    • To understand the biology and evolution of ruminants, the cattle genome was sequenced to about sevenfold coverage. The cattle genome contains a minimum of 22,000 genes, with a core set of 14,345 orthologs shared among seven mammalian species of which 1217 are absent or undetected in noneutherian (marsupial or monotreme) genomes. Cattle-specific evolutionary breakpoint regions in chromosomes have a higher density of segmental duplications, enrichment of repetitive elements, and species-specific variations in genes associated with lactation and immune responsiveness. Genes involved in metabolism are generally highly conserved, although five metabolic genes are deleted or extensively diverged from their human orthologs. The cattle genome sequence thus provides a resource for understanding mammalian evolution and accelerating livestock genetic improvement for milk and meat production.

    • Aims To develop a set of quality indicators (QIs) for the evaluation of the care and outcomes for atherosclerotic cardiovascular disease (ASCVD) prevention. Methods and results The Quality Indicator Committee of the European Society of Cardiology (ESC) formed the Working Group for Cardiovascular Disease Prevention Quality Indicators in collaboration with Task Force members of the 2021 ESC Guidelines on Cardiovascular Disease Prevention in Clinical Practice and the European Association of Preventive Cardiology (EAPC). We followed the ESC methodology for QI development, which involved (i) the identification of the key domains of care for ASCVD prevention by constructing a conceptual framework of care, (ii) the development of candidate QIs by conducting a systematic review of the literature, (iii) the selection of the final set of QIs using a modified Delphi method, and (iv) the evaluation of the feasibility of the developed QIs. In total, 17 main and 14 secondary QIs were selected across six domains of care for ASCVD prevention: (i) structural framework, (ii) risk assessment, (iii) care for people at risk for ASCVD, (iv) care for patients with established ASCVD, (v) patient education and experience, and (vi) outcomes. Conclusion We present the 2021 ESC QIs for Cardiovascular Disease Prevention, which have been co-constructed with EAPC using the ESC methodology for QI development. These indicators are supported by evidence from the literature, underpinned by expert consensus and aligned with the 2021 ESC Guidelines on Cardiovascular Disease Prevention in Clinical Practice to offer a mechanism for the evaluation of ASCVD prevention care and outcomes.

    • The therapeutic potential of glial cell line-derived neurotrophic factor ( GDNF) for Parkinson's disease is likely to depend on sustained delivery of the appropriate amount to the target areas. Recombinant adeno-associated viral vectors ( rAAVs) expressing GDNF may be a suitable delivery system for this purpose. The aim of this study was to define a sustained level of GDNF that does not affect the function of the normal dopamine (DA) neurons but does provide anatomical and behavioral protection against an intrastriatal 6-hydroxydopamine (6-OHDA) lesion in the common marmoset. We found that unilateral intrastriatal injection of rAAV resulting in the expression of high levels of GDNF ( 14 ng/mg of tissue) in the striatum induced a substantial bilateral increase in tyrosine hydroxylase protein levels and activity as well as in DA turnover. Expression of low levels of GDNF (0.04 ng/mg of tissue), on the other hand, produced only minimal effects on DA synthesis and only on the injected side. In addition, the low level of GDNF provided similar to 85% protection of the nigral DA neurons and their projections to the striatum in the 6-OHDA-lesioned hemisphere. Furthermore, the anatomical protection was accompanied by a complete attenuation of sensorimotor neglect, head position bias, and amphetamine-induced rotation. We conclude that when delivered continuously, a low level of GDNF in the striatum ( approximately threefold above baseline) is sufficient to provide optimal functional outcome.

  • Forty years of natural dynamics in Swiss beech forests : Structure, composition, and the influence of former management

    • We investigated forest development after the cessation of management based on inventory data from six beech forest reserves in Switzerland covering nearly 40 years, using observed changes to assess the textbook understanding of natural beech forest dynamics. Specifically, we evaluated the importance of light as a driver of tree species composition, and we aimed to disentangle the role of site characteristics and past management regimes for shaping today’s forest properties. Forest dynamics in the reserves showed a clear trend toward a broadening of the diameter distribution, an increase in basal area and standing dead wood, an increase in beech dominance, and a reduction of tree species diversity over time, conforming to expectations. However, the expected development of specific structural features, such as significant amounts of large living trees and snags or a small-scale mosaic of various developmental phases, appears to take longer than the time elapsed since the cessation of management. The observed loss in species richness can be attributed to decreasing light availability, as almost all species that disappeared were shade intolerant. Additionally, the shade-intolerant tree species had a characteristic bell-shaped diameter distribution in all reserves, indicating a lack of recruits, whereas shade-tolerant species had an irregular to monotonically decreasing diameter distribution, demonstrating sustained regeneration. Along the environmental gradient covered by the six reserves, abiotic factors are sufficient to explain tree species distribution, with management history not contributing additional information. This suggests that at larger scales, tree species composition is determined by abiotic factors, but historical management strategies were obviously adapted well to the species’ autecological requirements. Analyses such as ours provide the foundation for refining forest management systems as well as for developing effective and target-oriented conservation strategies. © 2009 by the Ecological Society of America beech forests;.
