- The Visual Object Tracking Segmentation VOTS2023 challenge is the eleventh annual tracker benchmarking activity of the VOT initiative. This challenge is the first to merge short-term and long-term as well as single-target and multiple-target tracking with segmentation masks as the only target location specification. A new dataset was created; the ground truth has been withheld to prevent overfitting. New performance measures and evaluation protocols have been created along with a new toolkit and an evaluation server. Results of the presented 47 trackers indicate that modern tracking frameworks are well-suited to deal with convergence of short-term and long-term tracking and that multiple and single target tracking can be considered a single problem. A leaderboard, with participating trackers details, the source code, the datasets, and the evaluation kit are publicly available at the challenge website1

- Ever since its introduction 40 years ago l-3,4-dihydroxyphenylalanine (l-DOPA) therapy has retained its role as the leading standard medication for patients with Parkinson's disease. With time, however, the shortcomings of oral l-DOPA treatment have become apparent, particularly the motor fluctuations and troublesome dyskinetic side effects. These side effects, which are caused by the excessive swings in striatal dopamine caused by intermittent oral delivery, can be avoided by delivering l-DOPA in a more continuous manner. Local gene delivery of the l-DOPA synthesizing enzymes, tyrosine hydroxylase and guanosine-tri-phosphate-cyclohydrolase-1, offers a new approach to a more refined dopaminergic therapy where l-DOPA is delivered continuously at the site where it is needed i.e. the striatum. In this study we have explored the therapeutic efficacy of adeno-associated viral vector-mediated l-DOPA delivery to the putamen in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-treated rhesus monkeys, the standard non-human primate model of Parkinson's disease. Viral vector delivery of the two enzymes, tyrosine hydroxylase and guanosine-5'-tri-phosphate-cyclohydrolase-1, bilaterally into the dopamine-depleted putamen, induced a significant, dose-dependent improvement of motor behaviour up to a level identical to that obtained with the optimal dose of peripheral l-DOPA. Importantly, this improvement in motor function was obtained without any adverse dyskinetic effects. These results provide proof-of-principle for continuous vector-mediated l-DOPA synthesis as a novel therapeutic strategy for Parkinson's disease. The constant, local supply of l-DOPA obtained with this approach holds promise as an efficient one-time treatment that can provide long-lasting clinical improvement and at the same time prevent the appearance of motor fluctuations and dyskinetic side effects associated with standard oral dopaminergic medication.

Design and Synthesis of BACE-1 Inhibitors Containing a New Hydroxyethylene (HE) Scaffold: Potent activities in a cellular assay

- In a preceding report from our group we disclosed the development of a novel HE transition state isostere with a difluorophenoxymethyl side chain in the P1 position and a methoxy group in the P1’ position furnishing highly potent inhibitors of BACE-1 (i.e. lead compound 1), which moreover exhibit very promising selectivity over cathepsin D. In a continuation of this work with the aim at improving on the cell-based activity and pharmacokinetic properties, we have further developed the SAR for the P1 side chain of inhibitor 1 whereby the P1 side chain oxygen has been substituted for an amine, a carbon or a bond. The chemistry developed for the previous HE inhibitor structure 1 has now been extended to readily accommodate the introduction of new P1 side chains into this new HE scaffold. These modifications have given rise to several highly potent inhibitors where the most potent displayed a BACE-1 Ki value of 0.2 nM and a cell-based Aβ40 IC50 value of 9 nM. Thus, regarding the enzyme inhibition in the cell assay a more than 600-fold improvement compared to compound 1 was achieved via minor structural alterations.

- Cancer-associated fibroblasts (CAFs) are a major constituent of the tumor microenvironment, although their origin and roles in shaping disease initiation, progression and treatment response remain unclear due to significant heterogeneity. Here, following a negative selection strategy combined with single-cell RNA sequencing of 768 transcriptomes of mesenchymal cells from a genetically engineered mouse model of breast cancer, we define three distinct subpopulations of CAFs. Validation at the transcriptional and protein level in several experimental models of cancer and human tumors reveal spatial separation of the CAF subclasses attributable to different origins, including the peri-vascular niche, the mammary fat pad and the transformed epithelium. Gene profiles for each CAF subtype correlate to distinctive functional programs and hold independent prognostic capability in clinical cohorts by association to metastatic disease. In conclusion, the improved resolution of the widely defined CAF population opens the possibility for biomarker-driven development of drugs for precision targeting of CAFs.

- I ett aktionsforskningsprojekt undersöktes hur relationer mellan matematik och bygg- och anläggningsämnet i gymnasieskolan kan beskrivas, med specifikt intresse av elevers kommande yrkesaktiviteter som byggnadsarbetare. Vidare undersöktes hur elever på yrkesprogram, lärare (i matematik och bygg- och anläggning) och en forskare beskriver dessa relationer i början och slutet av aktionsforskningsprojektet. Datamaterialet bestod av svar på frågeformulär, bilder, anteckningar och loggböcker. Resultaten visar hur mötespunkten mellan matematik och bygg- och anläggning bör ses som ett eget kunskapsområde som i sig har olika slags relationer till respektive ämne, vilket i artikeln sammanfattas i en generell modell. Resultaten pekar också på hur relationerna handlar om betydligt mer än att se matematik som något som appliceras i en yrkespraktik, utan också om, exempelvis, att matematik i sig kan förklara aspekter i bygg- och anläggningsämnet.

- This chapter addresses ways of characterising interfaces between mathematics and the educational subject of construction work in pre-vocational studies. A model is presented, which can be used for teaching and research. It is argued that the interfaces between mathematics and construction work should be viewed as a content area of its own. The authors also point to how the interfaces concern substantially more than viewing mathematics as simply being applied in a workplace practice. It may for example also be the case that mathematics in itself may explain aspects of the subject of construction work.

- Under februari 2019 togs 7 dygnsprover på det totala utgående flödet av avloppsvatten från Skånes Universitetssjukhus (SUS) i Malmö samt på det totala inkommande flödet av avlopssvatten till Sjölunda avloppsreningsverk i Malmö, som är Malmös största reningsverk. Syftet var att få kunskap om utgående koncentrationer av läkemedel från sjukhuset i relation till de inkommande koncentrationerna till avloppsreningsverket. Proverna togs från måndag till söndag under en och samma vecka och analyserades med avseende på förekomst av 33 läkemedel, antibiotika och andra mikroföroreningar. Detta innefattade 9 st antibiotika, 19 st läkemedel samt 5 andra kemikalier.
