    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • The complexity of computing along the cloud-to-edge continuum presents significant challenges to ICT operations and in particular reliable capacity planning and resource provisioning to meet unpredictable, fluctuating, and mobile demand. This chapter presents a high-level conceptual overview of RECAP—an architectural innovation to support reliable capacity provisioning for distributed clouds—and its operational modes and functional building blocks. In addition, the major design concepts informing its design—namely separation of concerns, model-centricism, modular design, and machine learning and artificial intelligence for IT operations—are also discussed.

    • Large-scale software systems are currently designed as distributed entities and deployed in cloud data centers. To overcome the limitations inherent to this type of deployment, applications are increasingly being supplemented with components instantiated closer to the edges of networks—a paradigm known as edge computing. The problem of how to efficiently orchestrate combined edge-cloud applications is, however, incompletely understood, and a wide range of techniques for resource and application management are currently in use.This article investigates the problem of reliable resource provisioning in joint edge-cloud environments, and surveys technologies, mechanisms, and methods that can be used to improve the reliability of distributed applications in diverse and heterogeneous network environments. Due to the complexity of the problem, special emphasis is placed on solutions to the characterization, management, and control of complex distributed applications using machine learning approaches. The survey is structured around a decomposition of the reliable resource provisioning problem into three categories of techniques: workload characterization and prediction, component placement and system consolidation, and application elasticity and remediation. Survey results are presented along with a problem-oriented discussion of the state-of-the-art. A summary of identified challenges and an outline of future research directions are presented to conclude the article.

    • Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection fatality rate (IFR) doubles with every 5 y of age from childhood onward. Circulating autoantibodies neutralizing IFN-α, IFN-ω, and/or IFN-β are found in ∼20% of deceased patients across age groups, and in ∼1% of individuals aged <70 y and in >4% of those >70 y old in the general population. With a sample of 1,261 unvaccinated deceased patients and 34,159 individuals of the general population sampled before the pandemic, we estimated both IFR and relative risk of death (RRD) across age groups for individuals carrying autoantibodies neutralizing type I IFNs, relative to noncarriers. The RRD associated with any combination of autoantibodies was higher in subjects under 70 y old. For autoantibodies neutralizing IFN-α2 or IFN-ω, the RRDs were 17.0 (95% CI: 11.7 to 24.7) and 5.8 (4.5 to 7.4) for individuals <70 y and ≥70 y old, respectively, whereas, for autoantibodies neutralizing both molecules, the RRDs were 188.3 (44.8 to 774.4) and 7.2 (5.0 to 10.3), respectively. In contrast, IFRs increased with age, ranging from 0.17% (0.12 to 0.31) for individuals <40 y old to 26.7% (20.3 to 35.2) for those ≥80 y old for autoantibodies neutralizing IFN-α2 or IFN-ω, and from 0.84% (0.31 to 8.28) to 40.5% (27.82 to 61.20) for autoantibodies neutralizing both. Autoantibodies against type I IFNs increase IFRs, and are associated with high RRDs, especially when neutralizing both IFN-α2 and IFN-ω. Remarkably, IFRs increase with age, whereas RRDs decrease with age. Autoimmunity to type I IFNs is a strong and common predictor of COVID-19 death.

    • Future cooperative autonomous vehicles will be able to organize into flexible platoons to improve both the efficiency and the safety of driving. However, platooning requires dependable coordination through the periodic wireless exchange of control messages. Therefore, challenging propagation scenarios as found, e.g., in dense urban areas, may hinder coordination and lead to undesirable vehicle behavior. While reconfigurable intelligent surfaces (RISs) have been advocated as a solution to improper coverage issues, no system-level simulation exists that accounts for realistic road mobility and communication aspects. To this end, we present one such simulator built on top of the OMNeT++-based PLEXE and Veins frameworks. Specifically, our contribution is a simulator that takes into account vehicle mobility, physical layer propagation, RIS coding, and networking protocols. To test our simulator, we implement an RIS-assisted autonomous platoon merging maneuver taking place at an intersection where the absence of any RIS would limit successful communications to an area dangerously close to the intersection itself. Our results validate the simulator as a feasible tool for system-level RIS-assisted cooperative autonomous vehicle maneuvering, and ultimately show the benefit of RIS as roadside infrastructure for wireless coverage extension. © 2023 IFIP.

    • Future cooperative autonomous vehicles will require high-performance communication means to support functions such as cooperative maneuvering and cooperative perception. The high-bandwidth requirements of these functions can be met through mmWave communications, whose utilization is often hindered by the harsh propagation conditions of typical vehicular environments. A solution to this problem is the use of reconfigurable intelligent surfaces (RISs), which enable the reflection of signals in a configurable direction, and have recently gained attention in the vehicular domain. In this paper, we provide an initial feasibility study, highlighting the challenges ahead and the performance RISs need to deliver in order to enable this type of communications. Specifically, we utilize CoopeRIS, a simulation framework for RISs integrated into the Plexe/Veins/SUMO ecosystem that we develop as further contribution and will release to the public. © 2023 IEEE.

  • A Review of Millimeter Wave Device-based Localization and Device-free Sensing Technologies and Applications

    • The commercial availability of low-cost millimeterwave (mmWave) communication and radar devices is starting to improve the adoption of such technologies in consumer markets, paving the way for large-scale and dense deployments in fifthgeneration (5G)-and-beyond as well as 6G networks. At the same time, pervasive mmWave access will enable device localization and device-free sensing with unprecedented accuracy, especially with respect to sub-6 GHz commercial-grade devices. This paper surveys the state of the art in device-based localization and device-free sensing using mmWave communication and radar devices, with a focus on indoor deployments. We overview key concepts about mmWave signal propagation and system design, detailing approaches, algorithms and applications for mmWave localization and sensing. Several dimensions are considered, including the main objectives, techniques, and performance of each work, whether they reached an implementation stage, and which hardware platforms or software tools were used. We analyze theoretical (including signal processing and machine learning), technological, and implementation (hardware and prototyping) aspects, exposing under-performing or missing techniques and items towards enabling a highly effective sensing of human parameters, such as position, movement, activity and vital signs. Among many interesting findings, we observe that device-based localization systems would greatly benefit from commercial-grade hardware that exposes channel state information, as well as from a better integration between standardcompliant mmWave initial access and localization algorithms, especially with multiple access points (APs). Moreover, more advanced algorithms requiring zero-initial knowledge of the environment would greatly help improve the adoption of mmWave simultaneous localization and mapping (SLAM). Machine learning (ML)-based algorithms are gaining momentum, but still require the collection of extensive training datasets, and do not yet generalize to any indoor environment, limiting their applicability. Device-free (i.e., radar-based) sensing systems still have to be improved in terms of: improved accuracy in the detection of vital signs (respiration and heart rate) and enhanced robustness/generalization capabilities across different environments; moreover, improved support is needed for the tracking of multiple users, and for the automatic creation of radar networks to enable largescale sensing applications. Finally, integrated systems performing joint communications and sensing are still in their infancy: theoretical and practical advancements are required to add sensing functionalities to mmWave-based channel access protocols based on orthogonal frequency-division multiplexing (OFDM) and multi-antenna technologies.

    • Optimisation of (the configuration and deployment of) distributed cloud applications is a complex problem that requires understanding factors such as infrastructure and application topologies, workload arrival and propagation patterns, and the predictability and variations of user behaviour. This chapter outlines the RECAP approach to application optimisation and presents its framework for joint modelling of applications, workloads, and the propagation of workloads in applications and networks. The interaction of the models and algorithms developed is described and presented along with the tools that build on them. Contributions in modelling, characterisation, and autoscaling of applications, as well as prediction and generation of workloads, are presented and discussed in the context of optimisation of distributed cloud applications operating in complex heterogeneous resource environments.

    • The REliable CApacity Provisioning and enhanced remediation for distributed cloud applications (RECAP) project aims to advance cloud and edge computing technology, to develop mechanisms for reliable capacity provisioning, and to make application placement, infrastructure management, and capacity provisioning autonomous, predictable and optimized. This paper presents the RECAP vision for an integrated edge-cloud architecture, discusses the scientific foundation of the project, and outlines plans for toolsets for continuous data collection, application performance modeling, application and component auto-scaling and remediation, and deployment optimization. The paper also presents four use cases from complementing fields that will be used to showcase the advancements of RECAP.
