  • Influence of the DIN 3962 Quality Class on the Efficiency in Honed Powder Metal and Wrought Steel Gears

    • To increase the efficiency of a gearbox, research on gear mesh loss is of importance. Britton et al. concluded that the surface finishing method affects the gear mesh efficiency. The efficiency benefits of superfinishing a surface and reducing the surface roughness have been reported by Kahraman. A novel method for calculating the bearing loss torque was proposed by Tu et al. Andersson et al. found that the efficiency can vary between 2 and 5% during repeated efficiency tests due to variations in the assembly process. This work investigates how the honing surface finishing process and DIN 3962 quality class affect the gear mesh efficiency by performing tests in an FZG back-to-back test rig. Two materials, a powder metal and a wrought steel, were tested. All gears were finished using a honing process and sorted according the measured quality class. Powder metal gears of class 6, 7, 8, and ≥9 and wrought steel gears of class 6, 7, and ≥9 were tested. The efficiency were calculated from measuring the torque required to maintain a constant velocity of the FZG test rig. The results from the efficiency tests showed no significant difference in efficiency between the wrought steel and powder metal steel gears. In addition, no obvious correlation between the DIN 3962 quality class and the gear mesh efficiency could be found. When examining the wrought steel material it was found that the reproducibility of the efficiency was comparable to the assembly error of the test rig, despite the variation in quality class.

  • Izokibep : Preclinical development and first-in-human study of a novel IL-17A neutralizing Affibody molecule in patients with plaque psoriasis

    • Psoriasis, an immune-mediated inflammatory disease, affects nearly 125 million people globally. The interleukin (IL)-17A homodimer is a key driver of psoriasis and other autoimmune diseases, including psoriatic arthritis, axial spondyloarthritis, hidradenitis suppurativa, and uveitis. Treatment with monoclonal antibodies (mAbs) against IL-17A provides an improvement in the Psoriasis Area and Severity Index compared to conventional systemic agents. In this study, the Affibody(CIRCLED LATIN CAPITAL LETTER R) technology was used to identify and optimize a novel, small, biological molecule comprising three triple helical affinity domains, izokibep (previously ABY-035), for the inhibition of IL-17A signaling. Preclinical studies show that izokibep, a small 18.6 kDa IL-17 ligand trap comprising two IL-17A-specific Affibody domains and one albumin-binding domain, selectively inhibits human IL-17A in vitro and in vivo with superior potency and efficacy relative to anti-IL-17A mAbs. A Phase 1 first-in-human study was conducted to establish the safety, pharmacokinetics, and preliminary efficacy of izokibep, when administered intravenously and subcutaneously as single doses to healthy subjects, and as single intravenous and multiple subcutaneous doses to patients with psoriasis (NCT02690142; EudraCT No: 2015-004531-13). Izokibep was well tolerated with no meaningful safety concerns identified in healthy volunteers and patients with psoriasis. Rapid efficacy was seen in all psoriasis patients after one dose which further improved in patients receiving multiple doses. A therapeutic decrease in joint pain was also observed in a single patient with concurrent psoriatic arthritis. The study suggests that izokibep has the potential to safely treat IL17A-associated diseases such as psoriasis, psoriatic arthritis, axial spondyloarthritis, hidradenitis suppurativa, and uveitis.

  • Sensor Fusion for Robotic Workspace State Estimation

    • We consider the problem of tool position and orientation state estimation for robot manipulators in workspace by sensor fusion of the internal robot joint measurements with inertial measurement unit data. A prerequisite for this to be successful is accurate calibration of the sensors used. Therefore, we discuss a method for calibration of the sensor with respect to the robot end-effector, which is straightforward to apply on an arbitrary industrial manipulator. We also consider two different workspace state-estimation algorithms requiring a minimum of robot modeling; the first is based on the extended Kalman filter and the second is based on the Rao-Blackwellized particle filter. The calibration procedure and the state-estimation algorithms were evaluated and compared in extensive experiments. Both state-estimation algorithms exhibited an accuracy improvement compared to estimates provided by the forward kinematics of the robot. Moreover, both algorithms were shown to satisfy the constraints of real-time execution at 4-ms sampling period. To further evaluate and compare the robustness of the methods, the algorithms were investigated with respect to the sensitivity of the filter parameters and the noise modeling.

  • Predictors of symptom trajectory in newly diagnosed ulcerative colitis: a 3-year follow-up cohort study.

    • Psychological symptoms are associated with poorer ulcerative colitis (UC)-related outcomes. However, the majority of research is cross-sectional. We aimed to identify subgroups based on the longitudinal evolution of GI symptom levels and health-related quality of life (HRQoL), and to disentangle the directionality of effects between GI symptom levels and psychological distress.Self-reported GI symptom severity, HRQoL, inflammatory biomarkers and psychological distress were assessed in 98 newly diagnosed UC patients at disease onset and yearly for 3 consecutive years. Latent class growth analysis was used to determine subgroups based on longitudinal trajectories of symptom severity and HRQoL, and baseline predictors of trajectory group membership were determined. Cross-lagged structural equation models were used to disentangle temporal relationships between psychological functioning and symptom severity.Patients with higher initial psychological distress had increased probability of maintaining higher levels of diarrhea and abdominal pain. Conversely, patients with lower initial levels of diarrhea and abdominal pain had higher chances of maintaining lower levels of psychological distress. Higher levels of C-reactive protein at baseline predicted greater improvements in mental health after anti-inflammatory treatment. Reductions in diarrhea and abdominal pain preceded reductions in psychological symptoms over time.Baseline psychological distress is predictive of increased GI symptom severity and reduced mental HRQoL over time, suggesting early assessment of psychological symptoms may identify patients who may have worse disease trajectories. Abdominal pain predicted increased psychological distress, but not the other way around. Intervening on abdominal pain may help prevent or reduce future psychological distress.
