    • Advances in antibody production render a growing supply of affinity reagents for immunohistochemistry (IHC), and tissue microarray (TMA) technologies facilitate simultaneous analysis of protein expression in a multitude of tissues. However, collecting validated IHC data remains a bottleneck problem, as the standard method is manual microscopical analysis. Here we present a high-throughput strategy combining IHC on a recently developed cell microarray with a novel, automated image-analysis application (TMAx). The software was evaluated on 200 digital images of IHC-stained cell spots, by comparing TMAx annotation with manual annotation performed by seven human experts. A high concordance between automated and manual annotation of staining intensity and fraction of IHC-positive cells was found. in a limited study, we also investigated the possibility to assess the correlation between mRNA and protein levels, by using TMAx output results for relative protein quantification and quantitative real-time PCR for the quantification of corresponding transcript levels. In conclusion, automated analysis of immunohistochemically stained in vitro-cultured cells in a microarray format can be used for high-throughput protein profiling, and extraction of RNA from the same cell lines provides a basis for comparing transcription and protein expression on a global scale.

  • A human protein atlas for normal and cancer tissues based on antibody proteomics

    • Antibody-based proteomics provides a powerful approach for the functional study of the human proteome involving the systematic generation of protein-specific affinity reagents. We used this strategy to construct a comprehensive, antibody-based protein atlas for expression and localization profiles in 48 normal human tissues and 20 different cancers. Here we report a new publicly available database containing, in the first version, similar to 400,000 high resolution images corresponding to more than 700 antibodies toward human proteins. Each image has been annotated by a certified pathologist to provide a knowledge base for functional studies and to allow queries about protein profiles in normal and disease tissues. Our results suggest it should be possible to extend this analysis to the majority of all human proteins thus providing a valuable tool for medical and biological research.

  • Mode monitoring and call-outs : An eye-tracking study of 2-crew automated flight deck operations

    • Mode awareness has been suggested as a critical factor in safe operations of automated aircraft. This study investigated mode awareness by measuring eye point of gaze of both pilots during simulated commercial flights, while recording call-outs and tracking aircraft performance. The results of this study show that the compliance to manufacturer or air carrier procedures regarding mode monitoring and call-outs was very low. However, this did not seem to have a negative effect on the flight path or safety during our observations. Crews exhibited a proliferation of strategies to keep track of status and behavior of the automation, often with little reliance on the flight mode annunciations of the primary flight display. The data confirm the limitations of current flight mode annunciator designs, and suggest that mode awareness is a more complex phenomenon than what can be captured by measuring eye point of gaze and communication alone.

    • This thesis is based on an ethnographic field study among a group of monolingual children from one and a half to three years old at a pre-school in a Swedish village. The primary aim of the study has been to gain knowledge about how young children conquer and express literacy in their everyday lives. One assumption was that children are active and competent in their search for meaning when dealing with texts, signs and images. A second aim has been to investigate whether children’s literacy could have an impact on the social and cultural context of pre-school.Research questions addressed in the study focus on children’s participation in literacy events and their actions in literacy practice. An additional question was  whether children were contributing to literacy practice when interacting with other children in the group.The theoretical framework draws upon socio-cultural theory. The empirical material consists of video recordings and the focus of observation has been on children’s actions related to literacy, expressed as early literacy, including reading and writing as well as telling and retelling narratives, singing and other verbal and non verbal communication. In children’s literacy events and literacy practice the specific context was of central interest. All the material collected has been transcribed and transferred into text and constitutes the basis for analyzing what children are performing in actual events and practices. The guiding principle for description was at first to give a close reproduction of children’s verbal utterances and their acting linked to the concept of early literacy. Secondly, the aim was to describe and analyze whether the utterances and actions could be linked to the specific social and cultural context.The analysed material demonstrates how children participate and interact with each other while engaged in literacy and the material also displays the content of their communication. Two different kinds of literacy appear: one is narrative tellings and the other is reading and drawing/writing. A deeper analysis shows that children are building knowledge of telling and also creating a specific manifesto of literacy. The children also underline what they are doing through verbal expressions where they defined  themselves as both readers and writers.The result gives a contribution to new knowledge and an understanding of early literacy among very young children as something they have created in pre-school as a social and cultural environment. Literacy in the studied group of children uncovers many more expressions, including several actions with regard to written material, than we usally relate to the youngest children in pre-school.

    • The aim of this study was to identify and characterize family caregivers' conceptions of usage of and information on products, technology, and Web-based services when caring at home for next of kin aged over 65 years. A phenomenographic method with semi-structured interviews with ten family caregivers was used. The results show that the family caregivers considerd products and technology to be essential for an active life, and they stressed both facilitators for and hindrances to their optimal usage. The family caregivers apprehended a lack of information regarding products, technology and Web-based services, and additionally showed limited interest in using Web-based services. There is a need for better information about existing availability and for improvement in follow-up regarding prescriptions given. Therefore, it is necessary to involve family caregivers in the prescription and decision process in line with their wishes, since this to a highly degree influences their possibility to take care of their next of kin at home.

    • This review underlines the importance of indoor contamination as a pathway of human exposure to hexabromocyclododecanes (HBCDs), polybrominated diphenyl ethers (PBDEs), and perfluoroalkyl compounds (PFCs). There is ample evidence of substantial contamination of indoor dust with these chemicals and that their concentrations in indoor air exceed substantially those outdoors. Studies examining the relationship between body burden and exposure via indoor dust are inconsistent while some indicate a link between body burdens and PBDE and HBCD exposure via dust ingestion, others find no correlation. Likewise, while concentrations in indoor dust and human tissues are both highly skewed, this does not necessarily imply causality. Evidence suggests exposure via dust ingestion is higher for toddlers than adults. Research priorities include identifying means of reducing indoor concentrations and indoor monitoring methods that provide the most ""biologically-relevant"" measures of exposure as well as monitoring a wider range of microenvironment categories. Other gaps include studies to improve understanding of the following: emission rates and mechanisms via which these contaminants migrate from products into indoor air and dust; relationships between indoor exposures and human body burdens; relevant physicochemical properties; the gastrointestinal uptake by humans of these chemicals from indoor dust; and human dust ingestion rates.

  • Dispatch of Firefighters and Police Officers in Out-of-Hospital Cardiac Arrest : A Nationwide Prospective Cohort Trial Using Propensity Score Analysis.

    • BACKGROUND: Dispatch of basic life support-trained first responders equipped with automated external defibrillators in addition to advanced life support-trained emergency medical services personnel in out-of-hospital cardiac arrest (OHCA) has, in some minor cohort studies, been associated with improved survival. The aim of this study was to evaluate the association between basic life support plus advanced life support response and survival in OHCA at a national level.METHODS AND RESULTS: This prospective cohort study was conducted from January 1, 2012, to December 31, 2014. People who experienced OHCA in 9 Swedish counties covered by basic life support plus advanced life support response were compared with a propensity-matched contemporary control group of people who experienced OHCA in 12 counties where only emergency medical services was dispatched, providing advanced life support. Primary outcome was survival to 30 days. The analytic sample consisted of 2786 pairs (n=5572) derived from the total cohort of 7308 complete cases. The median time from emergency call to arrival of emergency medical services or first responder was 9 minutes in the intervention group versus 10 minutes in the controls (P<0.001). The proportion of patients admitted alive to the hospital after resuscitation was 31.4% (875/2786) in the intervention group versus 24.9% (694/2786) in the controls (conditional odds ratio, 1.40; 95% confidence interval, 1.24-1.57). Thirty-day survival was 9.5% (266/2786) in the intervention group versus 7.7% (214/2786) in the controls (conditional odds ratio, 1.27; 95% confidence interval, 1.05-1.54).CONCLUSIONS: In this nationwide interventional trial, using propensity score matching, dispatch of first responders in addition to emergency medical services in OHCA was associated with a moderate, but significant, increase in 30-day survival.CLINICAL TRIAL REGISTRATION: URL: http://www.clinicaltrials.gov.   • Life cycle assessment in road infrastructure planning using spatial geological data

    • Purpose: The purpose of the study was to outline and demonstrate a new geographic information system (GIS)-based approach for utilising spatial geological data in three dimensions (i.e. length, width and depth) to improve estimates on earthworks during early stages of road infrastructure planning. Methods: This was undertaken by using three main methodological steps: mass balance calculation, life cycle inventory analysis and spatial mapping of greenhouse gas (GHG) emissions and energy use. The mass balance calculation was undertaken in a GIS environment using two assumptions of geological stratigraphy for two proposed alternative road corridors in Sweden. The estimated volumes of excavated soil, blasted rock and filling material were later multiplied with the GHG emission and energy use factors for these processes, to create spatial data and maps in order to show potential impacts of the studied road corridors. The proposed GIS-based approach was evaluated by comparing with actual values received after one alternative was constructed. Results and discussion: The results showed that the estimate of filling material was the most accurate (about 9 % deviation from actual values), while the estimate for excavated soil and blasted rock resulted in about 38 and 80 % deviation, respectively, from the actual values. It was also found that the total volume of excavated and ripped soils did not change when accounting for stratigraphy. Conclusions: The conclusion of this study was that more information regarding embankment height and actual soil thickness would further improve the model, but the proposed GIS-based approach shows promising results for usage in LCA at an early stage of road infrastructure planning. Thus, by providing better data quality, GIS in combination with LCA can enable planning for a more sustainable transport infrastructure.
