  • People and parking requirements : Residential attitudes and day-to-day consequences of a land use policy shift towards sustainable mobility

    • A land use policy shift is taking place in a growing number of cities regarding parking, whereby a conventional supply management approach is being replaced with a parking management approach. As part of this policy shift, many cities are lowering their parking requirements.This study analysed changes in car use, car ownership, spatial parking patterns and the consequences for the everyday life of residents in a housing area with a relatively restrictive parking requirement in Gothenburg, the second largest city in Sweden. The housing area, a concrete example of how lowering parking requirements can be used to achieve targets on reduced car use and sustainable urban development, is used to discuss how parking policy should be applied to achieve the desired effect.The results show that the consequences of the restrictive requirement was paradoxically small in the study area. In practice, the requirement did not result in a decrease in the number of parking spaces, because e.g. of access to parking in neighbouring residential areas. This shows how important it is to adopt a holistic approach in parking policy, by e.g.introducing more restrictive parking requirements in parallel with other measures, such as raising parking charges and decreasing the number of public parking spaces. It also shows that planning of parking must be coordinated with other urban planning functions. Otherwise, the actual contribution of a shift in parking policy to the development of a more environmentally friendly transport system and city risks being small, despite lower parking requirements

  • Studies of the C/EBP family of transcription factors : cloning, regulation, and expression

    • The CCAAT/Enhancer Binding Protein (C/EBP) family belongs to the basic leucine zipper class of transcription factors with a total of 6 members having been identified in this family so far. This gene family has been mainly implicated in regulation of cell specific gene expression and differentiation. In order to identify factors important for the regulation of the mouse C/EBPa gene the promoter region was characterised using nuclear extracts prepared from liver. Several sites within the promoter which bound nuclear proteins were identified. One of these sites was identified as a C/EBP binding site and it was further determined that the gene is autoregulated. Upstream of this site an E-box motif was identified which binds the transcription factor USF. It was also demonstrated that purified Myc and Max proteins can bind to this site, suggesting a role for these proteins in regulating C/EBP a expression. The expression of C/EBPa and C/EBPb was studied in brown adipose tissue. Dramatic changes of gene expression during cold exposure were observed. The C/EBPa expression was transiently downregulated whereas C/EBPb expression was strongly induced. Also norepinephrine affected the expression of both genes and it was concluded that both genes are under andrenergic control in this tissue. Following up the studies of the C/EBPa promoter, the role of Myc and Max in regulation of the C/EBPb gene was investigated. It was determined that overexpression of Myc strongly represses expression of the C/EBPa gene, and also blocks differentiation of the brown adipocyte cell line HIB-lB. The Myc repression however was not mediated via the Mycbinding site, but through the core promoter region. The last two studies describe the cloning of the human C/EBPa and C/EBPe genes. Both genes were sequenced and the sequences were submitted to the EMBL/GenBank databases. The coding region of the C/EBPa gene was seen to have very high similarity to the rat and mouse genes, but the promoter region was less conserved. By using Northern blot analysis on several human tissues, the C/EBPa gene was found to have a tissue restricted expression pattern. Sequence determination of the C/EBPe gene revealed that it shared homology to the rat gene termed CRPl. Its chromosomal localisation was determined both with fluorescent in situ hybridisation (FISH) and by linkage analysis to chromosome 14qll.2, between the TCRA and CTLA genes. This chromosomal region is frequently rearranged in T-cell leukaemias. Expression in normal human tissues was detected in peripheral blood mononuclear cells, ovaries and in bone marrow. Expression was also found in the human cell lines Jurkatand HL60. This was the first time expression of this C/EBP isoform has been reported. These results show that the C/EBPe gene has a very cell specific expression pattern and that it has an intriguing chromosome location.

    • Rapporten ”Landskapsanalys för transportinfrastruktur” har tagits fram inom forskningsprojektet ”Bättre landskapsanalys för transportsektorn” (2010-2013) i samverkan mellan SLU och VTI och handlar om hur man kan arbeta med landskapsanalys (LA) i planläggningsprocessen för väg- ochjärnvägsprojekt. Vi kallar den ett kunskapsunderlag, och dess främsta syfte är att förmedla relevantkunskap från forskarsamhället till Trafikverkets landskapsexperter och därmed stödja Trafikverketsmöjligheter att leverera den landskapsanpassade infrastruktur som regeringen kräver.Fokus för arbetet har varit planläggningsfasen, dvs. när man enligt den sammanhållna vägprocessenkommit så långt att vägen/järnvägen ska prövas i en fysisk miljö.

  • ERβ in Granulosa Cell Tumors and Its Clinical Potential

    • Granulosa cell tumors (GCTs) are rare ovarian tumors comprising an adult and a juvenile subtype. They have a generally good prognosis, but the survival rate drastically declines in patients with late-stage or recurring tumors. Due to the rarity of GCTs, the tumor type is largely understudied and lacks a specific treatment strategy. Estrogen receptor beta (ERβ/ESR2) has been found to be highly expressed in GCTs, which could be of therapeutic importance since it can be targeted with small molecules. However, its role in GCTs is not known. In this review, we summarize the current knowledge about the action of ERβ in the ovary and discuss its prospective role in GCTs.

  • Road, forestry and regional planners’ work for biodiversity conservation and public participation : a case study in Poland’s hotspots regions

    • We studied how regional, road and forestry planning sectors work to implement policies about biodiversity conservation and public participation. Evaluations were based on a normative model for planning derived from the existing international policies and relevant literature. Key planning actors were then interviewed with regard to their understanding of biodiversity and participation policies as well as ability to act and willingness to implement them. The results indicate several gaps in planning processes, for example, insufficient knowledge about biodiversity conservation and participation, limited resources and tools for planning of functional habitat networks and collaboration, poor connections between local and regional planning, and weakly developed public participation. The main problem for effective policy implementation seems to be related to planners' ability to act, which indicates that relatively low priority was given to provide resources for biodiversity conservation and public participation by the relevant units. We discuss our findings in relation to the implementation of environmental policies in the new EU countries of Eastern and Central Europe.

    • The European Landscape Convention indicates that assessment of different dimensions that exist in landscapes should be taken into account in planning. In this study, we first investigated ecological, cultural and social values as perceived by the local people in a highway-planning process in Sweden. Next, we explored which factors influenced the local people's participation in the road-planning process. We used questionnaires, planning documents and the Theory of Planned Behavior to investigate the relations between different factors and local people's participation in the planning process. The results showed that people presumed the ecological values in the landscape to be adversely affected by the new road, while the social values would remain the same. Landowners had heard of the participatory-process, but few participated. Those who lived within 300m of the road were more active in the planning process than people living further away. The findings suggest that people living within a few hundred metres of the road should be treated as key stakeholders in the planning process. The involvement of other stakeholders, and when in the public participation process stakeholders should be involved, is also discussed.

  • Parkeringsnormer för bostäder : Porslinsfabriken – ett exempel på samspelet mellan läge, kollektivtrafik- och parkeringsutbud

    • I denna rapport analyseras effekterna av sänkta parkeringsnormer för bostäder för invånarna i det centrumnära, och relativt nybyggda, bostadsområdet Porslinsfabriken i Göteborg. Porslinsfabriken är med en relativt låg parkeringsnorm på 0,57 platser per lägenhet ett bra exempel på en parkeringspolitik som förs i alltfler kommuner. Resultatet visar att 19 procent av hushållen har minskat bilinnehavet, och man kör även färre antal mil på grund av parkeringsmöjligheterna. En majoritet har emellertid inte förändrat bilinnehavet eller bilanvändningen. Det fanns få skillnader mellan grupper med olika sysselsättning, utbildning, inkomst, ålder, kön och/eller familjesituation i bilinnehav, bilanvändning, eller med avseende på parkeringens konsekvenser för vardagslivet.De allra flesta är nöjda med parkeringssituationen. En parkeringsnorm på 0,57 för centralt belägna bostadsområden med god kollektivtrafikförsörjning, närhet till service och bra förutsättningar för gång-, cykel-, och kollektivtrafik möjliggör sammantaget en minskad bilanvändning. Utbudet av parkeringsplatser i närområdet gav emellertid boende möjligheter att välja mellan parkeringsplatser. En majoritet boende privatbilspendlar som tidigare och många andra har kvar sin bil. Parkeringsnormen kunde därför ha varit lägre. En praktisk implikation av resultaten är att en lägre parkeringsnorm vid nyproduktion av flerfamiljshus måste åtföljas av ett väl samordnat batteri olika åtgärder som god tillgång på kollektivtrafik, höjning av parkeringsavgifter, minskning av allmänna parkeringsplatser och så vidare.

    • This report analyses the effects of lowered parking requirements for residents in the centrally located, relatively newly built housing area, Porslinsfabriken in Gothenburg. Porslinsfabriken has a relatively low parking requirement of 0.57 places per apartment and is a good example of a parking policy being introduced in many Swedish municipalities. The results show that 19 percent of households have decreased their car ownership and also drive fewer miles because of parking opportunities. However, the majority have not altered their car ownership or car use. There are few differences between groups with different occupations, education, age, gender and/or family situation as regards car ownership, car use or the consequences of parking for everyday life. Most are happy with the parking situation. A parking requirement of 0.57 for a centrally located housing area with good public transport, close proximity to services and good opportunities for cycling, walking and using public transport has decreased overall car use. However, the range of parking available in the immediate area gives residents a choice of parking spaces. The majority of residents commute by private car as before, and many others have kept their cars. The parking requirement could therefore have been lower. A practical implication of the results is that a lower parking requirement for construction of new apartment blocks must be accompanied by a well-coordinated battery of other measures, such as good access to public transport, higher parking charges, a reduction in public parking spaces etc.

  • Sex- and Female Age-Dependent Differences in Gene Expression in Diffuse Large B-Cell Lymphoma-Possible Estrogen Effects

    • Simple Summary Females show a favorable sex difference in incidence as well as in survival for many cancer types, so also in lymphoma. The reasons for this are unknown. We have therefore analyzed global gene expression in a large cohort of the most common lymphoma type, diffuse large B-cell lymphoma. We show that many genes are differentially expressed between males and females. Furthermore, the results demonstrate sex-dependent differences in gene expression between DLBCL subtypes. In addition, gene expression differs in pre- vs. postmenopausal women suggesting that estrogen regulation of genes is involved. Thus, estrogens may contribute to the sex and female age differences in incidence and prognosis observed. For most lymphomas, including diffuse large B-cell lymphoma (DLBCL), the male incidence is higher, and the prognosis is worse compared to females. The reasons are unclear; however, epidemiological and experimental data suggest that estrogens are involved. With this in mind, we analyzed gene expression data from a publicly available cohort (EGAD00001003600) of 746 DLBCL samples based on RNA sequencing. We found 1293 genes to be differentially expressed between males and females (adj. p-value < 0.05). Few autosomal genes and pathways showed common sex-regulated expression between germinal center B-cell (GCB) and activated B-cell lymphoma (ABC) DLBCL. Analysis of differentially expressed genes between pre- vs. postmenopausal females identified 208 GCB and 345 ABC genes, with only 5 being shared. When combining the differentially expressed genes between females vs. males and pre- vs. postmenopausal females, nine putative estrogen-regulated genes were identified in ABC DLBCL. Two of them, NR4A2 and MUC5B, showed induced and repressed expression, respectively. Interestingly, NR4A2 has been reported as a tumor suppressor in lymphoma. We show that ABC DLBCL females with a high NR4A2 expression showed better survival. Inversely, MUC5B expression causes a more malignant phenotype in several cancers. NR4A2 and MUC5B were confirmed to be estrogen-regulated when the ABC cell line U2932 was grafted to mice. The results demonstrate sex- and female reproductive age-dependent differences in gene expression between DLBCL subtypes, likely due to estrogens. This may contribute to the sex differences in incidence and prognosis.

    • Implementation of the European Landscape Convention requires new tools that link ecological, social and cultural dimensions in practical planning. Here, we propose connectivity as a conceptual tool to include different dimensions into landscape and spatial planning. We present a short review of the connectivity concept in relation to ecological, social and cultural dimensions and illustrate it by examples from a real landscape planning case.
