    • Cancer mortality is exacerbated by late-stage diagnosis. Liquid biopsies based on genomic biomarkers can noninvasively diagnose cancers. However, validation studies have reported ~10% sensitivity to detect stage I cancer in a screening population and specific types, such as brain or genitourinary tumors, remain undetectable. We investigated urine and plasma free glycosaminoglycan profiles (GAGomes) as tumor metabolism biomarkers for multi-cancer early detection (MCED) of 14 cancer types using 2,064 samples from 1,260 cancer or healthy subjects. We observed widespread cancer-specific changes in biofluidic GAGomes recapitulated in an in vivo cancer progression model. We developed three machine learning models based on urine (Nurine = 220 cancer vs. 360 healthy) and plasma (Nplasma = 517 vs. 425) GAGomes that can detect any cancer with an area under the receiver operating characteristic curve of 0.83-0.93 with up to 62% sensitivity to stage I disease at 95% specificity. Undetected patients had a 39 to 50% lower risk of death. GAGomes predicted the putative cancer location with 89% accuracy. In a validation study on a screening-like population requiring ≥ 99% specificity, combined GAGomes predicted any cancer type with poor prognosis within 18 months with 43% sensitivity (21% in stage I; N = 121 and 49 cases). Overall, GAGomes appeared to be powerful MCED metabolic biomarkers, potentially doubling the number of stage I cancers detectable using genomic biomarkers.

  • Mate acquisition by females in a butterfly : the effects of mating status and age on female mate-locating behaviour

    • In most species, female reproductive success is determined by realized fecundity, which depends on the amount of female reproductive reserves and the availability of time for oviposition. Consequently, selection is likely to favour behaviour in virgin females that increases the likelihood of encountering males and thereby minimizing time without sperm. We used the speckled wood butterfly, Pararge aegeria, to test the hypothesis that virgin females increase the probability of encountering males by behaving more conspicuously. We also tested for an effect of age on behaviour, with the prediction that females behave more conspicuously if they remain unmated for a longer period. To do this we conducted controlled behavioural studies in large outdoor cages, comparing the behaviour of young and old, virgin and mated, females. We also assessed the time it took for a male to discover virgin versus mated females. Our results show an effect of age and mating status: old virgin females behaved more conspicuously than young virgin females and mated females, and spent more time in flight and performed more individual flights. Males also discovered virgin females faster than mated females. Furthermore, virgin females did not specifically locate the large sunspot, where perching males are found. Hence, females of P. aegeria adjust their behaviour in accordance with mating status and age, making them more likely to encounter a male and thereby maximize their reproductive success. This study underlines the importance of taking the distribution and behaviour of receptive females into account when studying mate-locating behaviour.

  • The evolution of territoriality in butterflies

    • Competition over mating opportunities is a conspicuous characteristic of animal behaviour. In many butterfly species the males establish territories in places advantageous for encountering females. This thesis addresses questions about how territoriality has evolved and is maintained in butterflies. The studies have been conducted using the speckled wood butterfly, Pararge aegeria, as a model species. Males of P. aegeria are found in sunspots on the forest floor (paper I-V), on the lookout for females visiting the sunspots. However, males are only found in sunspots above a certain size (paper III). This behavior is maintained by a mating success advantage, where using large sunspots instead of small sunspots as perching areas generates a higher reproductive output (paper I). The mating success asymmetry is not explained by female choice or by a female preference for large sunspots per se (paper I, V), but rather the large sunspot facilitates visual performance of perching males and improves flight pursuit and interception of females (paper III). Winners of territorial contests gain sole ownership of large sunspot territories, while losers search for a new suitable sunspot territory (paper I, II & IV) or use smaller, suboptimal sunspots as perching sites (paper II). Territorial contests between P. aegeria males are not settled due to an obvious morphological/physiological asymmetry (paper I). Rather, variation in resource value and motivational asymmetries are important for settling contests (paper IV). A majority of male-female interactions (paper V) and matings (paper I) are initiated by a perching male detecting and intercepting a flying female. Furthermore, females can affect their chances of being detected by a perching male by behaving more conspicuously (paper V). This thesis highlights the role of female behaviour, variation in resource value and motivation asymmetries to understand the evolution of territoriality in butterflies.

    • Ever since the interest in organic environmental contaminants first emerged 50 years ago, there has been a need to present discussion of such chemicals and their transformation products using simple abbreviations so as to avoid the repetitive use of long chemical names. As the number of chemicals of concern has increased, the number of abbreviations has also increased dramatically, sometimes resulting in the use of different abbreviations for the same chemical. In this article, we propose abbreviations for flame retardants (FRs) substituted with bromine or chlorine atoms or including a functional group containing phosphorus, i.e. BFRs, CFRs and PFRs, respectively. Due to the large number of halogenated and organophosphorus FRs, it has become increasingly important to develop a strategy for abbreviating the chemical names of FRs. In this paper, a two step procedure is proposed for deriving practical abbreviations (PRABs) for the chemicals discussed. In the first step, structural abbreviations (STABs) are developed using specific STAB criteria based on the FR structure. However, since several of the derived STABs are complicated and long, we propose instead the use of PRABs. These are, commonly, an extract of the most essential part of the STAB, while also considering abbreviations previously used in the literature. We indicate how these can be used to develop an abbreviation that can be generally accepted by scientists and other professionals involved in FR related work. Tables with PRABs and STABs for BFRs, CFRs and PERs are presented, including CAS (Chemical Abstract Service) numbers, notes of abbreviations that have been used previously, CA (Chemical Abstract) name, common names and trade names, as well as some fundamental physicochemical constants.

  • Tuning skyrmions in B20 compounds by 4d and 5d doping

    • Skyrmion stabilization in novel magnetic systems with the B20 crystal structure is reported here, primarily based on theoretical results. The focus is on the effect of alloying on the 3d sublattice of the B20 structure by substitution of heavier 4d and 5d elements, with the ambition to tune the spin-orbit coupling and its influence on magnetic interactions. State-of-the-art methods based on density functional theory are used to calculate both isotropic and anisotropic exchange interactions. Significant enhancement of the Dzyaloshinskii-Moriya interaction is reported for 5d-doped FeSi and CoSi, accompanied by a large modification of the spin stiffness and spiralization. Micromagnetic simulations coupled to atomistic spin-dynamics and ab initio magnetic interactions reveal the spin-spiral nature of the magnetic ground state and field-induced skyrmions for all these systems. Especially small skyrmions similar to 50 nm are predicted for Co0.75Os0.25Si, compared to similar to 148 nm for Fe0.75Co0.25Si. Convex-hull analysis suggests that all B20 compounds considered here are structurally stable at elevated temperatures and should be possible to synthesize. This prediction is confirmed experimentally by synthesis and structural analysis of the Ru-doped CoSi systems discussed here, both in powder and in single-crystal forms.

  • Cyanobacterial genome evolution subsequent to domestication by a plant (Azolla)

    • Cyanobacteria are an ancient and globally distributed group of photosynthetic prokaryotes including species capable of fixing atmospheric dinitrogen (N2) into biologically available ammonia via the enzyme complex nitrogenase. The ability to form symbiotic interactions with eukaryotic hosts is a notable feature of cyanobacteria and one which, via an ancient endosymbiotic event, led to the evolution of chloroplasts and eventually to the plant dominated biosphere of the globe. Some cyanobacteria are still symbiotically competent and form symbiotic associations with eukaryotes ranging from unicellular organisms to complex plants. Among contemporary plant-cyanobacteria associations, the symbiosis formed between the small fast-growing aquatic fern Azolla and its cyanobacterial symbiont (cyanobiont), harboured in specialized cavities in each Azolla leaf, is the only one which is perpetual and in which the cyanobiont has lost its free-living capacity, suggesting a long-lasting co-evolution between the two partners. In this study, the genome of the cyanobiont in Azolla filiculoides was sequenced to completion and analysed. The results revealed that the genome is in an eroding state, evidenced by a high proportion of pseudogenes and transposable elements. Loss of function was most predominant in genetic categories related to uptake and metabolism of nutrients, response to environmental stimuli and in the DNA maintenance machinery. Conversely, function was retained in key symbiotic processes such as nitrogen-fixation and cell differentiation. A comparative analysis shows that the size of the cyanobiont genome has remained relatively stable, and that few genes have been completely eliminated, since the symbiotic establishment. Indications of genes acquired via horizontal gene transfer were discovered in thec yanobiont genome, some of which may have originated from the bacterial community in the Azolla leaf-cavities. It is concluded that the perpetual nature of the Azolla symbiosis has resulted in pronounced ongoing streamlining of the cyanobiont genome around core symbiotic functions, a process not described previously for complex cyanobacteria or for any bacterial plant symbiont. Further, the status of the genome indicates that the cyanobiont is at an early stage of adapting to its host-restricted environment and continued co-evolution with the plant may result in additional genome reductions. However, although a vertical transmission process is already established, the unusual extracellular location of the cyanobiont and the intricate nature of the symbiosis, may still impose restrictions on such a reductive process.

  • Four distinct trajectories of tau deposition identified in Alzheimer’s disease

    • Alzheimer’s disease (AD) is characterized by the spread of tau pathology throughout the cerebral cortex. This spreading pattern was thought to be fairly consistent across individuals, although recent work has demonstrated substantial variability in the population with AD. Using tau-positron emission tomography scans from 1,612 individuals, we identified 4 distinct spatiotemporal trajectories of tau pathology, ranging in prevalence from 18 to 33%. We replicated previously described limbic-predominant and medial temporal lobe-sparing patterns, while also discovering posterior and lateral temporal patterns resembling atypical clinical variants of AD. These ‘subtypes’ were stable during longitudinal follow-up and were replicated in a separate sample using a different radiotracer. The subtypes presented with distinct demographic and cognitive profiles and differing longitudinal outcomes. Additionally, network diffusion models implied that pathology originates and spreads through distinct corticolimbic networks in the different subtypes. Together, our results suggest that variation in tau pathology is common and systematic, perhaps warranting a re-examination of the notion of ‘typical AD’ and a revisiting of tau pathological staging. © 2021, The Author(s), under exclusive licence to Springer Nature America, Inc.
