  • Efficient production of native, biologically active human cystatin C by Escherichia coli

    • A cDNA encoding the mature human cysteine proteinase inhibitor cystatin C was fused to the coding sequence for the Escherichia coli outer membrane protein A signal peptide, and the recombinant gene was expressed in E. coli under the control of the λ PR promoter, an optimized Shine-Dalgarno sequence and the λ cI 857 repressor. When induced at 42°C, such cells expressed large amounts of recombinant cystatin C. The recombinant protein was isolated in high yield and characterized. All physicochemical properties investigated, including the positions of disulfide bonds, indicated that the E. coli derived cystatin C was identical to cystatin C isolated from human biological fluids, except that the proline residue in position three was not hydroxylated. The recombinant protein displayed full biological activity against papain, cathepsin B and dipeptidyl peptidase I.

  • Systematic exploration of scenario spaces

    • Purpose - Scenarios have become a vital methodological approach in business as well as in public policy. When scenarios are used to guide analysis and decision-making, the aim is typically robustness and in this context we argue that two main problems at scenario set level is conservatism, i.e. all scenarios are close to a perceived business-as-usual trajectory and lack of balance in the sense of arbitrarily mixing some conservative and some extreme scenarios. The purpose of this paper is to address these shortcomings by proposing a methodology for generating sets of scenarios which are in a mathematical sense maximally diverse. Design/methodology/approach - In this paper, we develop a systematic methodology, Scenario Diversity Analysis (SDA), which addresses the problems of broad span vs conservatism and imbalance. From a given set of variables with associated states, SDA generates scenario sets where the scenarios are in a quantifiable sense maximally different and therefore best span the whole set of feasible scenarios. Findings - The usefulness of the methodology is exemplified by applying it to sets of storylines of the emissions scenarios of the Intergovernmental Panel on Climate Change. This ex-post analysis shows that the storylines were not maximally diverse and given the challenges ahead with regard to emissions reduction and adaptation planning, we argue that it is important to strive for diversity when developing scenario sets for climate change research. Originality/value - The proposed methodology adds significant novel features to the field of systematic scenario generation, especially with regard to scenario diversity. The methodology also enables the combination of systematics with the distinct future logics of good intuitive logics scenarios.

    • Expression of the human cysteine proteinase inhibitor, cystatin C (CysC) in the cytoplasm of Escherichia coli was studied using a cDNA fragment encoding the cysteine proteinase inhibitor controlled by the phage λ pImage /cI857 system. The yield of CysC was low, probably due to proteolytic degradation. By fusing the cysC cDNA to a DNA fragment encoding the signal peptide of the E. coli outer membrane protein A, it was possible to produce a substantial amount of CysC in the periplasm. The processing of the signal peptide was shown to be quantitative and to result in CysC with the correct N-terminal amino acid. Yields higher than 1000 μg CysC/ml can be obtained by initiating the product formation at a moderate temperature (40 °C) late in an optimized fermentation process. A method that gives selective extraction of the periplasmic proteins and at the same time stabilizes CysC has been used.

  • Design and Development of a Hexacopter for the Search and Rescue of a Lost Drone

    • Search and rescue with an autonomous robot is an attractive and challenging task within the research community. This paper presents the development of an autonomous hexacopter that is designed for retrieving a lost object, like a drone, from a vast-open space, like a desert area. Navigating its path with a proposed coverage path planning strategy, the hexacopter can efficiently search for a lost target and locate it using an image-based object detection algorithm. Moreover, after the target is located, our hexacopter can grasp it with a customised gripper and transport it back to a destined location. It is also capable of avoiding static obstacles and dynamic objects. The proposed system was realised in simulations before implementing it in a real hardware setup, i.e. assembly of the drone, crafting of the gripper, software implementation and testing under real-world scenarios. The designed hexacopter won the best UAV design award at the CPS-VO 2018 Competition held in Arizona, USA.

    • ABSTRACT: INTRODUCTION: Cartilage oligomeric matrix protein (COMP) is a homopentameric protein in cartilage. The development of arthritis, like collagen-induced arthritis (CIA), involves cartilage as a target tissue. We have investigated the development of CIA in COMP-deficient mice. METHODS: COMP-deficient mice in the 129/Sv background were backcrossed for 10 generations against B10.Q mice, which are susceptible to chronic CIA. COMP-deficient and wild-type mice were tested for onset, incidence, and severity of arthritis in both the collagen and collagen antibody-induced arthritis models. Serum anti-collagen II and anti-COMP antibodies as well as serum COMP levels in arthritic and wild-type mice were measured by enzyme-linked immunosorbent assay. RESULTS: COMP-deficient mice showed a significant early onset and increase in the severity of CIA in the chronic phase, whereas collagen II-antibody titers were similar in COMP-deficient and wild-type controls. COMP antibodies were not found in wild-type mice. Finally, COMP-deficient and wild-type mice responded similarly to collagen antibody-induced arthritis, indicating no difference in how collagen II antibodies interact with COMP-deficient cartilage during the initial stages of arthritis. CONCLUSIONS: COMP deficiency enhances the early onset and development of chronic arthritis but does not affect collagen II autoimmunity. These findings accentuate the importance of COMP in cartilage stability.
