    • The present survey aimed to explore the fish fauna diversity, abundance and conservation status in the Jamuna River, a tributary of the River Brahmaputra, Bangladesh. During the study period from November 2018 to October 2019, a total of 55 species of fish were recorded, belonging to 6 orders, 20 families and 41 genera from the five selected stations near the river. Orders Cypriniformes and Siluriformes were recorded as the dominant group in the fish fauna community that comprises 34.55% and 30.91% of total species, respectively. Various types of Small Indigenous Species (SIS) and a total of 26 International Union for Conservation of Nature (IUCN) red-listed species were recorded. Population indices, viz. Shannon-Weaver index (H), Simpson's dominance index (D), Simpson's index of diversity (1-D), Margalef's index (d) and Evenness (E), were applied to demonstrate the species diversity, richness and evenness of fish, and their overall values were 1.28-1.48, 0.26-0.33, 0.67-0.74, 1.22-1.46 and 0.77-0.86, respectively. To sustain the prospect of fisheries biodiversity in the Jamuna River of Bangladesh, different fish management and conservation plan of action specifically establishing and maintaining fish sanctuaries, banning indiscriminate fishing and the use of destructive fishing gears for the protection of the breeding and nursery grounds of fish should be taken into consideration with utmost priority. © 2021 Sulav Indra Paul et al., published by Sciendo.

    • The ocean is considered to be an immense reservoir of biological and microbial diversity on the planet. In marine biospheres, microbial communities are ecologically significant as intermediaries of energy. By decomposing the dead as well as decaying organic matter with the assistance of microbial communities, it plays an indispensable role in nutrient regeneration cycles of marine ecosystems. Marine environments associated with microorganisms such as bacteria, fungi and bacterial virus have renowned potential to produce novel bioactive natural products and chemically diverse secondary metabolites like antibiotics, antifungal, antiviral, antitumor, anticancer and also different hydrolyzing enzymes, namely, protease, lipase, amylase, chitinase, etc. Hence, the bioprospecting for these compounds is of greater importance. Numerous effective and efficient application of marine microbial metabolites contribute to the fields of pharmaceuticals, biotechnological, agricultural, cosmetics industries, and so on. This review attempts to summarize the present status of bioprospecting marine microorganisms and their role in natural product discovery. © 2021 The Author(s).

  • Allosteric Binding Sites of Aβ Peptides on the Acetylcholine Synthesizing Enzyme ChAT as Deduced by In Silico Molecular Modeling

    • The native function of amyloid-β (Aβ) peptides is still unexplored. However, several recent reports suggest a prominent role of Aβ peptides in acetylcholine homeostasis. To clarify this role of Aβ, we have reported that Aβ peptides at physiological concentrations can directly enhance the catalytic efficiency of the key cholinergic enzyme, choline acetyltransferase (ChAT), via an allosteric interaction. In the current study, we further aimed to elucidate the underlying ChAT-Aβ interaction mechanism using in silico molecular docking and dynamics analysis. Docking analysis suggested two most probable binding clusters on ChAT for Aβ40 and three for Aβ42. Most importantly, the docking results were challenged with molecular dynamic studies of 100 ns long simulation in triplicates (100 ns × 3 = 300 ns) and were analyzed for RMSD, RMSF, RoG, H-bond number and distance, SASA, and secondary structure assessment performed together with principal component analysis and the free-energy landscape diagram, which indicated that the ChAT-Aβ complex system was stable throughout the simulation time period with no abrupt motion during the evolution of the simulation across the triplicates, which also validated the robustness of the simulation study. Finally, the free-energy landscape analysis confirmed the docking results and demonstrated that the ChAT-Aβ complexes were energetically stable despite the unstructured nature of C- and N-terminals in Aβ peptides. Overall, this study supports the reported in vitro findings that Aβ peptides, particularly Aβ42, act as endogenous ChAT-Potentiating-Ligand (CPL), and thereby supports the hypothesis that one of the native biological functions of Aβ peptides is the regulation of acetylcholine homeostasis.

  • Fetal/neonatal alloimmune thrombocytopenia : A systematic review of impact of HLA-DRB3∗01:01 on fetal/neonatal outcome

    • The most common, severe cases of fetal and neonatal alloimmune thrombocytopenia among whites are caused by antibodies against human platelet antigen 1a (HPA-1a). The aims of this systematic review and meta-analysis are to determine the association between maternal HLA-DRB3∗01:01 and: (1) HPA-1a-alloimmunization and (2) neonatal outcome in children born of HPA-1a-immunized women. A systematic literature search identified 4 prospective and 8 retrospective studies. Data were combined across studies to estimate pooled odds ratios (ORs) and the associated 95% confidence intervals (CIs). The population represented by the prospective studies was more than 150 000. In the prospective studies, there were 64 severely thrombocytopenic newborns (platelet count < 50 × 109/L) of whom 3 had intracranial hemorrhage. The mothers of all 64 children were HLA-DRB3∗01:01+. The number of severely thrombocytopenic children born of HPA-1a-alloimmunized women in the retrospective studies was 214; 205 of whom were born of HLA-DRB3∗01:01+ women. For HLA-DRB3∗01:01- women, the OR (95% CI) for alloimmunization was 0.05 (0.00-0.60), and for severe neonatal thrombocytopenia 0.08 (0.02-0.37). This meta-analysis demonstrates that the risk of alloimmunization and of having a child with severe thrombocytopenia are both very low for HPA-1a- women who are HLA-DRB3∗01:01-.
