  • Hydrogen sulfide upregulates K-ATP channel expression in vascular smooth muscle cells of spontaneously hypertensive rats

    • The study was designed to investigate whether H2S could upregulate expression of K-ATP channels in vascular smooth muscle cells (VSMCs), and by this mechanism enhances vasorelaxation in spontaneously hypertensive rats (SHR). Blood pressure, vascular structure, and vasorelaxation were analyzed. Plasma H2S was detected using polarographic sensor. SUR2B and Kir6.1 expressions were detected in VSMCs of SHR and in A7r5 cells as well as primarily cultured ASMCs using real-time PCR, western blot, immunofluorescence, and confocal imaging. Nuclear translocation of forkhead transcription factors FOXO1 and FOXO3a in ASMCs was detected using laser confocal microscopy, and their binding activity with SUR2B and Kir6.1 promoters was examined by chromatin immunoprecipitation. SHR developed hypertension at 18 weeks. They showed downregulated vascular SUR2B and Kir6.1 expressions in association with a decreased plasma H2S level. H2S donor, however, could upregulate vascular SUR2B and Kir6.1 expressions, causing a left shift of the vasorelaxation curve to pinacidil and lowered tail artery pressure in the SHR. Also, H2S antagonized endothelin-1 (ET-1)-inhibited K-ATP expression in A7r5 cells and cultured ASMCs. Mechanistically, H2S inhibited ET-1-stimulated p-FOXO1 and p-FOXO3a expressions (inactivated forms), but increased their nuclear translocation and the ET-1-inhibited binding of FOXO1 and FOXO3a with Kir6.1 and SUR2B promoters in ASMCs. Hence, H2S promotes vasorelaxation of SHR, at least in part, through upregulating the expression of K-ATP subunits by inhibiting phosphorylation of FOXO1 and FOXO3a, and stimulating FOXO1 and FOXO3a nuclear translocation and their binding activity with SUR2B and Kir6.1 promoters. H2S increased vascular SUR2B and Kir6.1 expression of SHR, promoting vasorelaxation. H2S antagonized ET-1-inhibited K-ATP expression in A7r5 cells and cultured ASMCs. H2S inhibited ET-1-induced FOXO1 and FOXO3a phosphorylation in ASMCs. H2S promoted FOXO1 and FOXO3a nuclear translocation and binding with target gene promoters.

    • The fast growth of Internet traffic, the growingimportance of cellular accesses and the escalating competitionbetween content providers and network operators result in agrowing interest in improving network performance and userexperience. In terms of network transport, different solutionsranging from tuning TCP to installing middleboxes are applied.It turns out, however, that the practical results sometimes aredisappointing and we believe that poor testing is one of thereasons for this. Indeed, many cases in the literature limittesting to the simple and rare use case of a single file download,while common and complex use cases like web browsing oftenare ignored or modelled only by considering smaller files. Tofacilitate better testing, we present a set of metrics by whichthe complexity around web pages can be characterised andthe potential for different optimisations can be estimated. Wealso derive numerical values of these metrics for a small set ofpopular web pages and study similarities and differences betweenpages with the same kind of content (newspapers, e-commerceand video) and between pages designed for the same platform(computer and smartphone).

  • Analysis of Data From Breast Diseases Treated With 5-Alpha Reductase Inhibitors for Benign Prostatic Hyperplasia

    • OBJECTIVE: 5-alpha reductase inhibitors (5ARIs) decrease the androgen levels in vivo and are currently used for the treatment of benign prostatic hyperplasia (BPH) in men. However, these inhibitors can also increase the risk of gynecomastia, breast tenderness, and breast cancer. Hence, we did a systematic review and meta-analysis to evaluate the rate of breast-related diseases in men treated with 5ARIs.MATERIALS AND METHODS: PubMed, Embase, Cochrane, and CNKI databases were searched for randomized controlled trials using 5ARIs in patients with BPH. Data were analyzed by using Cochrane Collaboration review manager program and Stata 12.0 software.RESULTS: In total, 14 studies were included in the meta-analysis. Gynecomastia was significantly more common with 5ARIs treatment when compared with placebo (3.30% vs. 1.84%; P < .00001) or alpha blockers (ABs) monotherapy (2.33% vs. 1.00%; P = .0009). Both dutasteride (2.03% vs. 0.90%; P < .00001) and finasteride (4.08% vs. 2.43%; P < .00001) are associated with significantly higher risk of gynecomastia than placebo. Risk for breast tenderness was elevated in 5ARIs users (0.83% vs. 0.25%; P = .01) or in users having combination therapy with ABs (2.48% vs. 0.58%; P < .0001). Finasteride is associated with significantly higher risk of breast tenderness than placebo (0.80% vs. 0.25%; P = .02).CONCLUSION: In male patients with BPH, 5ARIs have significantly increased the risk of gynecomastia and breast tenderness but may be not to the breast cancer. In addition, combination therapy is significantly associated with higher risk of breast tenderness compared to single ABs monotherapy. © 2019 Elsevier Inc. All rights reserved.

  • Comparative Analysis on Abnormal Methylome of Differentially Expressed Genes and Disease Pathways in the Immune Cells of RA and SLE

    • We identified abnormally methylated, differentially expressed genes (DEGs) and pathogenic mechanisms in different immune cells of RA and SLE by comprehensive bioinformatics analysis. Six microarray data sets of each immune cell (CD19+ B cells, CD4+ T cells and CD14+ monocytes) were integrated to screen DEGs and differentially methylated genes by using R package "limma." Gene ontology annotations and KEGG analysis of aberrant methylome of DEGs were done using DAVID online database. Protein-protein interaction (PPI) network was generated to detect the hub genes and their methylation levels were compared using DiseaseMeth 2.0 database. Aberrantly methylated DEGs in CD19+ B cells (173 and 180), CD4+ T cells (184 and 417) and CD14+ monocytes (193 and 392) of RA and SLE patients were identified. We detected 30 hub genes in different immune cells of RA and SLE and confirmed their expression using FACS sorted immune cells by qPCR. Among them, 12 genes (BPTF, PHC2, JUN, KRAS, PTEN, FGFR2, ALB, SERB-1, SKP2, TUBA1A, IMP3, and SMAD4) of RA and 12 genes (OAS1, RSAD2, OASL, IFIT3, OAS2, IFIH1, CENPE, TOP2A, PBK, KIF11, IFIT1, and ISG15) of SLE are proposed as potential biomarker genes based on receiver operating curve analysis. Our study suggests that MAPK signaling pathway could potentially differentiate the mechanisms affecting T- and B- cells in RA, whereas PI3K pathway may be used for exploring common disease pathways between RA and SLE. Compared to individual data analyses, more dependable and precise filtering of results can be achieved by integrating several relevant data sets.

  • Human interactive secure key and ID exchange protocols in body sensor networks

    • A body sensor network (BSN) is typically a wearable wireless sensor network. Security protection is critical to BSNs, since they collect sensitive personal information. Generally speaking, security protection of BSN relies on identity (ID) and key distribution protocols. Most existing protocols are designed to run in general wireless sensor networks, and are not suitable for BSNs. After carefully examining the characteristics of BSNs, the authors propose human interactive empirical channel-based security protocols, which include an elliptic curve Diffie–Hellman version of symmetric hash commitment before knowledge protocol and an elliptic curve Diffie–Hellman version of hash commitment before knowledge protocol. Using these protocols, dynamically distributing keys and IDs become possible. As opposite to present solutions, these protocols do not need any pre-deployment of keys or secrets. Therefore compromised and expired keys or IDs can be easily changed. These protocols exploit human users as temporary trusted third parties. The authors, thus, show that the human interactive channels can help them to design secure BSNs.

    • A total of 55 samples including soil, sediment, plants (cypress, reed and seepweed) and aquatic species were collected at locations around hexabromocyclododecane (HBCD) manufacturing facilities in Laizhou Bay area, East China. HBCD was determined at concentrations ranging between 0.88 and 6901 ng g(-1) dry weight (dw), 2.93-1029 ng g(-1) dw, 8.88-160241 ng g(-1) dw, and 7.09-815 ng g(-1) lipid weight (lw), respectively. Significant negative correlations (r(2) = 0.54, p = 0.006) were observed between HBCD concentrations in soils and the distance from the manufacturing facility, and the concentrations became constant when the distance was >4 km. The calculation results on the bioaccumulation factors (BAFs) suggested that HBCD may be accumulated in plants. Tissue-specific bioaccumulation of HBCD diastereoisomers was found in aquatic species. For example, in crabs the highest concentrations of HBCD (815 ng g(-1) lw for female and 446 ng g(-1) lw for male) were observed in the gill. Besides the gill, α-HBCD was more preferentially accumulated in the spermary and ovary, while β- and γ-HBCD were more accumulated in the muscle. A similar distribution was also observed in roe and muscle of goby fish.

  • Exploring askers' switching from free to paid social Q&A services : A perspective on the push-pull-mooring framework

    • The purpose of this study is to explore the factors that prompt askers to switch from free to paid social question-and-answer (SQA) services. Prior studies have investigated users' motivations and participation in free and paid SQA services; however, little attention has been paid to askers' switching behavior. We empirically analyzed the content of qualitative interviews from 64 askers on a well-known SQA platform in China. Based on the push-pull-mooring framework, we identified and classified factors that influenced askers' to switch from free to paid Q&A services, using the critical incident technique, after which we calculated the entropy weights of the 16 subcategories before and after the switch, using the entropy weight method. The findings suggest that askers' switching behavior was influenced by push factors (i.e., dissatisfaction with the free SQA service), pull factors (i.e., satisfaction with the paid SQA service), and mooring factors (i.e., social factors, personal factors, situational factors). Moreover, the findings show that the effects of these factors vary significantly before and after a switch. Dissatisfaction with the quality of information from the free SQA service would influence users before a switch, whereas satisfaction with the quality of information from the paid SQA service would influence them after a switch. In terms of mooring factors, the effects of social and personal factors on askers' switching behavior, especially subjective norms and cognitive lock-in, turn out to be less significant after a switch, whereas the effect of trust is more significant. Besides, the effects of situational factors are more or less the same before and after a switch. To the best of our knowledge, this paper is one of the first attempts to explore factors that affect askers' switching behavior and to shed light on the managerial strategies of paid SQA services.
