  • Loss of dopamine uptake sites labeled with [3H]GBR-12935 in Alzheimer's disease.

    • The binding of the dopamine uptake inhibitor [3H]GBR-12935 to postmortem putamen from a control group and patients with Alzheimer's disease/senile dementia of Alzheimer type (AD/SDAT) or vascular dementia (VD) was studied. The binding density (Bmax) in AD/SDAT was significantly reduced to 50% of control. A reduction of Bmax in VD was also noted, but it did not reach statistical significance. No differences in apparent binding affinity (Kd) between controls and dementia groups were obtained. The concentrations of dopamine (DA), dihydroxyphenylacetic acid (DOPAC), 3-methoxytyramine (3-MT) and homovanillic acid were also determined. The concentrations of DA and DOPAC were reduced by 30-40% in AD/SDAT and VD, but the reductions did not reach statistical significance. The concentration of 3-MT was reduced by 40% in AD/SDAT and by 30% in VD. The [3H]GBR-12935-binding densities correlated significantly with corresponding concentrations of DA in control brains. It is suggested that the loss of [3H]GBR-12935-binding sites in human putamen in AD/SDAT reflects a degeneration of dopamine neurites.

  • TEM-tomography of FAU-zeolite crystals containing Pt-clusters

    • A method for preparing ultrathin sections (- 20 nm) of inorganic solids has been developed using ultramicrotomy of resin-embedded crystal fragments. Undamaged crystals, oriented along a crystallographic direction, could be imaged with transmission electron microscopy (TEM) at a resolution better than 0.5 nm. The true internal structure of the crystals could be investigated by imaging the second in a series of at least three consecutive ultrathin sections. Such TEM-tomography proved that Pt-ion exchanged FAU zeolite crystals, after reduction and oxidation, are occupied internally and randomly of large platinum clusters mainly in the {111-twin planes. TEM-tomography could be useful in man made nanostructures like semiconductors, epitaxial thin films, hard metal coatings, ceramics, catalysts, and biomaterials.

    • Nitrite may be generated by bacteria in urine during urinary tract infections. Acidification of nitrite results in the formation of nitric oxide (NO) and other reactive nitrogen oxides, which are toxic to a variety of microorganisms. We have studied NO formation and bacterial growth in mildly acidified human urine containing nitrite and the reducing agent vitamin C. Urine collected from healthy subjects was incubated in closed syringes at different pH values with varying amounts of nitrite and/or ascorbic acid added. NO generation was measured in headspace gas using a chemiluminescence technique. A similar setup was also used to study the growth of three strains of bacteria in urine. Mildly acidified nitrite-containing urine generated large amounts of NO and this production was greatly potentiated by ascorbic acid. The growth of Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus saprophyticus was markedly reduced by the addition of nitrite to acidified urine. This inhibition was enhanced by ascorbic acid. In conclusion, we show that the growth of three common urinary pathogens is markedly inhibited in mildly acidified urine when nitrite is present. The bacteriostatic effect of acidified nitrite is likely related to the release of NO and other toxic reactive nitrogen intermediates. These results may help to explain the well-known beneficial effects of urinary acidification with, e.g., vitamin C in treatment and prevention of urinary tract infection.

  • Evidence for an adrenergic innervation of the adrenal cortical blood vessels in rats.

    • The aim of this study was to investigate the blood flow in the adrenal cortex of the rat. Relative changes in the adrenal cortical blood flow were continuously measured by Laser Doppler flowmetry in 33 chloralose-anaesthetized artificially ventilated rats during electrical stimulation (1 ms, 5 V) of the left great splanchnic nerve (LGSN), which conveys both pre- and post-ganglionic nerve fibres to the adrenal gland. Laser Doppler flux (LDF) was decreased and regional resistance (RR) was increased by augmenting nerve stimulation at increasingly higher frequencies (2, 4, 8, 20 and 40 Hz). The decrease in LDF, when compared to pre-drug stimulations at 4 Hz was partially or totally inhibited by the adrenergic blocking agents trimethaphan (TRIM), guanethidine (GUA) and alpha 1-blockade with prazosin (PRAZ). Furthermore, both the decrease in LDF and the increase in RR were either completely or partially blocked by stimulation at 40 Hz after TRIM-treatment and GUA-treatment. It is concluded that the adrenal cortex in the rat is innervated by post-ganglionic adrenergic nerve fibres, which are involved in the regulation of blood flow in the adrenal cortex.

  • Municipal Solid Waste Management from a Systems Perspective

    • Different waste treatment options for municipal solid waste have been studied in a systems analysis. Different combinations of incineration, materials recycling of separated plastic and cardboard containers, and biological treatment (anaerobic digestion and composting) of biodegradable waste, were studied and compared to landfilling. The evaluation covered use of energy resources, environmental impact and financial and environmental costs. In the study, a calculation model ( ) based on methodology from life cycle assessment (LCA) was used. Case studies were performed in three Swedish municipalities: Uppsala, Stockholm, and Älvdalen. The study shows that reduced landfilling in favour of increased recycling of energy and materials lead to lower environmental impact, lower consumption of energy resources, and lower economic costs. Landfilling of energy-rich waste should be avoided as far as possible, partly because of the negative environmental impacts from landfilling, but mainly because of the low recovery of resources when landfilling. Differences between materials recycling, nutrient recycling and incineration are small but in general recycling of plastic is somewhat better than incineration and biological treatment somewhat worse. When planning waste management, it is important to know that the choice of waste treatment method affects processes outside the waste management system, such as generation of district heating, electricity, vehicle fuel, plastic, cardboard, and fertiliser.
