    • In a genome-wide scan, we show that 30 variants in 25 genomic regions are associated with risk of TN breast cancer. Women carrying many of the risk variants may have 4-fold increased risk relative to women with few variants.Triple-negative (TN) breast cancer is an aggressive subtype of breast cancer associated with a unique set of epidemiologic and genetic risk factors. We conducted a two-stage genome-wide association study of TN breast cancer (stage 1: 1529 TN cases, 3399 controls; stage 2: 2148 cases, 1309 controls) to identify loci that influence TN breast cancer risk. Variants in the 19p13.1 and PTHLH loci showed genome-wide significant associations (P < 5 x 10(-) (8)) in stage 1 and 2 combined. Results also suggested a substantial enrichment of significantly associated variants among the single nucleotide polymorphisms (SNPs) analyzed in stage 2. Variants from 25 of 74 known breast cancer susceptibility loci were also associated with risk of TN breast cancer (P < 0.05). Associations with TN breast cancer were confirmed for 10 loci (LGR6, MDM4, CASP8, 2q35, 2p24.1, TERT-rs10069690, ESR1, TOX3, 19p13.1, RALY), and we identified associations with TN breast cancer for 15 additional breast cancer loci (P < 0.05: PEX14, 2q24.1, 2q31.1, ADAM29, EBF1, TCF7L2, 11q13.1, 11q24.3, 12p13.1, PTHLH, NTN4, 12q24, BRCA2, RAD51L1-rs2588809, MKL1). Further, two SNPs independent of previously reported signals in ESR1 [rs12525163 odds ratio (OR) = 1.15, P = 4.9 x 10(-) (4)] and 19p13.1 (rs1864112 OR = 0.84, P = 1.8 x 10(-) (9)) were associated with TN breast cancer. A polygenic risk score (PRS) for TN breast cancer based on known breast cancer risk variants showed a 4-fold difference in risk between the highest and lowest PRS quintiles (OR = 4.03, 95% confidence interval 3.46-4.70, P = 4.8 x 10(-) (69)). This translates to an absolute risk for TN breast cancer ranging from 0.8% to 3.4%, suggesting that genetic variation may be used for TN breast cancer risk prediction.

  • Trawling-induced change in benthic effect trait composition – A multiple case study

    • Introduction: The importance of the response-effect trait dichotomy in marine benthic ecology has garnered recent attention. Response traits, characterising species responses to environmental variations, have been a dominant focus in the development of ecological indicators for ecosystem health assessment. In contrast, effect traits, expressing effects of organism activities on the ecosystem, still do not benefit from an equal interest in spite of the complementary facet that they provide to complete our understanding of functional diversity and ecosystem vulnerability. In this study, we explore the consequences of disturbance by bottom trawl fisheries on benthic effect trait composition.Methods: To this end, we used different contexts of environmental and trawling conditions from thirteen case studies in European waters and apply the same analytical procedure to derive a gradient that solely account for trawling-induced disturbance (Partial RLQ analysis).Results: Bottom trawling was found to be a selective force of benthic effect trait composition in a majority of case studies. In general, tube-dwelling species were more typical of low trawling frequencies, whereas deep burrowing species were more resistant at high trawling frequencies. Although we report significantly deleterious effects of trawling on benthic ecosystem functions, the effect trait pattern along the gradient was never related to life span, a key response trait generally assumed to express recoverability following disturbance. Furthermore, we show that trends in species multi-functionality and community functional diversity can be negative or positive along the trawling intensity gradient.Discussion: We discuss the relevance of these results in light of recent developments in the framework of response and effect trait dichotomy, and provide guidelines of trait data analysis in the context of trawl fisheries impact on the sea floor. Our findings emphasize the importance of fundamental concepts from functional ecology in this context and represent a first step toward an assessment of trawling effect more oriented on benthos-mediated biogeochemical processes.

  • Different Strategies for Transient Control of the Air-Fuel Ratio in a SI Engine

    • This paper compares several strategies for air-fuel ratio tran-sient control. The strategies are: A factory-standard look-up table based system (a SAAB Trionic 5), a feedback PI controller with and without feed-forward throttle correction, a linear feed-forward control algorithm, and two nonlinear feed- forward algorithms based on artificial neural networks. The control strategies have been implemented and evaluated in a SAAB 9000 car during a transient driving test, consisting of an acceleration in the second gear from an engine speed of 1500 rpm to 3000 rpm. The best strategies are found to be the neural network based ones, followed by the table based factory system. The two feedback PI controllers offer the poorest performance.

  • Structural and tribological properties of cluster-assembled CNx films

    • We report the structural and tribological characterization of nanostructured CNx thin films produced by the deposition of a supersonic carbon cluster beam assisted by nitrogen ion bombardment. The influence of the deposition parameters on the chemical composition and structure of the films has been systematically studied by X-ray photoelectron spectroscopy, elastic recoil detection analysis, transmission electron microscopy and atomic force microscopy. Depending on the deposition parameters, the films show a structure ranging from amorphous to disordered graphitic with interlinked planes. Nitrogen content depends on the nitrogen ion kinetic energy. The films have a very low density with a high surface roughness. Friction measurements at the nanoscale show a correlation between nitrogen content and mechanical properties of the system.

  • A System Perspective on Energy End-Use Measures in a District Heated Region : Renovation of Buildings and Hydronic Pavement Systems

    • A district heating and cooling (DHC) system can be a viable piece of the puzzle in the efforts of reducing the greenhouse gas (GHG) emissions. Especially if the DHC system include combined heat and power (CHP) plants which enable electricity production from renewable resources. This is set forth in national energy targets and sustainable development goals (SDGs), adopted by the United Nations in 2015. Moreover, improved energy efficiency and energy savings are important factors in fulfilling the national targets of decreased energy intensity as well as reducing the use of fossil fuels.The aim of this thesis is to analyze the impacts of two energy end-use measures in a DHC network and their consequences on the efforts towards sustainable development. The end-use measures studied are (1) renovation of a multifamily building stock and (2) the use of a hydronic pavement system (HPS) including analysis of different control strategies. The end-use measures are assessed in terms of energy use and efficiency, use of renewable and fossil resources, and local and global GHG emissions. Lastly, it is analyzed how the results relate to national energy targets and SDGs. By using simulation and optimization models, several scenarios of end-use measures are analyzed in the two studies. In the first study, six scenarios are analyzed, as the renovation packages include measures on the envelope, ventilation and conversion from district heating to ground source heat pump. In the second study three scenarios are analyzed, where the HPS are operated all-time at a temperature below 4°C or are shut down at temperatures below -10°C or at temperatures below -5°C.The results of the study regarding the renovation of a multifamily building stock indicate a future reduction in heat demand. All scenarios show energy savings of the studied building, which ranged from 11% to 56%. All scenarios show a reduction in local GHG emissions, as well as reduced fossil fuel use. Although the largest reduction was found in the use of renewable resources. From a global perspective on GHG emissions, the scenarios with district heating out-performed measures with heat pump solutions in the studied system. Moreover, the study point to positive impacts on the efforts towards SDGs.To mitigate the reduced heat demand from the renovation of the building stock, an HPS may be used. The results show mostly renewable resources were used for the HPS. The use of HPS was found to generate a positive impact on global GHG emissions. A control strategy that shuts down the HPS at temperatures below -10°C would result in 10% energy saving and would maintain acceptable performance of the HPS. Furthermore, it would reduce the use of fossil fuel and reduce local GHG emissions by 25%. Moreover, an HPS may contribute to SDGs.It is concluded that energy end-use measures of renovating a multifamily building stock are vital in the work towards an improved energy intensity. However, these measures result in a decreased demand for heat in the DHC network. This can then lead to reduced electricity production from renewable resources in the CHP plants, which in turn have a negative impact on the global GHG emissions. By finding new applications, like HPS, the infrastructure of DHC networks could be utilized efficiently and serve as one piece of the puzzle that is the efforts towards sustainable development.

  • Analysing Conflicting Energy and Climate Targets between Municipalities, Energy Utilities and Housing Companies in Sweden

    • To achieve the Sustainable Development Goals (SDGs), it is important that the SDGs are integrated atboth national and municipal levels, and that the business sector is involved in this work. This paper analyses howwell the SDGs are integrated as well if there are any conflicting energy and climate targets among municipalities,energy utilities and housing companies in Sweden, and in relation to national and EU targets. The targets arecategorized as; climate impact, efficient energy use, and share of renewable resources. Three key indicators arein focus; timeline and level of targets, terminology, and system boundaries. The study focuses on the Linköpingand Norrköping region, which has 300,000 inhabitants and is located 150 km southwest of Stockholm, Sweden.By using directives, company annual reports and interviews, the companies’ decision-making processes andtargets have been mapped.The results identify differences in system boundaries and timelines as reasons to potential conflictsregarding climate targets. Ambitious targets at the municipal level are identified as a driving force, but with targetfulfilment two decades or more before national and EU targets. Regarding the category of efficient energy use,the level of targets and terminology differs. Lastly, regarding renewable resources, terminology and timelinediffer. Furthermore, it is concluded that the SDGs are not integrated to a large extent in the business sector orat a local level. Overall, this may create challenges in communication, collaboration and exchange of knowledgein order to succeed and achieve the SDGs.
