  • Identification of human S100A9 as a novel target for treatment of autoimmune disease via binding to quinoline-3-carboxamides.

    • Despite more than 25 years of research, the molecular targets of quinoline-3-carboxamides have been elusive although these compounds are currently in Phase II and III development for treatment of autoimmune/inflammatory diseases in humans. Using photoaffinity cross-linking of a radioactively labelled quinoline-3-carboxamide compound, we could determine a direct association between human S100A9 and quinoline-3-carboxamides. This interaction was strictly dependent on both Zn++ and Ca++. We also show that S100A9 in the presence of Zn++ and Ca++ is an efficient ligand of receptor for advanced glycation end products (RAGE) and also an endogenous Toll ligand in that it shows a highly specific interaction with TLR4/MD2. Both these interactions are inhibited by quinoline-3-carboxamides. A clear structure-activity relationship (SAR) emerged with regard to the binding of quinoline-3-carboxamides to S100A9, as well as these compounds potency to inhibit interactions with RAGE or TLR4/MD2. The same SAR was observed when the compound's ability to inhibit acute experimental autoimmune encephalomyelitis in mice in vivo was analysed. Quinoline-3-carboxamides would also inhibit TNFalpha release in a S100A9-dependent model in vivo, as would antibodies raised against the quinoline-3-carboxamide-binding domain of S100A9. Thus, S100A9 appears to be a focal molecule in the control of autoimmune disease via its interactions with proinflammatory mediators. The specific binding of quinoline-3-carboxamides to S100A9 explains the immunomodulatory activity of this class of compounds and defines S100A9 as a novel target for treatment of human autoimmune diseases.

  • Fatigue in Patients With Head and Neck Cancer Treated With Radiation Therapy: A Prospective Study of Patient-Reported Outcomes and Their Association With Radiation Dose to the Cerebellum

    • Purpose: Although fatigue is a known side effect in patients with head and neck cancer (HNC) receiving radiation therapy, knowledge regarding long-term fatigue and dose-response relationships to organs at risk is scarce. The aim of this prospective study was to analyze patient-reported fatigue in patients with HNC receiving radiation therapy and to explore any possible association with organ-at-risk doses. Methods and Materials: Patients with HNC referred for curative radiation therapy were eligible for inclusion in the study. To assess patient-reported fatigue, quality of life questionnaires (European Organization for Research and Treatment of Cancer QLQ-C30 and QLQ-FA12) were distributed before treatment and 1, 3, 6, 12, 24, and 60 months after the start of treatment. Mean dose (Dmean) and near maximum dose (D2%) of the cerebellum and brain stem were evaluated in relation to baseline-adjusted fatigue scores at 3 months. Results: One hundred twenty-six patients treated with intensity modulated radiation therapy between 2008 and 2010 were available for final analysis. Female sex and age <60 years were associated with higher fatigue at baseline, whereas patients also treated with chemotherapy had reduced physical and emotional fatigue at 6 months. Physical fatigue (QLQ-FA12 scale) increased from baseline up to 3 months (29 vs 59; P < .0001) but showed no difference compared with baseline from 1 to 5 years. Emotional fatigue was significantly lower at 5 years compared with baseline (14 vs 28; P < .0001). Patients with cerebellum Dmean > 3.5 Gy had higher mean physical fatigue scores at 3 months (38 vs 27; P = .036). Conclusions: Although there is a significant increase in fatigue scores for patients with HNC up to 1 year after radiation therapy, this study showed a return to baseline levels at 5 years. A possible association was found between physical fatigue and a higher mean dose to the cerebellum.

    •     We investigate the possibility of an arbitrage free model for the term structure of in-terest rates where the yield curve only changes through a parallel shift. We consider HJMtype forward rate models driven by a multidimensional Wiener process as well as by a gen-eral marked point process. Within this general framework we show that there does indeedexist a large variety of nontrivial parallel shift term structure models, and we also describethese in detail. We also show that there exists no nontrivial flat term structure model. Thesame analysis is repeated for the similar case, where the yield curve only changes throughproportional shifts.

Målet med projektet har varit att uppnå samarbetsformer mellan Individ- och Familjeomsrog (IoF) samt Vård- och Omsorg (VoF) som gör att personer som är, eller riskerar att bli, hemlösa och vars problematik berör båda verksamheterna ska kunna ta del av förbättrade och samordnade insatser.

    • Internal crowdsourcing utilizes a firm’s employees, of which many have a strong understanding of the domains in which the firm operates, for contributing with, developing and evaluating ideas. On the one hand, these employees can use their domain-specific knowledge to identify the value of what may seem a far-fetched solution to the average employee. On the other hand, previous research has shown that employees typically evaluate ideas in their domains less favorably if they do not align with ongoing exploitation activities. Hence, this study focuses on whether a higher degree of relevant domain-specific knowledge makes employees participating in internal crowdsourcing prefer exploitative solutions when evaluating ideas. An empirical study of an online platform for firm-internal innovation in a multinational engineering company showed that employees who only infrequently participated in internal crowdsourcing mostly contributed to and evaluated ideas within their own domain. Employees who frequently participated also contributed to and evaluated ideas outside their own domains. By statistically analyzing group differences during idea evaluation, we show that employees participating infrequently favor exploitable solutions, whereas employees participating frequently are more uncertain. The former difference is only seen concerning ideas that require domain-specific knowledge to understand, but the latter is observed for all types of ideas. This study makes three substantial contributions. First, employees with domain-specific knowledge, through their preference for exploitative solutions, bias the outcome of internal crowdsourcing when idea evaluation requires domain-specific knowledge. Second, this bias is aggravated by the overall higher level of uncertainty displayed by employees participating frequently in internal crowdsourcing and thereby tend to reach out to other domains. Third, in order to mitigate this, bias management can build engagement in internal crowdsourcing through idea challenges that do not require domain-specific knowledge and consider avoiding employees with a strongly associated domain knowledge for idea evaluation.

    • The emergence of technology increasingly depends on innovation ecosystems and frequently involves actors from both industry and academia. However, value creation may experience challenges due to bias formed during public-private innovation ecosystem genesis.This empirical study of bias in a new pan-European public-private initiative provides results regarding innovation ecosystems and the individuals typically active during their genesis: value creation is biased towards the selection of incumbent firms and complement challenges, and participation is biased towards engineers with knowledge of exploitation from multiple domains and researchers with knowledge of exploitation from single domains.This suggests that the implications of the loose coupling emphasised by the innovation ecosystems discourse and the knowledge of the different contexts in which firms capture value are more complex than previously acknowledged. The practical implications are that the ability of public innovation ecosystem leadership to act early on novel technology might be offset by the inability of involved firms to commit to bringing the technology to market and that individuals typically active during public-private innovation ecosystems genesis are not ideal for handling this challenge. In fact, increasingly connected public leadership could smother the innovation ecosystem unless well-connected and multidisciplinary researchers are brought in as brokers.

Det kan vara människor med funktionsnedsättning, med invandrarbakgrund, som fastnat i långtidsarbetslöshet etc. Rapporten belyser några av de insatser som pågår idag för att skapa arbeten för dessa grupper genom framväxten av socialt företagande med det primära målet att få människor i sysselsättning
