  • Particle physics at the European Spallation Source
  • 2023
  • Ingår i: Physics reports. - : Elsevier. - 0370-1573 .- 1873-6270. ; 1023, s. 1-84
  • Forskningsöversikt (refereegranskat)abstract
    • Presently under construction in Lund, Sweden, the European Spallation Source (ESS) will be the world’s brightest neutron source. As such, it has the potential for a particle physics program with a unique reach and which is complementary to that available at other facilities. This paper describes proposed particle physics activities for the ESS. These encompass the exploitation of both the neutrons and neutrinos produced at the ESS for high precision (sensitivity) measurements (searches).

  • Societal Impacts of Storm Damage
  • 2013
  • Ingår i: Living with Storm Damage to Forests. - : European Forest Institute. - 9789525980080 - 9789525980097 ; 3, s. 70-77

    • Wind damage to forests can be divided into (1) the direct damage done to the forest and (2) indirect effects. Indirect effects may be of different kinds and may affect the environ- ment as well as society. For example, falling trees can lead to power and telecommunica- tion failures or blocking of roads. The salvage harvest of fallen trees is another example and one that involves extremely dangerous work. In this overview we provide examples of different entities, services, and activities that may be affected by wind damage to for- ests. We illustrate how valuation of the damage depends on the perspective applied and how the affected entities, services, and activities may represent different types of values. Finally we suggest means for how to actively manage the risk in an ethically sustaina- ble way. Many of our examples will be drawn from the experiences of the wind damage Gudrun in southern Sweden on 8–9 January 2005. The direct as well as indirect effects, which are described, are by no means unique to the Gudrun wind damage event and similar or even worse effects have been described after the wind damage events Martin and Lothar in 1999, and Klaus in 2009.

  • Mass spectrometry-based candidate reference measurement procedure for quantification of amyloid-β in cerebrospinal fluid.
  • 2014
  • Ingår i: Clinical chemistry. - : Oxford University Press (OUP). - 1530-8561 .- 0009-9147. ; 60:7, s. 987-94
  • Tidskriftsartikel (refereegranskat)abstract
    • Cerebrospinal fluid (CSF) amyloid-β (Aβ42) is a well-established biomarker for Alzheimer disease. Several immunoassays for Aβ42 exist but differ in absolute concentrations and may suffer from matrix interference, thereby hampering interlaboratory comparisons and the use of general cutoff levels. Together with the IFCC Working Group on CSF Proteins, we developed a candidate reference measurement procedure (RMP) for Aβ42.

  • Diagnostic Accuracy of a Plasma Phosphorylated Tau 217 Immunoassay for Alzheimer Disease Pathology
  • 2024
  • Ingår i: JAMA NEUROLOGY. - 2168-6149 .- 2168-6157.
  • Tidskriftsartikel (refereegranskat)abstract
    • ImportancePhosphorylated tau (p-tau) is a specific blood biomarker for Alzheimer disease (AD) pathology, with p-tau217 considered to have the most utility. However, availability of p-tau217 tests for research and clinical use has been limited. Expanding access to this highly accurate AD biomarker is crucial for wider evaluation and implementation of AD blood tests. ObjectiveTo determine the utility of a novel and commercially available immunoassay for plasma p-tau217 to detect AD pathology and evaluate reference ranges for abnormal amyloid beta (A beta) and longitudinal change across 3 selected cohorts. Design, Setting, and ParticipantsThis cohort study examined data from 3 single-center observational cohorts: cross-sectional and longitudinal data from the Translational Biomarkers in Aging and Dementia (TRIAD) cohort (visits October 2017-August 2021) and Wisconsin Registry for Alzheimer's Prevention (WRAP) cohort (visits February 2007-November 2020) and cross-sectional data from the Sant Pau Initiative on Neurodegeneration (SPIN) cohort (baseline visits March 2009-November 2021). Participants included individuals with and without cognitive impairment grouped by amyloid and tau (AT) status using PET or CSF biomarkers. Data were analyzed from February to June 2023. ExposuresMagnetic resonance imaging, A beta positron emission tomography (PET), tau PET, cerebrospinal fluid (CSF) biomarkers (A beta 42/40 and p-tau immunoassays), and plasma p-tau217 (ALZpath pTau217 assay). Main Outcomes and MeasuresAccuracy of plasma p-tau217 in detecting abnormal amyloid and tau pathology, longitudinal p-tau217 change according to baseline pathology status. ResultsThe study included 786 participants (mean [SD] age, 66.3 [9.7] years; 504 females [64.1%] and 282 males [35.9%]). High accuracy was observed in identifying elevated A beta (area under the curve [AUC], 0.92-0.96; 95% CI, 0.89-0.99) and tau pathology (AUC, 0.93-0.97; 95% CI, 0.84-0.99) across all cohorts. These accuracies were comparable with CSF biomarkers in determining abnormal PET signal. The detection of abnormal A beta pathology using a 3-range reference yielded reproducible results and reduced confirmatory testing by approximately 80%. Longitudinally, plasma p-tau217 values showed an annual increase only in A beta-positive individuals, with the highest increase observed in those with tau positivity. Conclusions and RelevanceThis study found that a commercially available plasma p-tau217 immunoassay accurately identified biological AD, comparable with results using CSF biomarkers, with reproducible cut-offs across cohorts. It detected longitudinal changes, including at the preclinical stage.

To evaluate references ranges for abnormal Aβ across three selected cohorts.Three single-centre observational cohorts were involved in the study: Translational Biomarkers in Aging and Dementia (TRIAD), Wisconsin Registry for Alzheimer's Prevention (WRAP), and Sant Pau Initiative on Neurodegeneration (SPIN). MRI, Aβ-PET, and tau-PET data were available for TRIAD and WRAP, while CSF biomarkers were additionally measured in a subset of TRIAD and SPIN. Plasma measurements of pTau181, pTau217 (ALZpath), pTau231, Aβ42/40, GFAP, and NfL, were available for all cohorts. Longitudinal blood biomarker data spanning 3 years for TRIAD and 8 years for WRAP were included.MRI, Aβ-PET, tau-PET, CSF biomarkers (Aβ42/40 and pTau immunoassays) and plasma pTau217 (ALZpath Simoa). The accuracy of plasma pTau217 for detecting abnormal amyloid and tau pathology. • Assessing the commutability of reference material formats for the harmonization of amyloid beta measurements.
  • 2016
  • Ingår i: Clinical chemistry and laboratory medicine : CCLM / FESCC. - : Walter de Gruyter GmbH. - 1437-4331 .- 1434-6621. ; 54:7, s. 1177-91
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: The cerebrospinal fluid (CSF) amyloid-β (Aβ42) peptide is an important biomarker for Alzheimer's disease (AD). Variability in measured Aβ42 concentrations at different laboratories may be overcome by standardization and establishing traceability to a reference system. Candidate certified reference materials (CRMs) are validated herein for this purpose. METHODS: Commutability of 16 candidate CRM formats was assessed across five CSF Aβ42 immunoassays and one mass spectrometry (MS) method in a set of 48 individual clinical CSF samples. Promising candidate CRM formats (neat CSF and CSF spiked with Aβ42) were identified and subjected to validation across eight (Elecsys, EUROIMMUN, IBL, INNO-BIA AlzBio3, INNOTEST, MSD, Simoa, and Saladax) immunoassays and the MS method in 32 individual CSF samples. Commutability was evaluated by Passing-Bablok regression and the candidate CRM termed commutable when found within the prediction interval (PI). The relative distance to the regression line was assessed. RESULTS: The neat CSF candidate CRM format was commutable for almost all method comparisons, except for the Simoa/MSD, Simoa/MS and MS/IBL where it was found just outside the 95% PI. However, the neat CSF was found within 5% relative distance to the regression line for MS/IBL, between 5% and 10% for Simoa/MS and between 10% and 15% for Simoa/MSD comparisons. CONCLUSIONS: The neat CSF candidate CRM format was commutable for 33 of 36 method comparisons, only one comparison more than expected given the 95% PI acceptance limit. We conclude that the neat CSF candidate CRM can be used for value assignment of the kit calibrators for the different Aβ42 methods.

  • Comparative study of dielectric spectroscopy measurements and anayses of oil and paper samples from a transformer undergoing repair
  • 2005
  • Ingår i: In Proc. of 19th Nordic Insulation Symposium, Trondheim, Norway, June 13-15.
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • New methods, based on characterisation of dielectric response measurements, for assessing moisture content of pressboard and paper are gaining ground. However, there is a need for calibrating results with results from other techniques. This communication presents results from a study of a 19.5/6.8/6.8 kV - 40 MVA power transformer, which was opened on-site during maintenance work, thus allowing for collecting paper samples. Dielectric spectroscopy measurements were conducted before and after its opening. Results were compared with measurements on oil and paper samples. These showed that the moisture levels in the interior of the solid insulation were unchanged. Further, it was found that FDS measurements gave more accurate estimates of moisture levels than estimates based on oil analyses and equilibrium curves.

  • Sequential Comparative Study of Dielectric Response and Analyses of Oil and Paper from a Power Transformer Undergoing Repair
  • 2005
  • Ingår i: In Proc. 14th International Symposium on High Voltage Engineering.
  • Konferensbidrag (refereegranskat)abstract
    • Methods, based on characterisation of dielectric response, for assessing moisture content in pressboard and paper are gaining ground. However, there is a need for calibrating obtained results with results from other techniques. This communication presents results from a study of a 50/10 kV - 40 MVA power transformer, which was sent to a workshop for repair, thus allowing for collecting samples of the solid insulation. Dielectric spectroscopy measurements in frequency domain were conducted at several occasions before and after the repair. Moisture estimation based on dielectric response measurements as well as on titration of oil samples yielded similar results to those obtained by direct titration of paper samples, as long as appropriate temperature values were used in all the analyses mentioned. The temperature value during measurements should be used for the analysis of the dielectric response, while in-service temperature of oil should be used for the evaluation based on Karl Fisher titration.

  • Longitudinal Data in Patients with Niemann-Pick Type C Disease Under Combined High Intrathecal and Low Intravenous Dose of 2-hydroxylpropyl-β-cyclodextrin.
  • 2021
  • Ingår i: Innovations in clinical neuroscience. - 2158-8333. ; 18:1-3, s. 11-16
  • Tidskriftsartikel (refereegranskat)abstract
    • Niemann-Pick Type C disease (NPC) is a rare, incurable, autosomal-recessive, lysosomal storage disorder with protean and progressive neurovisceral manifestations characterized by accumulation of intracellular unesterified cholesterol. The investigational use of 2-hydroxypropyl-beta-cyclodextrin (HP-β-CD) in the treatment of NPC has shown promising results in improving life expectancy and reducing neurological damage in this patient population. This case report describes two children with the neurological form of NPC: a 5-year-old male patient in advanced stage of the disease and an 11-year-old female patient in moderately advanced stage. Despite treatment with the enzyme inhibitor, miglustat, both patients continued to exhibit severe neurodegeneration. High intrathecal (900mg) and low intravenous (350-500mg/kg) doses of HP-β-CD (Trappsol®Cyclo™) were administrated twice monthly to the patients in addition to miglustat therapy. The patients were monitored clinically as well as by imaging, laboratory, and biomarker (e.g., total tau protein [T-tau]; phosphorylated tau [P-tau]; neurofilament light [NFL], oxysterols) studies over a period of 16 to 22 months. The combination therapy of miglustat and HP-β-CD resulted in disease stabilization in both patients. The combination therapy demonstrated a good safety profile, and no adverse effects on hearing were observed. Additionally, CSF biomarkers appeared useful in monitoring neuronal damage. Large, randomized studies are needed to confirm these findings.
