinogenic process.

  • Coherence in Synchronizing Power Networks with Distributed Integral Control

    • We consider frequency control of synchronous generator networks and study transient performance under both primary and secondary frequency control. We model random step changes in power loads and evaluate performance in terms of expected deviations from a synchronous frequency over the synchronization transient; what can be thought of as lack of frequency coherence. We compare a standard droop control strategy to two secondary proportional integral (PI) controllers: centralized averaging PI control (CAPI) and distributed averaging PI control (DAPI). We show that the performance of a power system with DAPI control is always superior to that of a CAPI controlled system, which in turn has the same transient performance as standard droop control. Furthermore, for a large class of network graphs, performance scales unfavorably with network size with CAPI and droop control, which is not the case with DAPI control. We discuss optimal tuning of the DAPI controller and describe how internodal alignment of the integral states affects performance. Our results are demonstrated through simulations of the Nordic power grid.

  • Performance and scalability of voltage controllers in multi-terminal HVDC networks

    • In this paper, we compare the transient performance of a multi-terminal high-voltage DC (MTDC) grid equipped with a slack bus for voltage control to that of two distributed control schemes: A standard droop controller and a distributed averaging proportional-integral (DAPI) controller. We evaluate performance in terms of an ℋ2 metric that quantifies expected deviations from nominal voltages, and show that the transient performance of a droop or DAPI controlled MTDC grid is always superior to that of an MTDC grid with a slack bus. In particular, by studying systems built up over lattice networks, we show that the ℋ2 norm of a slack bus controlled system may scale unboundedly with network size, while the norm remains uniformly bounded with droop or DAPI control. We simulate the control strategies on radial MTDC networks to demonstrate that the transient performance for the slack bus controlled system deteriorates significantly as the network grows, which is not the case with the distributed control strategies.

    • Background: Vascular complications occurring before the diagnosis of myeloproliferative neoplasms (MPN) in 612 patients from four centers in Sweden, Denmark and France were retrospectively studied.Results: Vascular complications were observed in 151 (25%) of the 612 patients. Of these, 66% occurred during the two years preceding diagnosis. The majority of events were thromboembolic (95%), and included myocardial infarction (n = 46), ischemic stroke (n = 43), transient ischemic attack (TIA) (n = 22), deep vein thrombosis/pulmonary embolism (n = 19), splanchnic vein thrombosis (n = 7), and peripheral embolism (n = 7). Bleeding was observed in only 7 (5%) of the 151 patients with vascular events (3 with intracranial bleeding, 2 with epistaxis and 2 with gastrointestinal bleeding). Full blood counts obtained at least 3 months prior to the MPN diagnosis showed that 269 (44%) had abnormal blood values, fulfilling the diagnostic criteria for MPN. During the time from the abnormal blood test to the diagnosis of MPN, 50 patients suffered from a vascular complication.Conclusion: We therefore conclude that a large proportion of MPN patients suffer severe thromboembolic complications prior to diagnosis. If MPN were diagnosed earlier, a large proportion of these events might be prevented. An MPN should always be suspected and ruled out in patients with unexplained elevated hematocrit, leukocyte and/or platelet counts.
