    • Previously, we reported germline DNA variants associated with risk of urinary bladder cancer (UBC) in Dutch and Icelandic subjects. Here we expanded the Icelandic sample set and tested the top 20 markers from the combined analysis in several European case-control sample sets, with a total of 4,739 cases and 45,549 controls. The T allele of rs798766 on 4p16.3 was found to associate with UBC (odds ratio = 1.24, P = 9.9 x 10(-12)). rs798766 is located in an intron of TACC3, 70 kb from FGFR3, which often harbors activating somatic mutations in low-grade, noninvasive UBC. Notably, rs798766[T] shows stronger association with low-grade and low-stage UBC than with more aggressive forms of the disease and is associated with higher risk of recurrence in low-grade stage Ta tumors. The frequency of rs798766[T] is higher in Ta tumors that carry an activating mutation in FGFR3 than in Ta tumors with wild-type FGFR3. Our results show a link between germline variants, somatic mutations of FGFR3 and risk of UBC.

    • [1] Large spatial and temporal variations in stream total organic carbon (TOC) concentration and export occurred during an 11-year observation period (1993-2003) in a boreal headwater catchment. TOC flux and concentration patterns from mire- and forest-dominated subcatchments differed ( mean annual flux 8.2 g m(-2)a(-1) versus 5.8 g m(-2) a(-1)). Temporal variations in stream TOC concentrations in both landscape types were primarily driven by variations in streamflow, with the mire stream generally diluting by half with increased runoff during spring flood and TOC from the forested landscape increasing during runoff peaks irrespective of season. Average TOC concentration in the mire stream in the snow-free season increased with increased seasonal precipitation from around 20 to 40 mg L-1 but then dropped to around 35 mg L-1 during very wet years. Average snow-free season TOC concentration at the forested site remained stable when summer precipitation was below average but then increased from 10 to around 25 mg L-1 during exceptionally wet years. For both the forested subcatchment and the whole catchment, TOC concentrations increased during the warm summer months during wet years, but no such increase occurred during dry years. Interannual variations in TOC flux were primarily driven by variations during the snow-free period. Wet years decreased the relative TOC export from the mire and favored the relative export of TOC from areas dominated by forest, an observation that also held true on a larger scale when similar landscape types were considered. Predicted climate change in rainfall and temperature patterns will affect the amount and character of TOC exported downstream from boreal landscapes with a mix of forest and mire.

  • Controls on snowmelt water mean transit times in northern boreal catchments

    • Catchment-scale transit times for water are increasingly being recognized as an important control on geochemical processes. In this study, snowmelt water mean transit times (MTTs) were estimated for the 15 Krycklan research catchments in northern boreal Sweden. The snowmelt water MTTs were assumed to be representative of the catchment-scale hydrologic response during the spring thaw period and, as such, may be considered to be a component of the catchment's overall MTT. These snowmelt water MTTs were empirically related to catchment characteristics and landscape structure represented by using different indices of soil cover, topography and catchment similarity. Mire wetlands were shown to be significantly correlated to snowmelt MTTs for the studied catchments. In these wetlands, shallow ice layers form that have been shown to serve as impervious boundaries to vertical infiltration during snowmelt periods and, thus, alter the flow pathways of water in the landscape. Using a simple thought experiment, we could estimate the potential effect of thawing of ice layers on snowmelt hydrologic response using the empirical relationship between landscape structure (represented using a catchment-scale Pe number) and hydrologic response. The result of this thought experiment was that there could be a potential increase of 20-45% in catchment snowmelt water MTTs for the Krycklan experimental catchments. It is therefore possible that climatic changes present competing influences on the hydrologic response of northern boreal catchments that need to be considered. For example, MTTs may tend to decrease during some times of the year due to an acceleration in the hydrologic cycle, while they tend to increase MTTs during other times of the year due to shifts in hydrologic flow pathways. The balance between the competing influences on a catchment's MTT has consequences on climatic feedbacks as it could influence hydrological and biogeochemical cycles at the catchment scale for northern latitude boreal catchments.   • Soil frost effects on soil water and runoff dynamics along a boreal transect : 2. Simulations

    • A physically based SVAT-model was rested with soil and snow physical measurements, as well as runoff data fr om an 8600 m(2) catchment in northern Sweden in order to quantify the influence of soil frost on spring snowmelt runoff in a moderately sloped, boreal forest. The model was run as an array of connected profiles cascading to the brook. For three winter seasons (1995-98) it was able to predict the onset and total accumulation of the runoff with satisfactory accuracy. Surface runoff was identified as only a minor fraction of the total runoff occurring during short periods in connection with ice blocking of the water-conducting pores. Little surface runoff, though, does not mean that soil frost is unimportant for spring runoff. Simulations without frost routines systematically underestimated the total accumulated runoff. The possibility of major frost effects appearing in response to specific combinations of weather conditions were also tested. Different scenarios of critical initial conditions for the winter, e.g, high water saturation and delayed snow accumulation leading to an increased frost penetration, were tested. These showed that under special circumstances there is potential for increased spring runoff due to soil frost.

    • We conducted a genome-wide association study on 969 bladder cancer cases and 957 controls from Texas. For fast-track validation, we evaluated 60 SNPs in three additional US populations and validated the top SNP in nine European populations. A missense variant (rs2294008) in the PSCA gene showed consistent association with bladder cancer in US and European populations. Combining all subjects (6,667 cases, 39,590 controls), the overall P-value was 2.14 x 10(-10) and the allelic odds ratio was 1.15 (95% confidence interval 1.10-1.20). rs2294008 alters the start codon and is predicted to cause truncation of nine amino acids from the N-terminal signal sequence of the primary PSCA translation product. In vitro reporter gene assay showed that the variant allele significantly reduced promoter activity. Resequencing of the PSCA genomic region showed that rs2294008 is the only common missense SNP in PSCA. Our data identify rs2294008 as a new bladder cancer susceptibility locus.
