  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Carotid-Femoral Pulse Transit Time Variability Predicted Mortality and Improved Risk Stratification in the Elderly

    • The carotid-to-femoral pulse wave velocity, determined by pulse transit time (PTT) and distance, is a well-established measure of arterial stiffness and predicts adverse outcomes. However, its predictive value decreases with aging. To explore new risk indicator in the elderly, we investigated if the variation of carotid-to-femoral pulse wave velocity, registered as beat-to-beat variability of carotid-to-femoral PTT (cf-PTT), could predict outcome. Totally 3015 (median age, 72.4 years; 39.6% men) and 1181 (75.6 years; 42.2% men) subjects from communities of Malmö, Sweden, and Shanghai, China, were analyzed, respectively. Continuous pulse waves for 10 seconds were recorded sequentially at carotid and femoral arterial sites with applanation tonometry (SphygmoCor, Atcor, Australia). During a median of 6.6 and 10.2 years, 389 and 427 deaths occurred in the Malmö and Shanghai cohorts, respectively. Each one-SD increase in the log-transformed coefficient of variation of cf-PTT was associated with 24% (95% CI, 13%–37%) and 21% (10%–33%) increased risk for all-cause mortality in the Malmö and Shanghai subjects, and 60% (33%–91%) for cardiovascular mortality in the Malmö subjects. Adding the coefficient of variation of cf-PTT to the models including conventional risk factors and carotid-to-femoral pulse wave velocity significantly (P<0.05) improved prediction for all-cause mortality in both cohorts (integrated discrimination improvement, 0.005–0.008) and cardiovascular mortality in the Malmö cohort (net reclassification improvement, 0.206). In both cohorts, a coefficient of variation of cf-PTT <6% was not associated with increased mortality risk. In conclusion, the beat-to-beat variability of cf-PTT predicted mortality and improved risk stratification, which might be a novel risk indicator for elderly people.

  • Microstructure and Mechanical Properties of High Chromium Steel Weld Metals

    • The microstructure of three types of Cr steel weld metals, containing 5%, 9% and 12% chromium, was studied in different heat treated conditions using optical microscopy, analytical transmission electron microscopy and atom probe field-ion microscopy. The mechanical properties of the weld metals were investigated. Scanning electron microscopy (SEM) and optical microscopy were used to carry out the fractography investigation of the impacted specimens. In the 5% Cr steel weld metal, it was found that M7C3 and M2C started to precipitate during tempering at 500 °C. M23C6 precipitated out from 600 °C. The composition of the precipitates also changed according to the tempering temperature. The development of microstructure during tempering was discussed. The microstructure of three 9% Cr steel weld metals consisted of lath martensite, some retained .delta.-ferrite and precipitates. The predominant precipitates were M23C6. In weld metals containing V and Nb, MC and M2C were present. The failure mode of the impacted weld metals was quasi-cleavage. Non- metallic inclusions were found to be the nucleation sites for cleavage cracks during impact. The cleavage stress of the weld metals was determined by the size of the inclusions and the size of martensite packets or recrystallized ferrite grains (in the case that the quantity of recrystallized ferrite was high). The impact toughness of the weld metals depended on the yield stress and fracture stress of the materials. The microstructure of three varieties of 12% Cr steel weld metals consisted also of martensite, retained .delta.-ferrite and precipitates. The precipitates included M23C6, MX and M2X. The morphology, quantity and distribution of the precipitates were determined by composition and heat treatment of the weld metals. The microstructure development in the weld metals in different heat treatment conditions was investigated. The failure mode of impacted specimens was mainly cleavage. Intercooling the weld metals to 125 °C before post-weld heat treatment did not affect the tensile strength of the weld metals very much, but improved their impact toughness dramatically. The mechanism by which the microstructure of the weld metals affects their mechanical properties was discussed.

    • Field monitoring carried out in a deep tunnel of the Dingji coal mine in China confirmed the zonal disintegration phenomenon by using the borehole TV. Based on field monitoring, an analogical model test was conducted to research the fracture shape and forming conditions of the rock mass in the Dingji mine. To perform the model test, an analogical material and optical sensor were developed independently. Through the test, the occurrence of zonal disintegration was confirmed and the forming process was monitored. The fracture pattern of zonal disintegration was determined, and the radii of the fractured zones were found to fulfil the relationship of geometric progression. The displacement laws of surrounding rocks during zonal disintegration were obtained and found to be non-monotonic. The test results are in agreement with the field-monitoring results. Through a theory analysis based on fracture mechanics, the mechanism of zonal disintegration was revealed. The fracture zones occur as circles concentric to the cavern periphery, which is the “false face”. Each fracture zone ruptures at the elastic–plastic boundary of surrounding rocks and then coalesces into a circle. The geometric progression ratio was determined; it is related to the mechanical parameters and ground stress of the surrounding rocks and calculated as follows: And the mechanism of the non-monotonic displacement law is revealed; the continuous formation of the “false face” causes the geostress redistribution and crack opening.

  • Outcome-Driven Thresholds for Ambulatory Blood Pressure Based on the New American College of Cardiology/American Heart Association Classification of Hypertension

    • The new American College of Cardiology/American Heart Association guideline reclassified office blood pressure and proposed thresholds for ambulatory blood pressure (ABP). We derived outcome-driven ABP thresholds corresponding with the new office blood pressure categories. We performed 24-hour ABP monitoring in 11 152 participants (48.9% women; mean age, 53.0 years) representative of 13 populations. We determined ABP thresholds resulting in multivariable-adjusted 10-year risks similar to those associated with elevated office blood pressure (120/80 mm Hg) and stages 1 and 2 of office hypertension (130/80 and 140/90 mm Hg). Over 13.9 years (median), 2728 (rate per 1000 person-years, 17.9) people died, 1033 (6.8) from cardiovascular disease; furthermore, 1988 (13.8), 893 (6.0), and 795 (5.4) cardiovascular and coronary events and strokes occurred. Using a composite cardiovascular end point, systolic/diastolic outcome-driven thresholds indicating elevated 24-hour, daytime, and nighttime ABP were 117.9/75.2, 121.4/79.6, and 105.3/66.2 mm Hg. For stages 1 and 2 ambulatory hypertension, thresholds were 123.3/75.2 and 128.7/80.7 mm Hg for 24-hour ABP, 128.5/79.6 and 135.6/87.1 mm Hg for daytime ABP, and 111.7/66.2 and 118.1/72.5 mm Hg for nighttime ABP. ABP thresholds derived from other end points were similar. After rounding, approximate thresholds for elevated 24-hour, daytime, and nighttime ABP were 120/75, 120/80, and 105/65 mm Hg, and for stages 1 and 2, ambulatory hypertension 125/75 and 130/80 mm Hg, 130/80 and 135/85 mm Hg, and 110/65 and 120/70 mm Hg. Outcome-driven ABP thresholds corresponding to elevated blood pressure and stages 1 and 2 of hypertension are similar to those proposed by the current American College of Cardiology/American Heart Association guideline.

  • Characterization of cross-species transcription and splicing from Penicillium to Saccharomyces cerevisiae

    • Heterologous expression of eukaryotic gene clusters in yeast has been widely used for producing high-value chemicals and bioactive secondary metabolites. However, eukaryotic transcription cis-elements are still undercharacterized, and the cross-species expression mechanism remains poorly understood. Here we used the whole expression unit (including original promoter, terminator, and open reading frame with introns) of orotidine 5'-monophosphate decarboxylases from 14 Penicillium species as a showcase, and analyzed their cross-species expression in Saccharomyces cerevisiae. We found that pyrG promoters from the Penicillium species could drive URA3 expression in yeast, and that inefficient cross-species splicing of Penicillium introns might result in weak cross-species expression. Thus, this study demonstrates cross-species expression from Penicillium to yeast, and sheds light on the opportunities and challenges of cross-species expression of fungi expression units and gene clusters in yeast without refactoring for novel natural product discovery.
