    • To identify new risk variants for cutaneous basal cell carcinoma, we performed a genome-wide association study of 16 million SNPs identified through whole-genome sequencing of 457 Icelanders. We imputed genotypes for 41,675 Illumina SNP chip-typed Icelanders and their relatives. In the discovery phase, the strongest signal came from rs78378222[C] (odds ratio (OR) = 2.36, P = 5.2 × 10(-17)), which has a frequency of 0.0192 in the Icelandic population. We then confirmed this association in non-Icelandic samples (OR = 1.75, P = 0.0060; overall OR = 2.16, P = 2.2 × 10(-20)). rs78378222 is in the 3' untranslated region of TP53 and changes the AATAAA polyadenylation signal to AATACA, resulting in impaired 3'-end processing of TP53 mRNA. Investigation of other tumor types identified associations of this SNP with prostate cancer (OR = 1.44, P = 2.4 × 10(-6)), glioma (OR = 2.35, P = 1.0 × 10(-5)) and colorectal adenoma (OR = 1.39, P = 1.6 × 10(-4)). However, we observed no effect for breast cancer, a common Li-Fraumeni syndrome tumor (OR = 1.06, P = 0.57, 95% confidence interval 0.88-1.27).

  • 3D-Printed Hydrogel Composites for Predictive Temporal (4D) Cellular Organizations and Patterned Biogenic Mineralization

    • Materials chemistries for hydrogel scaffolds that are capable of programming temporal (4D) attributes of cellular decision-making in supported 3D microcultures are described. The scaffolds are fabricated using direct-ink writing (DIW)-a 3D-printing technique using extrusion to pattern scaffolds at biologically relevant diameters (<= 100 mu m). Herein, DIW is exploited to variously incorporate a rheological nanoclay, Laponite XLG (LAP), into 2-hydroxyethyl methacrylate (HEMA)-based hydrogels-printing the LAP-HEMA (LH) composites as functional modifiers within otherwise unmodified 2D and 3D HEMA microstructures. The nanoclay-modified domains, when tested as thin films, require no activating (e.g., protein) treatments to promote robust growth compliances that direct the spatial attachment of fibroblast (3T3) and preosteoblast (E1) cells, fostering for the latter a capacity to direct long-term osteodifferentiation. Cell-to-gel interfacial morphologies and cellular motility are analyzed with spatial light interference microscopy (SLIM). Through combination of HEMA and LH gels, high-resolution DIW of a nanocomposite ink (UniH) that translates organizationally dynamic attributes seen with 2D gels into dentition-mimetic 3D scaffolds is demonstrated. These analyses confirm that the underlying materials chemistry and geometry of hydrogel nanocomposites are capable of directing cellular attachment and temporal development within 3D microcultures-a useful material system for the 4D patterning of hydrogel scaffolds.

  • A flow immunoassay for alkylphenol ethoxylate surfactants and their metabolites - questions associated with cross-reactivity, matrix effects, and validation by chromatographic techniques

    • This paper describes the application and evaluation of a competitive enzyme flow injection immunoassay (EFIIA) for screening of alkylphenol ethoxylate (APEO) surfactants in different water samples based on a generic immunoassay system previously developed ( see E. Burestedt, C Nistor, U. Schagerlof and J. Emneus, Anal. Chem., 2000, 72, 4171 - 4177). The detection limits for octylphenol ethoxylates (OPEOs), nonylphenol ethoxylates (NPEOs), and nonylphenol (NP) were 0.5 mug l(-1), between 2 and 3 mug l(-1), and 50 mug l(-1), respectively, with a sample throughput of 6 h(-1) (i.e., for triplicate analysis of each sample). Different OPEOs and NPEOs were highly cross-reactive within the assay, with sensitivities in the same order of magnitude for all the ethoxylates tested, thus the result obtained by the EFIIA method could be used as an "alkylphenol ethoxylate index". No or minor matrix effects with recoveries between 70 - 120% for the reference analyte NPEO10 in tap, and surface water, and acceptable for rainwater, were observed. Influent and effluent surfactant containing wastewater samples were analysed by EFIIA, LC-MS, LC-Fluoresence (LC-FL), and a commercial microplate ELISA. High recoveries for different concentrations of APEO(10) spiked into a 200 times diluted raw influent and effluent wastewater were achieved with the EFIIA method, however, the found APEO content of the same diluted wastewater samples, before spiking, could not be correlated directly to the chromatographic result by any of the immunoassays, and the possible reasons for this are discussed. The same trend of decreasing APEO content from influent to effluent wastewater could, however, be followed for all methods employed.

    • Metropolis II aims to provide insights in what is needed to enable high-density urban air operations. It does this by investigating the foundation for U-space U3/U4 services. The final goal is to provide a unified approach for strategic deconfliction, tactical deconfliction, and dynamic capacity management. Highly-dense operations in constrained urban airspace will likely require a degree of complexity that does not exist in modern-day air traffic management. The expected high traffic demand will require a shared use of the airspace instead of assigning exclusive use of blocks of the airspace to some flights. A unified approach for traffic management is needed because at high-densities, airspace design, flight planning, and separation management become increasingly interdependent. Metropolis II builds upon the results of the first Metropolis project. Three concepts with a varying degree of centralisation will be compared using simulations. (1) The centralised concept will take a global approach for separation management. (2) The decentralised concept aims to give the individual agents separation responsibility. (3) The hybrid concept tries to combine a centralised strategic planning agent with a robust tactical separation strategy.
