    • Antibody localization was analyzed following intraperitoneal (i.p.) or intravenous (i.v.) injection of the 125I-labelled anti-CEA-MAb I-38S1 in 44 nude rats, in order to evaluate the influence of tumour implantation site and the route of MAb administration. The animals were xenografted with a human colonic cancer (LS 174 T), either in the form of hepatic metastases, subcutaneous (s.c.) tumours or both. Tissue measurements, 4 days after MAb injection, showed better uptake for hepatic than for s.c. tumours, irrespective of the route of antibody administration. Antibody accumulation per g liver metastases was not size dependent for noduli weighing between 4 and 1,110 mg. MAb excretion evaluated in 20 animals and blood activity studied in 11 rats were equivalent 24-96 h following i.p. and i.v. injection. Dissimilar autoradiographic patterns were seen in hepatic metastases with predominantly peripherally located clusters following i.p. and more homogeneously distributed grains after i.v. MAb administration. The results indicate that tumour implantation site has a quantitative, and the route of administration at least a qualitative impact on the tumour accretion of anti-CEA MAb I-38S1 in the present xenograft model.

  • Influence of solar radiation on the availability of dissolved organic matter to bacteria in the Southern Ocean

    • The influence of solar radiation on the ability of dissolved organic matter (DOM) to support bacterial growth, was studied in the eastern Atlantic sector of the Southern Ocean during the SWEDARP 1997/1998 cruise with SA Agulhas December 1997–February 1998. Vertical profiles of water samples (2–3000 m) were obtained from the Spring Ice Edge (SIE, 60°S, high chlorophyll-a) and from the Winter Ice Edge (WIE, 56°S, low chlorophyll-a) areas. Filter-sterilized water from each area and depth was incubated under natural solar radiation and in the dark for comparison. Photobleaching of humic substance fluorescence occurred in all studied water samples. The bleaching was typically larger in the initially more fluorescent deep waters, compared to the low-fluorescent surface waters. Both the irradiated water and the dark controls were re-inoculated with a mixed bacterial inoculum from the initially sampled water. Bacterial growth (accumulating cells) and bacterial production (protein synthesis) were monitored during a 16–19 day incubation of these cultures at near in situ temperature (2 °C). Bacterial growth in cultures prepared from SIE water was largest at the surface (2–25 m), while the growth in corresponding cultures from the WIE did not vary much over depth. In contrast to the observed photobleaching, no clear effects of the irradiation on the ability of the DOM to support bacterial growth could be observed in either of the experiments. Hence, the degradation of fluorescent structures and other photochemical alterations of the organic matter did not have a major effect on the total pool of biodegradable organic substrates. The lack of effects of photoreactions on bacterial growth potential in the present study disagrees with the short-term bacterial growth response observed in other oceanic environments. This could be due to the different experimental approaches employed (short-vs. long-term incubations) or may indicate that the impact of photoreactions on bacterial growth in the Southern Ocean differ substantially from aquatic systems that are more influenced by terrestrial environments, as well as the warmer oligotrophic oceanic environments.

    • Bioassay experiments were performed two times (with 2 years in between) in order to investigate if nitrogen (N, ammonium), phosphorus (P, phosphate) and carbon (C, glucose) additions would stimulate the growth of bacteria and phytoplankton differently in three different tropical aquatic environments. The water and their indigenous microbial communities were taken from a freshwater coastal lake (Cabiunas), a coastal (Anjos), and an offshore marine station (Sonar) in the Atlantic outside Cabo Frio, Rio de Janeiro State, Brazil. Ammonium, phosphate and glucose were added alone or in combination to triplicate bottles. In the lake, P seemed to be the primary limiting factor during the first experiment, since both bacterial production and phytoplankton growth was stimulated by the P addition. Two years later, however, addition of P inhibited phytoplankton growth. During both years, C was closely co-limiting for bacteria since CP additions increased the response considerably. For both the coastal and offshore seawater stations, phytoplankton growth was clearly stimulated by N addition in both years and the bacteria responded either to the P, N or C additions (alone or in combination). To conclude, the results from these tropical aquatic systems show that it is possible that phytoplankton and bacteria may compete for a common resource (P) in lakes, but can be limited by different inorganic nutrients in marine waters as well as lakes, suggesting that phytoplankton and bacteria do not necessarily compete for the same growth limiting nutrient in these environments. Det låter högtravande och starkt. Men å andra sidan, ibland hör vi inte alls vad de säger in en serie på TV. Vad går förlorat när röstens uttryck förminskas även på scenen?

På operascenen skyms den talade textens innehåll ofta av det vokala uttrycket.Vad kommer vi att fåta del av under din presentation?Jag kommer redogöra för min undersökning av hur skådespelaren kan närma sig musikaliteten och ett expanderat vokalt uttryck före att gestalta textens innehåll. Och hur operasångaren kan söka sig mot en mer betydelsediger gestaltning av texten med bibehållen vokal kraft.

    • Background: Clinical trials with short and intermediate-term follow-up have demonstrated superior results for total hip replacement as compared with internal fixation with regard to hip function and the need for secondary surgery in elderly patients with a displaced intracapsular femoral neck fracture. The aim of the present study was to compare the results of total hip replacement with those of internal fixation over a long-term follow-up period of seventeen years.Methods: We enrolled 100 patients who had sustained a femoral neck fracture in a single-center, randomized controlled trial; all patients had had a healthy hip before the injury. The study group included seventy-nine women and twenty-one men with a mean age of seventy-eight years (range, sixty-five to ninety years). The subjects were randomly assigned to either total hip replacement (the arthroplasty group) (n = 43) or internal fixation (the control group) (n = 57). The primary end point was hip function, evaluated with use of the Harris hip score. Secondary end points included mortality, reoperations, gait speed, and activities of daily life. Follow-up evaluations were performed at threemonths and at one, two, four, eleven, and seventeen years.Results: The Harris hip score was higher in the total hip arthroplasty group, with a mean difference of 14.7 points (95% confidence interval, 9.2 to 20.1 points; p < 0.001 [analysis of covariance]) during the study period. We found no difference in mortality between the two groups. Four patients (9%) in the total hip replacement group and twenty-two patients (39%) in the internal fixation group had undergone a major reoperation (relative risk, 0.24; 95% confidence interval, 0.09 to 0.64). The overall reoperation rate was 23% (ten of forty-three) in the total hip replacement group and 53% (thirty of fifty-seven) in the internal fixation group (relative risk, 0.44; 95% confidence interval, 0.24 to 0.80). The results related to gait speed and activities of daily living favored the arthroplasty group during the first year.Conclusions: Over a period of seventeen years in a group of healthy, elderly patients with a displaced femoral neck fracture, total hip replacement provided better hip function and significantly fewer reoperations compared with internal fixation without increasing mortality.

    • Abstract:My dissertation is taken from an opera singer’s perspective and the aim is three-fold: ·         to investigate and problematize how gender and performance norms are reproduced on the opera stage·         to provide tools and propose approaches to transform gender perspective and norm critique into a creative artistic practice, enabling conscious and active artistic choices in relation to gender and performance norms·         to explore ways to expand the singer's artistic agency. The artistic research project draws on knowledge from both my own practice as a professional opera singer and my collaborators’ experiences from the worlds of opera, classical music and theatre.     The thesis consists of four artistic investigations:1. In Mönster och möjligheter (Patterns and Possibilities), gender markers were explored and identified, both in vocal and dramatic performance. The five Mozart scenes that were studied revealed a structure suggesting that femininity in the operatic art often can be expressed by taking responsibility for the music, for facial features and bodily behaviour, and for the audience. Correspondingly, masculinity seemed to be expressed by actively taking less responsibility in these areas.2. The second investigation was connected to Norrbottensmusiken's production of the new opera Under en kvinnas hjärta (Beneath the Heart of a Woman). This study exposed how performance norms affect singers' artistic possibilities also in contemporary opera. The investigation found that if the director's reading of an opera is norm-creative, it can facilitate a less traditionally gender-coded performance. However, even in more traditional opera productions, there are still options for the singer to work norm-creatively with their role, for example by mixing feminine and masculine gender markers.3. The third investigation, the concert production Sångernas sång (The Song of Songs), explored an expanded artistic agency for the singer. Actively using my experience from my career as an opera singer, I wrote the libretto for the chamber mini-opera Stark som döden (Strong as Death) in collaboration with a composer. The study suggested that singers, in addition to their creative work on the role, could also contribute with other important perspectives. Concert and opera productions, as well as newly written works, can be enriched by a dialogue with the singers early on in the artistic process. In the creation of the two roles in the chamber pieces Hög visa (Songs of Solomona) and Stark som döden, useful methods for creating a subject position for the characters were developed, as well as ways to work in a concert situation with the portrayal of sexual desire.4. The fourth and final investigation, Interaktion och intimitet (Interaction and Intimacy), was an exploration of intimate scenes. The study showed that the method and know-how of an intimacy coordinator can provide a sense of security that can make the singers freer in their portrayals of sexuality. A greater focus on the story can also make the scenes more interesting artistically. In addition, this study revealed the potential of an expanded agency for the singers. Allowing singers to adapt the text and the distribution of lines in repertoire opera, as well as creating a more collective rehearsal process, where the singers take an active part in the stage directing work, could open up for more norm-creative representations of gender on stage.

  • Voicelanding - Exploring the scenographic potential of acoustic sound in site-sensitive performance

    • This practical artistic research project (Documented Artistic Research Project (Doctoral Thesis)) explores how the performance of acoustic sound in dialogue with site can create a sonic scenography, experienced by an audience from within the sonic structures. Six art projects were carried out in the context of this research. Their form varies due to the site-sensitive approach that is employed: the space and the participating musicians are both the source and the frame for the resulting spatial sound performances.During workshops the collaborating musicians are introduced to site-sensitive methods. They learn full-body listening, spatial sounding, and space-care. The musicians learn to co-create with the space. In a collaborative process, spatial sound compositions are created using the site-specific sonic material that is elicited from the dialogue between the performers and the space. The relation to the audience plays an important role in the sharing of the performance space and the experience of the sonic scenographies. Therefore, active audience encounter is considered practiced during the creative process towards the performance and it is further explored during each performance.As sound is invisible and ephemeral it is a vulnerable material to engage with when creating scenographies. In this research its instability has revealed itself as an indispensable quality of a scenography that aims to connect the elements of a shared space and to make their relations perceivable.There is a tendency to make ‘reliable’ material scenographies and to sustain spatial sound through audio systems while attempting to overcome the challenges a site brings to performance. This approach to performance, scenography, and spatial sound composition, however, limits the relation between acoustic sound and site. In my sonic scenographies the performers are dependent on the dialogue with the space in order to create sonic structures that can be experienced by an audience. The attention needed for this collaboration is space-care. It includes care for all entities in the space, and especially the audience. The ephemeral quality of acoustic sound creates an active sonic scenography that performs together with the musicians, and engages multimodal listening.The resulting spatial sound performance includes the placement and movement of sonic expressions that are specific for each instrument-site relation. In the created performance, as the audience can ‘roam through’ it, they can experience a sonic scenography that unfolds around them. In the interaction of performers and audience in these shared spaces (architectural space and sonic space) a social space can develop that allows for an ephemeral community to emerge.

  • The role of maternal age and episiotomy in the risk of anal sphincter tears during childbirth

    • BACKGROUND: Many women who experience anal sphincter tear will suffer from anal incontinence. The most important tool to avoid this is to recognise the obstetric risk factors involved and thereby prevent injury. AIMS: The aim of this study was to analyse and evaluate the risk factors of anal sphincter tear during delivery. METHODS: Of a total of 57,943 vaginal deliveries, we identified 565 women with partial or total rupture of the anal sphincter and compared these women with 565 controls without sphincter tear with respect to possible risk factors. RESULTS: Several factors were significantly associated with sphincter tears, including nulliparity, birthweight, instrumental delivery, episiotomy, malpresentation, maternal age and epidural analgesia. The importance of these variables was further confirmed in a stepwise logistic regression analysis. Age was found to be a significant risk factor only when the birthweight was less than 4000 g. Episiotomy more than doubled the risk of sphincter tear when delivery was non-instrumental. CONCLUSION: There are several independent risk factors that should be considered when making decisions regarding delivery mode. Maternal age and episiotomy in non-instrumental delivery are two of these.
