    • In this paper, which is of programmatic rather than quantitative nature, we aim to further delineate and sharpen the future potential of the LISA mission in the area of fundamental physics. Given the very broad range of topics that might be relevant to LISA,we present here a sample of what we view as particularly promising fundamental physics directions. We organize these directions through a "science-first" approach that allows us to classify how LISA data can inform theoretical physics in a variety of areas. For each of these theoretical physics classes, we identify the sources that are currently expected to provide the principal contribution to our knowledge, and the areas that need further development. The classification presented here should not be thought of as cast in stone, but rather as a fluid framework that is amenable to change with the flow of new insights in theoretical physics.

  • Dicer ablation in Kiss1 neurons impairs puberty and fertility preferentially in female mice

    • Kiss1 neurons, producing kisspeptins, are essential for puberty and fertility, but their molecular regulatory mechanisms remain unfolded. Here, we report that congenital ablation of the microRNA-synthesizing enzyme, Dicer, in Kiss1 cells, causes late-onset hypogonadotropic hypogonadism in both sexes, but is compatible with pubertal initiation and preserved Kiss1 neuronal populations at the infantile/juvenile period. Yet, failure to complete puberty and attain fertility is observed only in females. Kiss1-specific ablation of Dicer evokes disparate changes of Kiss1-cell numbers and Kiss1/kisspeptin expression between hypothalamic subpopulations during the pubertal-transition, with a predominant decline in arcuate-nucleus Kiss1 levels, linked to enhanced expression of its repressors, Mkrn3, Cbx7 and Eap1. Our data unveil that miRNA-biosynthesis in Kiss1 neurons is essential for pubertal completion and fertility, especially in females, but dispensable for initial reproductive maturation and neuronal survival in both sexes. Our results disclose a predominant miRNA-mediated inhibitory program of repressive signals that is key for precise regulation of Kiss1 expression and, thereby, reproductive function. Kiss1 neurons are essential for puberty and fertility. Here, the authors show that canonical microRNA biosynthesis in Kiss1 neurons plays an essential role in the control of puberty and fertility, especially in females, likely via repression of repressors on the Kiss1 gene.

    • Background: Lipid homeostasis is an evolutionarily conserved process that is crucial for energy production, storage and consumption. Drosophila larvae feed continuously to achieve the roughly 200-fold increase in size and accumulate sufficient reserves to provide all energy and nutrients necessary for the development of the adult fly. The mechanisms controlling this metabolic program are poorly understood.Results: Herein we identified a highly conserved gene, orsai (osi), as a key player in lipid metabolism in Drosophila. Lack of osi function in the larval fat body, the regulatory hub of lipid homeostasis, reduces lipid reserves and energy output, evidenced by decreased ATP production and increased ROS levels. Metabolic defects due to reduced Orsai (Osi) in time trigger defective food-seeking behavior and lethality. Further, we demonstrate that downregulation of Lipase 3, a fat body-specific lipase involved in lipid catabolism in response to starvation, rescues the reduced lipid droplet size associated with defective orsai. Finally, we show that osi-related phenotypes are rescued through the expression of its human ortholog ETFRF1/LYRm5, known to modulate the entry of β-oxidation products into the electron transport chain; moreover, knocking down electron transport flavoproteins EtfQ0 and walrus/ETFA rescues osi-related phenotypes, further supporting this mode of action.Conclusions: These findings suggest that Osi may act in concert with the ETF complex to coordinate lipid homeostasis in the fat body in response to stage-specific demands, supporting cellular functions that in turn result in an adaptive behavioral response.

    • The Laser Interferometer Space Antenna (LISA) has the potential to reveal wonders about the fundamental theory of nature at play in the extreme gravity regime, where the gravitational interaction is both strong and dynamical. In this white paper, the Fundamental Physics Working Group of the LISA Consortium summarizes the current topics in fundamental physics where LISA observations of gravitational waves can be expected to provide key input. We provide the briefest of reviews to then delineate avenues for future research directions and to discuss connections between this working group, other working groups and the consortium work package teams. These connections must be developed for LISA to live up to its science potential in these areas.

    • COLLECTiEF (Collective Intelligence for Energy Flexibility) is an EU-funded H2020 project running from June 2021 to May 2025. COLLECTiEF aims to enhance, implement, test, and evaluate an interoperable and saleable energy management system based on collective intelligence that allows easy and seamless integration of legacy equipment into a collaborative network within and between existing buildings and urban energy systems with reduced installation cost, data transfer, and computational power while increasing user comfort, energy flexibility, climate resilience, and data security. To achieve this goal, the COLLECTiEF solution requires the development of software and hardware packages to smart up buildings and their legacy equipment on a large scale while maintaining simple and robust communication with the energy grid. Here, we present the project concept, structure, objectives, and working packages. Furthermore, the main progress and achievements obtained during the first two years of the project are presented.

    • Electrochromic (EC) films of nickel oxide, with and without vanadium,   were prepared by reactive dc magnetron sputtering. They were   characterized by electrochemical and optical measurements and studied   by X-ray photoelectron spectroscopy (PES) using synchrotron radiation.  The films were analyzed under as-deposited conditions and after   bleaching/coloration by insertion/extraction of protons from a basic   solution and ensuing charge stabilization. Optical measurements were consistent with a coloration process due to charge-transfer transitions   from Ni2+ to Ni3+ states. The PES measurements showed a higher   concentration of Ni3+ in the colored films. Moreover, two peaks were   present in the O 1s spectra of the bleached film and pointed to contributions of Ni(OH)(2) and NiO. The changes in the O 1s spectra   upon coloration treatment indicate the presence of Ni2O3 in the colored   film and necessitated an extension of the conventional model for the   mechanism of EC coloration. The model involves not only proton   extraction from nickel hydroxide to form nickel oxyhydroxide but also participation of NiO in the coloration process to form Ni2O3.
