    • In this work, we present a new catechol amperometric biosensor fabricated on the basis of naturally available enzymes in common mushrooms. The biosensor response mechanism comprises the reduction of the quinone exclusively produced in the oxidation of the catechol present in the sample, which is catalyzed by tyrosinase enzyme. The new catechol biosensor has demonstrated excellent analytical performance at increasing catechol concentrations in the sample solution, which includes superior reproducibility for several electrodes and long-term stability. On top of that, the biosensing element used in the fabrication is a sustainable material, of low-cost and presents an excellent lifetime of years. Whether the catechol biosensor is operating in the presence of a compound influencing the reactions underlying the amperometric response (such as ascorbic, benzoic, gallic and kojic acids), this serves as an analytical platform to detect these compounds in real samples. Particularly, we introduce herein for the first time different treatments to process the current signal of the biosensor pursuing the linearity needed for the analytical application in real samples. In this sense, the catechol biosensor has been successfully applied to the detection of benzoic, gallic and kojic acids in juices, teas and cosmetic products, respectively.

  • EuroPineDB : a high-coverage web database for maritime pine transcriptome

    • BACKGROUND: Pinus pinaster is an economically and ecologically important species that is becoming a woody gymnosperm model. Its enormous genome size makes whole-genome sequencing approaches are hard to apply. Therefore, the expressed portion of the genome has to be characterised and the results and annotations have to be stored in dedicated databases. DESCRIPTION: EuroPineDB is the largest sequence collection available for a single pine species, Pinus pinaster (maritime pine), since it comprises 951 641 raw sequence reads obtained from non-normalised cDNA libraries and high-throughput sequencing from adult (xylem, phloem, roots, stem, needles, cones, strobili) and embryonic (germinated embryos, buds, callus) maritime pine tissues. Using open-source tools, sequences were optimally pre-processed, assembled, and extensively annotated (GO, EC and KEGG terms, descriptions, SNPs, SSRs, ORFs and InterPro codes). As a result, a 10.5× P. pinaster genome was covered and assembled in 55 322 UniGenes. A total of 32 919 (59.5%) of P. pinaster UniGenes were annotated with at least one description, revealing at least 18 466 different genes. The complete database, which is designed to be scalable, maintainable, and expandable, is freely available at: http://www.scbi.uma.es/pindb/. It can be retrieved by gene libraries, pine species, annotations, UniGenes and microarrays (i.e., the sequences are distributed in two-colour microarrays; this is the only conifer database that provides this information) and will be periodically updated. Small assemblies can be viewed using a dedicated visualisation tool that connects them with SNPs. Any sequence or annotation set shown on-screen can be downloaded. Retrieval mechanisms for sequences and gene annotations are provided. CONCLUSIONS: The EuroPineDB with its integrated information can be used to reveal new knowledge, offers an easy-to-use collection of information to directly support experimental work (including microarray hybridisation), and provides deeper knowledge on the maritime pine transcriptome.

    • Several new initiatives have been launched recently to sequence conifer genomes including pines, spruces and Douglas-fir. Owing to the very large genome sizes ranging from 18 to 35 gigabases, sequencing even a single conifer genome had been considered unattainable until the recent throughput increases and cost reductions afforded by next generation sequencers. The purpose of this review is to describe the context for these new initiatives. A knowledge foundation has been acquired in several conifers of commercial and ecological interest through large-scale cDNA analyses, construction of genetic maps and gene mapping studies aiming to link phenotype and genotype. Exploratory sequencing in pines and spruces have pointed out some of the unique properties of these giga-genomes and suggested strategies that may be needed to extract value from their sequencing. The hope is that recent and pending developments in sequencing technology will contribute to rapidly filling the knowledge vacuum surrounding their structure, contents and evolution. Researchers are also making plans to use comparative analyses that will help to turn the data into a valuable resource for enhancing and protecting the world's conifer forests.

    • Intraocular lens (IOL) implantation following cataract extraction is the most common ophthalmic surgical procedure across the world. The IOL designs are constantly evolving and the postoperative visual outcomes are also improving significantly [1-3]. However, these reported outcomes are measured mainly for the central vision. Similar to other forms of vision correction such as spectacle lenses and contact lenses, IOLs are also designed to provide optimal image quality in the central visual field. Both theoretical and experimental evidence suggest that the peripheral optical errors are larger in eyes implanted with spherical and aspherical monofocal IOLs than phakic eyes [4-7]. Tabernero et al., reported an increase in the peripheral optical errors by analysing eyes that were implanted with monofocal IOLs from a variety of manufacturing companies and different optical designs, both spherical and aspherical [4]. The gradient index of the crystalline lens in the phakic eyes is suggested to partly compensate for the peripheral corneal astigmatism and the replacement of the crystalline lens with an IOL of constant index therefore results in higher The impact of peripheral optical errors induced by intraocular lenses was evaluated by simulating the average phakic and pseudophakic image qualities. An adaptive optics system was used to simulate the optical errors in 20 degrees nasal and inferior visual field in phakic subjects. Peripheral resolution acuity, contrast sensitivity and hazard detection were evaluated. Pseudophakic errors typical for monofocal designs had a negative effect on resolution acuity and contrast sensitivity and the hazard detection task also showed increased false positive and misses and a longer reaction time compared to phakic optical errors. The induced peripheral pseudophakic optical errors affect the peripheral visual performance and thereby impact functional vision.
