    • Doubtlessly, using teachable moments is an efficient approach to promote student learning, and few occasions lend themselves better to this than when teachers and students meet to talk about students’ writing. However, these conversations may be difficult to establish and even if the intention of the feedback given to the students is to be formative and useful, there is a risk that it is perceived primarily as summative rather than something that expands the students’ understanding of writing and writing processes. Therefore, we would like to provide examples of a variety of tutoring and feedback designs for academic writing that are used to promote students’ further development as writers. The examples come from several different courses and writing interventions across the disciplines at a university of technology with both group and individual tutorials, peer and teacher feedback, self-assessment and reflection, as well as different setups for feedback on individual and group assignments. The background for this presentation is shaped by many years of negotiating and designing writing instruction, feedback and assessment practices within STEM education (Science, Technology, Engineering, Mathematics). A variety of setups for writing assignments and writing processes have been tested, and have resulted in practices that approach skills and understanding of academic and disciplinary writing in different ways. In this presentation we will discuss how tutoring and feedback practices have been adapted to different teaching situations depending on scope and variation in genres and writing assignments, as well as on logistics and numbers of students. The aim for this presentation is not to provide a solution or a state-of-the-art description of how to design writing tutorials and feedback, instead we hope that the examples and ideas behind our different setups will generate a useful discussion on tutoring, writing pedagogy and didactics.

    • This article describes different feedback designs that have been developed at Chalmers University of Technology, Sweden. These feedback activities are part of courses and programmes that faculty at the Department of Communication and Learning in Science, Division for Language and Communication, are involved in. The feedback setup has evolved from many years of designing and delivering writing instruction within STEM education, grounded in the challenge to make feedback a meaningful learning experience for all students and improve students’ understanding of disciplinary academic writing. The feedback designs described are based on dialogue to provide feedback and as a means for students to verbalize their own understanding of text, textual features and how discipline specific content is communicated. Examples of setups are large class active feedback lectures, scaffolded peer response sessions, and guided feedback workshops. These feedback activities are explored, and we argue for how they, potentially, result in more (useful) feedback and feedforward compared to traditional written teacher-student feedback.

  • DNA Electrophoresis in Polymer Solutions. Separation Mechanisms studied by Fluorescence and Linear Dichroism Spectroscopy

    • Capillary electrophoresis of DNA in polymer solutions is a promising technique for separation of DNA molecules according to size. The primary aim of the work presented in this thesis was to get a better understanding of the electrophoretic separation mechanism in these media by studying the DNA migration behavior during electrophoresis in both entangled and non-entangled polymer solutions using linear dichroism spectroscopy and fluorescence video microscopy. Since the fluorescence microscopy technique requires optical probing, the DNA was stained with the cyanine dye YOYO, a dimer of oxazole yellow (YO) with excellent fluorescent properties. The first part of the work deals with photophysical properties of YO and YOYO, their binding to DNA and the influence of probing with YOYO on the electrophoretic behavior of DNA. We found that YO and YOYO intercalates and bisintercalates, respectively, in DNA. Furthermore, binding of YOYO to DNA has quite a small effect on the electrophoretic behavior of DNA; it slows down the orientation dynamics and lowers the mobility but does not seem to affect the global migration mode.     • The photophysical properties of the optical DNA probe YOYO (homodimeric derivative of oxazole yellow) have been characterized in terms of the monomeric part, the YO chromophore. In aqueous solutions YO is virtually nonfluorescent but upon binding to DNA its fluorescence quantum yield is strongly increased. A similar enhancement of the fluorescence is observed for YO in the viscous solvent glycerol. The high fluorescence quantum yield of YO, when bound to DNA or in a viscous solution, is proposed to be a result of decreased rotational mobility around the internuclear bridge between the two aromatic ring systems. This hypothesis is based on similar values of the activation energies for the temperature-dependent nonradiative decay processes (E(A) = 53 kJ/mol) and viscous flow (E(A) = 63 kJ/mol), suggesting related rate-limiting mechanisms. A single electronic transition is found to be responsible for the intense visible absorption band. This conclusion is based on the observation of an essentially wavelength-independent reduced linear dichroism and similarly wavelength independent fluorescence anisotropy, and the fact that the emission spectrum is very nearly a mirror image of the absorption spectrum. The conclusion is further supported by quantum mechanical calculations (CNDO/S). By combination of measurements of fluorescence anisotropy of YO in glycerol and linear dichroism of YO in a stretched poly(vinyl alcohol) film, the transition moment of the strong visible absorption band was found to be nearly long axis polarized, in agreement with the CNDO/S calculations. The low-energy electronic transition and its polarization direction in the YO chromophore remain essentially unperturbed in the YOYO dye, suggesting that the results obtained for the excited state of the YO chromophore are applicable also to YOYO. One difference, though, is that in aqueous solutions the two YO chromophores of YOYO interact with each other, forming an internal dimer, resulting in a distorted absorption spectrum.
