  • Down-Regulation of Cilia-Localized IL-6R{alpha} by 17{beta}-Estradiol in Mouse and Human Fallopian Tubes.

    • The action of Interleukin-6 (IL-6) impacts female reproduction. Although IL-6 was recently shown to inhibit cilia activity in human fallopian tubes in vitro, the molecular mechanisms underlying IL-6 signaling to tubal function remain elusive. Here, we investigate the cellular localization, regulation, and possible function of two IL-6 receptors (IL-6Ralpha and gp130) in mouse and human fallopian tubes in vivo. We show that IL-6Ralpha is restricted to the cilia of epithelial cells in both mouse and human fallopian tubes. Exogenous 17beta-estradiol (E2), but not progesterone (P4), causes a time-dependent decrease in IL-6Ralpha expression which is blocked by the estrogen receptor (ER) antagonist ICI 182,780. Exposure of different ER-selective agonists, PTT or DPN, demonstrated an ER subtype-specific regulation of IL-6Ralphaalpha in mouse fallopian tubes. In contrast to IL-6Ralpha, gp130 was detected in tubal epithelial cells in mice but not in humans. In humans, gp130 was found in the muscle cells and was decreased in the periovulatory and luteal phases during the reproductive cycles, indicating a species-specific expression and regulation of gp130 in the fallopian tube. Expression of tubal IL-6Ralpha and gp130 in IL-6 knockout mice was found to be normal; however, E2 treatment increased IL-6Ralpha, but not gp130, in IL-6 knockout mice compared to wild-type mice. Furthermore, expression levels of IL-6Ralpha, but not gp130, decreased in parallel with estrogenic accelerated oocyte-cumulus complex (OCC) transport in mouse fallopian tubes. Our findings unveil a potential role for cilia-specific IL-6Ralpha in the regulation of OCC transport and suggest an estrogen-regulatory pathway of IL-6Ralpha in the fallopian tube.     • In the area of governmental information for and services to citizens digitalization has certainly become a buzzword. Framed within the discourses on e-government or e-governance – or differ­ent mixtures of the two – various analyses have tried to point out, or even anticipate possible consequences of the appropriation of digital technologies, mainly the internet, in governmental services.On the one hand, these analyses have pointed out a great deal of opportunities connected to the incorporation of the internet into governmental information and services. Policy makers have been quick to point to the increased accessibility as a great opportunity for the citizens; in Sweden this vision of accessibility has even been referred to as ‘24:7-governmental agencies’. Academics, among others, have also suggested that the digitalization of information and services opens up new possibilities for citizen control of governments.On the other hand, a number of problems have also been identified. For instance, the digitalization of public registers holding personal information has been interpreted as a threat to the citizens’ integrity: Will digitalization bring a new surveillance society? The most frequently debated prob­lem, however, at least within research, has been the fear of digital divides. Will the internet create digital cleavages between different groups of citizens?So far, however, neither the hopeful nor the dystopian analyses have made enough efforts to critically evaluate their claims. Such evaluations can start from different points of departure, and in this paper the starting point is the citizens as users of governmental information and services through the internet: What patterns of inclusion and exclusion emerge as a governmental agency digitalizes its information and services by making them increasingly internet based?The paper presents statistical data from a survey of 762 unemployed citizens using the Swedish Public Employment Service, a governmental agency that has come to rely specifically heavy on internet based information and service. The initial analysis of data reveals interesting differences between social groups in terms of both perception and use of the internet – in general – and the resources offered by SPES in particular. For instance, the users’ various degrees of education is a strong, determining factor when it comes to use of internet SPES’ services.The paper starts from a conceptual elaboration of various notions of e-government and e-gov­ernance. Thereafter, the survey data is described and elaborated on before moving into a discus­sion of the wider significance of the findings: What does data suggest in terms of patterns of in­clusion in and exclusion from a society in which governmental agencies, to an increasing extent, use the internet for their information and services?

    • The aim of this paper is to describe an ongoing project with the aim of improving safety in an organization working with maintenance and development of the railway infrastructure in Sweden. The first sub goal was to investigate the Genta method, with 62 employees resulted in a description of these latent errors in the organization.eral Failure Types in the organization. Seminars and interviews, based on the Tripod Del Recommendations for an improvement toward a safety culture was suggested, action plans were formulated and, in some cases, implemented. A follow up study is planned in a two year perspective.

    • The purpose of this study was to investigate possible risk factors related to the quality of spoken communication between train drivers and train dispatchers at Banverket (Swedish Rail Administration). In a first stage 1422 recorded and transcribed dialogues were subject to an exploratory linguistic analysis. In a second stage 200 dialogues were sampled and made subject to a detailed analysis in terms of communication error types. Results showed an overall low frequency of communication errors. Errors actually observed were mainly related to deficiencies in identification and designation.

    • Phenytoin is a human and animal teratogen. Accumulating evidence suggests that the teratogenicity is associated with a potential of phenytoin to cause embryonic cardiac arrhythmia and resultant generation of toxic reactive oxygen species via hypoxia-reoxygenation mechanisms. The A/J mouse is more susceptible to phenytoin teratogenicity than other mouse strains. The aim of this study was to investigate whether A/J mice have other antioxidant enzyme activities than C57BL/6J and CD-1 mice. Also, strain differences in phenytoin effects on embryonic heart rate and rhythm were determined. Another objective was to determine whether a spin trapping agent with capacity to capture reactive oxygen species alter the developmental toxicity of phenytoin. Treatment with this agent resulted in a marked decrease in phenytoin teratogenicity, which supports the idea that reactive oxygen species are important mediators for the teratogenic action of phenytoin. The A/J mice embryos were most susceptible to the adverse cardiac effects of phenytoin and had the highest activity of superoxide dismutase and glutathione peroxidase, while the activity of catalase was the same in embryos of the three different strains. The high activities of antioxidant enzymes in the A/J stain indicate that the sensitivity to develop malformations is caused by excessive arrhythmia-related generation of reactive oxygen species rather than impaired antioxidant defense.
