  • Blood-based epigenetic estimators of chronological age in human adults using DNA methylation data from the Illumina MethylationEPIC array

    • Background: Epigenetic clocks have been recognized for their precise prediction of chronological age, age-related diseases, and all-cause mortality. Existing epigenetic clocks are based on CpGs from the Illumina HumanMethylation450 BeadChip (450 K) which has now been replaced by the latest platform, Illumina MethylationEPIC BeadChip (EPIC). Thus, it remains unclear to what extent EPIC contributes to increased precision and accuracy in the prediction of chronological age. Results: We developed three blood-based epigenetic clocks for human adults using EPIC-based DNA methylation (DNAm) data from the Norwegian Mother, Father and Child Cohort Study (MoBa) and the Gene Expression Omnibus (GEO) public repository: 1) an Adult Blood-based EPIC Clock (ABEC) trained on DNAm data from MoBa (n = 1592, age-span: 19 to 59 years), 2) an extended ABEC (eABEC) trained on DNAm data from MoBa and GEO (n = 2227, age-span: 18 to 88 years), and 3) a common ABEC (cABEC) trained on the same training set as eABEC but restricted to CpGs common to 450 K and EPIC. Our clocks showed high precision (Pearson correlation between chronological and epigenetic age (r) > 0.94) in independent cohorts, including GSE111165 (n = 15), GSE115278 (n = 108), GSE132203 (n = 795), and the Epigenetics in Pregnancy (EPIPREG) study of the STORK Groruddalen Cohort (n = 470). This high precision is unlikely due to the use of EPIC, but rather due to the large sample size of the training set. Conclusions: Our ABECs predicted adults’ chronological age precisely in independent cohorts. As EPIC is now the dominant platform for measuring DNAm, these clocks will be useful in further predictions of chronological age, age-related diseases, and mortality.

  • Prehospital Identification of Patients with a Final Hospital Diagnosis of Stroke.

    • Introduction the early phase of stroke, minutes are critical. Since the majority of patients with stroke are transported by the Emergency Medical Service (EMS), the early handling and decision making by the EMS clinician is important. Problem The study aim was to evaluate the frequency of a documented suspicion of stroke by the EMS nurse, and to investigate differences in the clinical signs of stroke and clinical assessment in the prehospital setting among patients with regard to if there was a documented suspicion of stroke on EMS arrival or not, in patients with a final hospital diagnosis of stroke.METHODS: The study had a retrospective observational design. Data were collected from reports on patients who were transported by the EMS and had a final diagnosis of stroke at a single hospital in western Sweden (630 beds) in 2015. The data sources were hospital and prehospital medical journals.RESULTS: In total, 454 patients were included. Among them, the EMS clinician suspected stroke in 52%. The findings and documentation on patients with a suspected stroke differed from the remaining patients as follows: a) More frequently documented symptoms from the face, legs/arms, and speech; b) More frequently assessments of neurology, face, arms/legs, speech, and eyes; c) More frequently addressed the major complaint with regard to time and place of onset, duration, localization, and radiation; d) Less frequently documented symptoms of headache, vertigo, and nausea; and e) More frequently had an electrocardiogram (ECG) recorded and plasma glucose sampled. In addition to the 52% of patients who had a documented initial suspicion of stroke, seven percent of the patients had an initial suspicion of transitory ischemic attack (TIA) by the EMS clinician, and a neurologist was approached in another 10%.CONCLUSION: Among 454 patients with a final diagnosis of stroke who were transported by the EMS, an initial suspicion of stroke was not documented in one-half of the cases. These patients differed from those in whom a suspicion of stroke was documented in terms of limited clinical signs of stroke, a less extensive clinical assessment, and fewer clinical investigations.   • Flexographic Ink-Coating Interactions : Effects of Porous Structure Variations of Coated Paperboard

    • Ink transfer and ink penetration into a coated surface, and variations thereof, influences the print density, mottling and dot gain, which affects the achievable print quality and visual appearance. The pressure in the printing nip and the porosity of the substrate are conditions and properties that will regulate the amount of ink that penetrates into a porous coating structure. The purpose of this study was to relate print quality aspects to ink penetration of water-based flexographic ink into calcium carbonate based coatings of differently engineered structures. Pilot-coated paper-boards with different coating porosities were printed in a laboratory flexographic printer. Results indicate that ink transfer distribution is strongly affected by the roughness and the porosity of the substrate. A coating layer of broad pigment particle size distribution resulted in a lower print density, compared to coatings of narrowly distributed particle sizes. A structure characterized by larger pore volume and greater dominating pore radius, showed a higher amount of z-directional ink penetration, which was supported by estimating the penetration using a physical model accounting for both capillary- and pressure driven penetration. A coating with narrow particle size distribution also showed a lower dot gain.

  • Optics of coated paperboard : Aspects of surface treatment on porous structures

    • Calendering of coated and uncoated paper is widely used to enhance optical properties such as gloss and print quality. The aim of this thesis is to characterize coatings and prints, and to validate models using experimental results from optical measurements of physical samples. Calendering of coated paper often leads to a brightness decrease. The mechanism for this is not altogether clear. One common explanation is that the porosity of the coating layer decreases and hence the scattering decreases. By comparing simulated and measured results it was shown that modifications of the surface properties account for the brightness decrease of ground calcium carbonate coated substrates with calendering. Monte Carlo light scattering simulations, taking into account the measured decrease of surface microroughness and increased effective refractive index, showed that surface modifications accounted for most of the observed brightness decrease of the ground calcium carbonate coated substrate, whereas the bulk scattering and absorption coefficients were not affected by calendering. It was also shown that the scattering coefficient is significantly dependent on the coat weight whereas the physical absorption coefficient is not. The penetration of ink in the z-direction of a substrate influences the quality of the print. The ink penetration affects print density, mottling and dot gain, common print effects that influence the preference of consumers. The pressure in the printing nip and the porosity of the substrate both affect the amount of ink that is pressed into the porous structure of a coating layer during printing. By printing pilot coated paperboard with different coating porosity and measuring the resulting optical properties of the prints, a basis for simulations of the different layers, that is to say the coating, the print and the mixed layer in between, was created. Results show that ink distribution is strongly affected by the roughness of the substrate. Fibres and fibre flocks underneath the two coating layers created an unevenly distributed coating thickness that affected the print quality. Differences in pore size and pore size distribution also affected the behaviour of the ink. A coating layer of broad pigment particle size distribution resulted in a relatively low print density, in comparison to coatings of narrowly distributed particle sizes. Comparison of dot gain showed that the coating layer of a narrow particle size distribution had a relatively low dot gain compared to others. In this work, these results are explained by the differences in ink distributions on and in the coating layers.
