  • Anchorless surface associated glycolytic enzymes from Lactobacillus plantarum 299v bind to epithelial cells and extracellular matrix proteins

    • An important criterion for the selection of a probiotic bacterial strain is its ability to adhere to the mucosal surface. Adhesion is usually mediated by proteins or other components located on the outer cell surface of the bacterium. In the present study we characterized the adhesive properties of two classical intracellular enzymes glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and enolase (ENO) isolated from the outer cell surface of the probiotic bacterium Lactobacillus plantarum 299v. None of the genes encoded signal peptides or cell surface anchoring motifs that could explain their extracellular location on the bacterial surface. The presence of the glycolytic enzymes on the outer surface was verified by western blotting using polyclonal antibodies raised against the specific enzymes. GAPDH and ENO showed a highly specific binding to plasminogen and fibronectin whereas GAPDH but not ENO showed weak binding to mucin. Furthermore, a pH dependent and specific binding of GAPDH and ENO to intestinal epithelial Caco-2 cells at pH 5 but not at pH 7 was demonstrated. The results showed that these glycolytic enzymes could play a role in the adhesion of the probiotic bacterium L. plantarum 299v to the gastrointestinal tract of the host. Finally, a number of probiotic as well non-probiotic Lactobacillus strains were analyzed for the presence of GAPDH and ENO on the outer surface, but no correlation between the extracellular location of these enzymes and the probiotic status of the applied strains was demonstrated.   • Strategies for improving kraftliner pulp properties

    • A large part of the world paper manufacturing consists of production of corrugated board components, kraftliner and fluting, that are used in many different types of corrugated boxes. Because these boxes are stored and transported, they are often subjected to changes in relative humidity. These changes together with mechanical loads will increase the deformation of the boxes compared to the case where the same loads are applied in a static environment. This enlarged creep due to the changes in relative humidity is called mechano-sorptive or accelerated creep. Mechano-sorptive creep forces producers to use high safety factors when designing boxes, and therefore, this is one of the key properties of kraftliner boards.   Different strategies to decrease mechano-sorptive creep, and to simultaneously gain more knowledge about the causes for this phenomenon in paper, are the aim of this work. Derivatised and underivatised black liquor lignins, a by-product produced in pulp mills in large quantities, have been used together with biomimetic methods, to modify the properties of kraftliner pulp. Furthermore, the properties of kraftliner pulp have been compared to other pulps in order to evaluate the influence of fibre morphological factors, such as fibre width and shape factor, on the mechano-sorptive creep. In addition the influence of the chemical composition of the kraftliner pulp has been evaluated both by means of treating a kraftliner pulp with chlorite and xylanase and by producing pulps with different chemical composition.   By using lignin and biomimetic methods, to create radical coupling reactions, it has been shown that it is possible to increase the wet strength of kraftliner pulp sheets. This method of treating the pulp showed, however, no significant effects on the mechano-sorptive creep. The addition of an apolar suberin-like lignin derivative, which has been shown to be possible to produce from natural resources, did show a positive effect on mechano-sorptive creep properties, but at the expense of stiffness properties in constant climate. Different pulps were compared with a kraftliner pulp and it was observed that the ratio between tensile stiffness and hygroexpansion can be used to estimate the mechano-sorptive creep properties. The hardwood kraft pulps investigated had lower hygroexpansion, probably due to more slender and straighter fibres, and higher tensile stiffness, probably due to lower lignin content. As the lignin content was varied by different methods in kraft pulps, it was observed that increased lignin content gives an increased hygroexpansion and decreased tensile stiffness as well as an increased mechano-sorptive creep. There were also indications of increased mechano-sorptive creep due to higher xylan content.

  • Molecular switch controlling expression of the mannose-specific adhesin, Msa, in Lactobacillus plantarum

    • Some lactic acid bacteria, especially Lactobacillus spp., possess adhesive properties enabling colonization of the human gastrointestinal tract. Two probiotic Lactobacillus plantarum strains, WCSF1 and 299v, display highly different mannose-specific adhesion, with L. plantarum 299v being superior to L. plantarum WCFS1 based on a yeast agglutination assay. A straightforward correlation between the mannose adhesion capacity and domain composition of the mannose-specific adhesin (Msa) in the two strains has not been demonstrated previously. In this study, we analyzed the promoter regions upstream of the msa gene encoding a mannose-specific adhesin in these two strains. The promoter region was mapped by primer extension and DNA sequence analysis, and only a single nucleotide change was identified between the two strains. However, Northern blot analysis showed a stronger msa transcript band in 299v than in WCFS1 correlating with the different adhesion capacities. During the establishment of a high-throughput yeast agglutination assay, we isolated variants of WCFS1 that displayed a very strong mannose-specific adhesion phenotype. The region upstream of the msa gene in these variants showed an inversion of a 104-bp fragment located between two perfectly inverted repeats present in the untranslated leader region. The inversion disrupts a strong hairpin structure that otherwise most likely would terminate the msa transcript. In addition, the ribosome binding site upstream of the msa gene, which is also masked within this hairpin structure, becomes accessible upon inversion, thereby increasing the frequency of translation initiation in the variant strains. Furthermore, Northern blot analysis showed a higher abundance of the msa transcript in the variants than in the wild type, correlating with a strong-Msa phenotype.

    • Typical ways of disseminating and using results of clinical research are scientific journals and reports. Presentation forms are condensed and comprehensible mainly to the experts following the specific topics. A vast amount of information remains unutilized due to the complex form of presenting the knowledge. Subject of this research is to explore possibilities of representation and also visualization of the results obtained using data mining methodologies. The intention is to formulate more than scientific ways to communicate facts that are of interest for the clinicians, medical students and even patients. Internet technologies as already widely established media support knowledge representation forms such as hypertext documents and structured knowledge components. The "Assist Me" decision support system for surgical treatment of cardiac patients integrates several forms of data mining and representation methodologies. We are showing a feasibility study in which scientific outcomes were forwarded to a broad group of potential users.

    • Migrations between different habitats are key events in the lives of many organisms. Such movements involve annually recurring travel over long distances usually triggered by seasonal changes in the environment. Often, the migration is associated with travel to or from reproduction areas to regions of growth. Young anadromous Atlantic salmon (Salmo salar) emigrate from freshwater nursery areas during spring and early summer to feed and grow in the North Atlantic Ocean. The transition from the freshwater ('parr') stage to the migratory stage where they descend streams and enter salt water ('smolt') is characterized by morphological, physiological and behavioural changes where the timing of this parr-smolt transition is cued by photoperiod and water temperature. Environmental conditions in the freshwater habitat control the downstream migration and contribute to within- and among-river variation in migratory timing. Moreover, the timing of the freshwater emigration has likely evolved to meet environmental conditions in the ocean as these affect growth and survival of the post-smolts. Using generalized additive mixed-effects modelling, we analysed spatio-temporal variations in the dates of downstream smolt migration in 67 rivers throughout the North Atlantic during the last five decades and found that migrations were earlier in populations in the east than the west. After accounting for this spatial effect, the initiation of the downstream migration among rivers was positively associated with freshwater temperatures, up to about 10 °C and levelling off at higher values, and with sea-surface temperatures. Earlier migration occurred when river discharge levels were low but increasing. On average, the initiation of the smolt seaward migration has occurred 2.5 days earlier per decade throughout the basin of the North Atlantic. This shift in phenology matches changes in air, river, and ocean temperatures, suggesting that Atlantic salmon emigration is responding to the current global climate changes.
