    • Introduction: Nitrogen (N) gradient studies in some cases use N deposition inthroughfall as measure of N deposition to forests.For evaluating critical loadsof N, however, information on total N deposition is required, i.e., the sum ofestimates of dry, wet and occult deposition.Methods: The present paper collects a number of studies in Europe wherethroughfall and total N deposition were compared in different forest types.From this dataset a function was derived which allows to estimate total Ndeposition from throughfall N deposition.Results: At low throughfall N deposition values, the proportion of canopyuptake is high and thus the underestimation of total deposition by throughfallN needs to be corrected.At throughfall N deposition values > 20 kg Nha?1 yr?1 canopy uptake is getting less important.Conclusion: This work shows that throughfall clearly underestimates totaldeposition of nitrogen. With the present data set covering large parts of Europe it is possible to derive a critical load estimate from gradient studiesusing throughfall data.

  • Dendrochemical indicators of tree rings reveal historical soil acidification in Swiss forest stands

    • The deposition of acidifying nitrogen and sulphur compounds from agriculture and fossil fuel combustion has drastically altered the chemical balance of forest soils in many regions of the world, leading to soil acidification with negative impacts on nutrient availability and thus also on tree vitality. The change of nutrient concentrations in the soil solution can be assessed by long-term investigations, however meaningful indicators, reflecting environmental changes, are needed to compare the current nutrient status with past values. We used dendrochemical indicators in stem wood of different tree species to access the impact of acidifying depositions on soil quality and tree nutrition. We selected 328 stem wood samples from 96 trees of Norway spruce (Picea abies), European beech (Fagus sylvatica), Sessile oak (Quercus petrea) and English oak (Quercus robur) from 22 forest sites, which are part of the long-term Intercantonal Forest Observation Program in Switzerland. Four time periods of 20 years were defined according to the emissions of air pollutants between 1910 and 2017. Our results showed a trend of increasing Al concentrations in tree rings of spruce peaking in the most recent time period (2000–2017). Mn and Ca concentrations in spruce and beech wood have decreased significantly throughout the time period 1910–2017. These dendrochemical indicators depended on the soil pH, with higher Al and lower Mn and Ca concentrations for soils with a low pH (pH<4.2). In oak trees the observed dendrochemical changes are confounded with dendrochemical differences between heartwood and sapwood. K and Mg showed inconsistent patterns in all three tree species, which are probably caused by translocation within the stem discs. With the use of piecewise structural equation models (SEM) we highlighted the direct and indirect influences of N deposition on element concentrations in stem wood. The data suggest a relation between increased N deposition and lower base saturation values in the forest soils for all three tree species, which were linked to higher Al concentrations in spruce and lower Mn concentrations in spruce and beech. The relation between Al concentrations in tree rings of Norway spruce and measured base saturation was used to reconstruct past soil base saturation values. It revealed a progressive soil acidification in the long-term forest observation sites. These reconstructed base saturation values were further used to validate modelled values from dynamic biogeochemical models such as SAFE/ForSAFE. This comparison pointed out possible shortcomings such as the lack of organic complexation in those models. Taken together, our analyses showed that element concentrations of Al, Mn, Ca in Norway spruce and European beech stem wood were suitable dendrochemical indicators of environmental change due to soil acidification, as they reflect both direct and indirect effects of air pollutants and chemical soil properties.

    • A dynamic model of forest ecosystems was used to investigate the effects of climate change, atmospheric deposition and harvest intensity on 48 forest sites in Sweden (n = 16) and Switzerland (n = 32). The model was used to investigate the feasibility of deriving critical loads for nitrogen (N) deposition based on changes in plant community composition. The simulations show that climate and atmospheric deposition have comparably important effects on N mobilization in the soil, as climate triggers the release of organically bound nitrogen stored in the soil during the elevated deposition period. Climate has the most important effect on plant community composition, underlining the fact that this cannot be ignored in future simulations of vegetation dynamics. Harvest intensity has comparatively little effect on the plant community in the long term, while it may be detrimental in the short term following cutting. This study shows: that critical loads of N deposition can be estimated using the plant community as an indicator; that future climatic changes must be taken into account; and that the definition of the reference deposition is critical for the outcome of this estimate.     • The ForSAFE-VEG model was used to investigate the impacts of climate change and air pollution scenarios on soil chemistry and ground vegetations composition. In particular, the model involves a ground vegetation model incorporating plant changes to ambient site conditions in terms of climate and chemistry, but the model also incorporate competition between the different plant groups. The model was validated against observed values and reproduced observations of tree growth, soil chemistry, and ground vegetation compositions to satisfaction. The results show that the ground vegetation reacts strongly to changes in air pollution, in particular nitrogen as well as to climate change with major shifts in plant composition. A procedure for estimating critical loads for nitrogen, using ground vegetation biodiversity as criterion, was tested and the method seems operable. It suggests that if we want to protect the present biodiversity of the ground vegetation, this will face significant difficulties because of permanent climate change that induced changes in the ecosystem. We conclude that the reference state for ground vegetation biodiversity is rather to be sought for in the future, hopefully using models, than in the past or present.

    • We have investigated the regulation of hormone secretion from rat pancreatic islets by the GABAB receptors (GABABRs). Inclusion of the specific GABABR antagonist CGP 55845 in the extracellular medium increased glucose-stimulated insulin secretion 1.6-fold but did not affect the release of glucagon and somatostatin. Conversely, addition of the GABABR agonist baclofen inhibited glucose-stimulated insulin secretion by ∼60%. Using RT-PCR, transcription of GABABR1a-c,f and GABABR2 subunits was detected in β-cells. Measurements of membrane currents and cell capacitance were applied to single β-cells to investigate the mechanisms by which GABABR activation inhibits insulin secretion. In perforated-patch measurements, baclofen inhibited exocytosis elicited by 500-ms voltage-clamp depolarizations to 0 mV by ≤ 80% and voltage-gated Ca2+ entry by only ∼30%. Both effects were concentration-dependent with IC50 values of ∼2 μm. The inhibitory action of baclofen was abolished in the presence of CGP 55845. The ability of baclofen to suppress exocytosis was prevented by pre-treatment with pertussis toxin and by inclusion of GDPβS in the intracellular medium, and became irreversible in the presence of GTPγS as expected for a process involving inhibitory G-proteins (Gi/o-proteins). The inhibitory effect of baclofen resulted from activation of the serine/threonine protein phosphatase calcineurin and pre-treatment with cyclosporin A or intracellular application of calcineurin autoinhibitory peptide abolished the effect. Addition of baclofen had no effect on [Ca2+]i and electrical activity in glucose-stimulated β-cells. These data indicate that GABA released from β-cells functions as an autocrine inhibitor of insulin secretion in pancreatic islets and that the effect is principally due to direct suppression of exocytosis.

    • © 2017 Elsevier B.V.For human health studies, epidemiology has been established as important tool to examine factors that affect the frequency and distribution of disease, injury, and other health-related events in a defined population, serving the purpose of establishing prevention and control programs. On the other hand, gradient studies have a long tradition in the research of air pollution effects on plants. While there is no principal difference between gradient and epidemiological studies, the former address more one-dimensional transects while the latter focus more on populations and include more experience in making quantitative predictions, in dealing with confounding factors and in taking into account the complex interplay of different factors acting at different levels. Epidemiological analyses may disentangle and quantify the contributions of different predictor variables to an overall effect, e.g. plant growth, and may generate hypotheses deserving further study in experiments. Therefore, their use in ecosystem research is encouraged. This article provides a number of recommendations on: (1) spatial and temporal aspects in preparing predictor maps of nitrogen deposition, ozone exposure and meteorological covariates; (2) extent of a dataset required for an analysis; (3) choice of the appropriate regression model and conditions to be satisfied by the data; (4) selection of the relevant explanatory variables; (5) treatment of interactions and confounding factors; and (6) assessment of model validity.

    • Radial stem growth dynamics at seasonal resolution are essential to understand how forests respond to climate change. We studied daily radial growth of 160 individuals of seven temperate tree species at 47 sites across Switzerland over 8 years. Growth of all species peaked in the early part of the growth season and commenced shortly before the summer solstice, but with species-specific seasonal patterns. Day length set a window of opportunity for radial growth. Within this window, the probability of daily growth was constrained particularly by air and soil moisture, resulting in intermittent growth to occur only on 29 to 77 days (30% to 80%) within the growth period. The number of days with growth largely determined annual growth, whereas the growth period length contributed less. We call for accounting for these non-linear intra-annual and species-specific growth dynamics in tree and forest models to reduce uncertainties in predictions under climate change.

  • Update of EULAR recommendations for the treatment of systemic sclerosis

    • The aim was to update the 2009 European League against Rheumatism (EULAR) recommendations for the treatment of systemic sclerosis (SSc), with attention to new therapeutic questions. Update of the previous treatment recommendations was performed according to EULAR standard operating procedures. The task force consisted of 32 SSc clinical experts from Europe and the USA, 2 patients nominated by the pan-European patient association for SSc (Federation of European Scleroderma Associations (FESCA)), a clinical epidemiologist and 2 research fellows. All centres from the EULAR Scleroderma Trials and Research group were invited to submit and select clinical questions concerning SSc treatment using a Delphi approach. Accordingly, 46 clinical questions addressing 26 different interventions were selected for systematic literature review. The new recommendations were based on the available evidence and developed in a consensus meeting with clinical experts and patients. The procedure resulted in 16 recommendations being developed (instead of 14 in 2009) that address treatment of several SSc-related organ complications: Raynaud's phenomenon (RP), digital ulcers (DUs), pulmonary arterial hypertension (PAH), skin and lung disease, scleroderma renal crisis and gastrointestinal involvement. Compared with the 2009 recommendations, the 2016 recommendations include phosphodiesterase type 5 (PDE-5) inhibitors for the treatment of SSc-related RP and DUs, riociguat, new aspects for endothelin receptor antagonists, prostacyclin analogues and PDE-5 inhibitors for SSc-related PAH. New recommendations regarding the use of fluoxetine for SSc-related RP and haematopoietic stem cell transplantation for selected patients with rapidly progressive SSc were also added. In addition, several comments regarding other treatments addressed in clinical questions and suggestions for the SSc research agenda were formulated. These updated data-derived and consensus-derived recommendations will help rheumatologists to manage patients with SSc in an evidence-based way. These recommendations also give directions for future clinical research in SSc.

    • γ-Aminobutyric acid (GABA) has been proposed to function as a paracrine signaling molecule in islets of Langerhans. We have shown that rat β-cells release GABA by Ca2+-dependent exocytosis of synaptic-like microvesicles. Here we demonstrate that GABA thus released can diffuse over sufficient distances within the islet interstitium to activate GABAA receptors in neighboring cells. Confocal immunocytochemistry revealed the presence of GABAA receptors in glucagon-secreting α-cells but not in β- and δ-cells. RT-PCR analysis detected transcripts of α1 and α4 as well as β1–3 GABAA receptor subunits in purified α-cells but not in β-cells. In whole-cell voltage-clamp recordings, exogenous application of GABA activated Cl− currents in α-cells. The GABAA receptor antagonist SR95531 was used to investigate the effects of endogenous GABA (released from β-cells) on pancreatic islet hormone secretion. The antagonist increased glucagon secretion at 1 mmol/l glucose twofold and completely abolished the inhibitory action of 20 mmol/l glucose on glucagon release. Basal and glucose-stimulated secretion of insulin and somatostatin were unaffected by SR95531. The L-type Ca2+ channel blocker isradipine evoked a paradoxical stimulation of glucagon secretion. This effect was not observed in the presence of SR95531, and we therefore conclude that isradipine stimulates glucagon secretion by inhibition of GABA release.

  • Why trees grow at night

    • The timing of diel stem growth of mature forest trees is still largely unknown, as empirical data with high temporal resolution have not been available so far. Consequently, the effects of day-night conditions on tree growth remained uncertain. Here we present the first comprehensive field study of hourly-resolved radial stem growth of seven temperate tree species, based on 57 million underlying data points over a period of up to 8 yr. We show that trees grow mainly at night, with a peak after midnight, when the vapour pressure deficit (VPD) is among the lowest. A high VPD strictly limits radial stem growth and allows little growth during daylight hours, except in the early morning. Surprisingly, trees also grow in moderately dry soil when the VPD is low. Species-specific differences in diel growth dynamics show that species able to grow earlier during the night are associated with the highest number of hours with growth per year and the largest annual growth increment. We conclude that species with the ability to overcome daily water deficits faster have greater growth potential. Furthermore, we conclude that growth is more sensitive than carbon uptake to dry air, as growth stops before stomata are known to close.
