  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Precision Orbit of delta Delphini and Prospects for Astrometric Detection of Exoplanets

    • Combining visual and spectroscopic orbits of binary stars leads to a determination of the full 3D orbit, individual masses, and distance to the system. We present a full analysis of the evolved binary system delta Delphini using astrometric data from the MIRC and PAVO instruments on the CHARA long-baseline interferometer, 97 new spectra from the Fairborn Observatory, and 87 unpublished spectra from the Lick Observatory. We determine the full set of orbital elements for delta Del, along with masses of 1.78 +/- 0.07 M-circle dot and 1.62 +/- 0.07 M-circle dot for each component, and a distance of 63.61 +/- 0.89 pc. These results are important in two contexts: for testing stellar evolution models and for defining the detection capabilities for future planet searches. We find that the evolutionary state of this system is puzzling, as our measured flux ratios, radii, and masses imply a similar to 200 Myr age difference between the components, using standard stellar evolution models. Possible explanations for this age discrepancy include mass transfer scenarios with a now-ejected tertiary companion. For individual measurements taken over a span of two years, we achieve <10 mu as precision on the differential position with 10 minute observations. The high precision of our astrometric orbit suggests that exoplanet detection capabilities are within reach of MIRC at CHARA. We compute exoplanet detection limits around delta Del and conclude that, if this precision is extended to wider systems, we should be able to detect most exoplanets >2M(J) on orbits >0.75 au around individual components of hot binary stars via differential astrometry.

    • Ordered nanoporous structures are among the most fascinating and industrially important materials currently in use. The archetypal zeolite material has now been joined by an eclectic array of new structures that exhibit porosity over a wide range of length scales and with order/disorder expressed in a multitude of ways. This raises the bar in terms of characterization and extends a real challenge to the scientific community to fully understand the properties and potential future applications of such materials. In this review we discuss the importance of modern microscopy tools combined with diffraction in this endeavour and show how the details of even the most complex quasi-crystalline nanoporous architectures can be elucidated. We show by using the appropriate spherical aberration (C-s) corrections in scanning transmission electron microscopy it is possible to decipher all the individual silicon and aluminum atoms in a zeolite structure. Automated routines for using large electron diffraction datasets for crystal structure determination of nanocrystals is described making the need for large single crystal synthesis less-and-less important. The power of complementary combinations of surface tools such as atomic force microscopy and high-resolution scanning electron microscopy is discussed to elucidate crystal growth mechanisms. For mesoporous materials synthesized from self-organized organic mesophases electron microscopy reveals the details of the complex hierarchy of porosity so crucial for the functional performance of the structure.

  • Does income redistribution prevent residential segregation?

    • Living in low-income neighborhoods can have adverse effects. Public policies that reduce income inequality might prevent residential segregation by income. However, previously documented associations between income inequality and residential segregation may not reflect residential sorting effects. We use rich full-population data for Sweden 1990-2017 and take advantage of how in-moving residents change the municipal income composition to rule out the influence of reverse causation and mechanical effects. We find that changing taxes and transfers has limited residential sorting effects on segregation. However, our results strongly suggest that raising the education levels of low-income residents is effective for fighting segregation.

    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

  • Constraining the formation and transport of lunar impact glasses using the ages and chemical compositions of Chang’e-5 glass beads

    • Impact glasses found in lunar soils provide a possible window into the impact history of the inner solar system. However, their use for precise reconstruction of this history is limited by an incomplete understanding of the physical mechanisms responsible for their origin and distribution and possible relationships to local and regional geology. Here, we report U-Pb isotopic dates and chemical compositions of impact glasses from the Chang’e-5 soil and quantitative models of impact melt formation and ejection that account for the compositions of these glasses. The predominantly local provenance indicated by their compositions, which constrains transport distances to <~150 kilometers, and the age-frequency distribution are consistent with formation mainly in impact craters 1 to 5 kilometers in diameter. Based on geological mapping and impact cratering theory, we tentatively identify specific craters on the basaltic unit sampled by Chang’e-5 that may have produced these glasses and compare their ages with the impact record of the asteroid belt.
