    • The local regulation of ovarian aromatase enzyme in polycystic ovary syndrome (PCOS) was studied with aromatase conversion and [C-11]vorozole-binding assays to analyze aromatase activity, substrate-enzyme affinity and number of aromatase binding sites in non-cultured human granulosa cells (GC) incubated with different sources and preparations of follicular fluid (FF). Incubation with FF from women stimulated in in vitro fertilization cycles with follicle-stimulating hormone yielded higher conversion activity than with FF from healthy women and PCOS patients, paralleled with higher substrate affinity (lower K-d) than with FF from healthy women. In PCOS women, charcoal-pretreated FF yielded higher conversion, whereas the ether-pretreated FF yielded lower conversion activity, than with untreated PCOS FF. Both preparations of FF yielded higher affinity to substrate (lower Kd values) and the ether-pretreated FF a lower number of binding sites (B-max). It seems that steroids with the presence of proteins in PCOS FF reduced aromatase conversion activity through decreased substrate affinity, whereas FF preparations devoid of proteins reduced the aromatase conversion activity mainly through blocking of aromatase active sites. Identification of specific agents responsible for this rapid regulation of aromatase function might help to understand normal regulation of the menstrual cycle and supposed imbalances of inhibitors/activators in PCOS.

    • The Global Lung Function Initiative (GLI) has recently published international reference values for diffusing capacity of the lung for carbon monoxide (DLCO). Lower limit of normal (LLN), i.e. the 5th percentile, usually defines impaired DLCO. We examined if the GLI LLN for DLCO differs from the LLN in a Swedish population of healthy, never-smoking individuals and how any such differences affect identification of subjects with respiratory burden.Spirometry, DLCO, chest high-resolution computed tomography (HRCT) and questionnaires were obtained from the first 15 040 participants, aged 50–64 years, of the Swedish CArdioPulmonary bioImage Study (SCAPIS). Both GLI reference values and the lambda-mu-sigma (LMS) method were used to define the LLN in asymptomatic never-smokers without respiratory disease (n=4903, of which 2329 were women).Both the median and LLN for DLCO from SCAPIS were above the median and LLN from the GLI (p<0.05). The prevalence of DLCO DLCO >GLI LLN but DLCO >GLI LLN but versus 4.5%, p<0.001), chronic airflow limitation (8.5% versus 3.9%, p<0.001) and chronic bronchitis (8.3% versus 4.4%, p<0.01) than subjects (n=13 600) with normal DLCO (>GLI LLN and >SCAPIS LLN). No differences were found with regard to physician-diagnosed asthma.The GLI LLN for DLCO is lower than the estimated LLN in healthy, never-smoking, middle-aged Swedish adults. Individuals with DLCO above the GLI LLN but below the SCAPIS LLN had, to a larger extent, an increased respiratory burden. This suggests clinical implications for choosing an adequate LLN for studied populations.

    • Human land use such as pastoralism may be a key determinant of wildlife populations. We studied the influence of land use on wildlife and livestock in south-western Kalahari, Botswana, during the wet and dry seasons. We included two types of livestock areas and two types of wildlife protection areas. Wild and domestic mammals >0.2 kg were sampled by distance technique. Small mammals <0.2 kg were trapped and a vegetation survey was performed. Livestock and some medium-sized wildlife species were most abundant in livestock areas, while large-sized wildlife species were mainly restricted to wildlife areas. Most red-listed species were confined to the national park. The influence of land use on abundances changed moderately between seasons. However, cattle moved into wildlife areas during the wet season. Differences in species richness and diversity were most pronounced during the dry season. The trapping frequency of small mammals was highest in the national park. Differences in vegetation structure included sparser field layer and higher cover of an invasive shrub in livestock areas. Our study shows the importance of protected areas for many wildlife species in the Kalahari. It also shows the value of a multi-species approach for investigating the effects of land use on wildlife communities.

  • Mammal community structure in relation to disturbance and resource gradients in southern Africa

    • The arrangements of mammals differ along environmental gradients, such as of disturbance and resources. We examined how mammal community composition in the Kalahari, Botswana, varied in relation to disturbance and resource gradients. We predicted that livestock-keeping villages are disturbances and pans are resources for wildlife, that the responses of mammals to disturbance and resources depend on their functional types and that increased disturbance over time has reduced the numbers and distributions of wildlife. The methods involved road-side counts of mammals > 0.2 kg and trapping of small mammals, < 0.2 kg. The disturbance gradient was more important than the resource gradient for explaining the distribution of mammals > 0.2 kg. Communities in low disturbance-high resource areas were most diverse regarding species and functional types. Small mammal species richness and abundances were unaffected by villages, but increased with distance from pans. Villages were particularly deterring to large wild herbivores, functionally similar to livestock. Most large wild herbivores had decreased since 1975-1983. We conclude that large and medium-sized mammals are highly affected by large-scale disturbance gradients, while small mammals are most dependent on small-scale variation in resources, probably shelter and food. Increased disturbance over time leads to decreasing ranges and numbers of the large wild herbivores.

  • Pancreatic neuroendocrine tumors : diagnosis with PET

    • PURPOSE: To evaluate the use of carbon-11-labeled L-dihydroxyphenylalanine (L-DOPA) and hydroxytryptophan (5-HTP) in the diagnosis of pancreatic endocrine tumors with positron emission tomography (PET). MATERIALS AND METHODS: Twenty-two consecutive patients with clinically and biochemically verified pancreatic endocrine tumors were examined with computed tomography (CT) and PET with L-DOPA alone (n = 16) or both C-11-L-DOPA and C-11-5-HTP (n = 6). RESULTS: Tumor uptake of L-DOPA was found in 11 patients, eight of whom had metastatic disease. Heterogeneity of tracer uptake was noted among different lesions in the same patient (ie, high uptake in some lesions and low uptake in others). Results in patients examined with both L-DOPA and 5-HTP correlated well, but the uptake levels of 5-HTP were higher in two of three patients with positive findings. In two additional patients, CT enabled detection of tumors not detected at PET. CONCLUSION: The current PET technique can be a valuable complement to CT in demonstration of functional pancreatic endocrine tumors, in particular, glucagonomas, but is less useful in detection of nonfunctional tumors.
