    • A nonvanishing entropy production rate is one of the defining characteristics of any nonequilibrium system, and several techniques exist to determine this quantity directly from experimental data. The short-time inference scheme, derived from the thermodynamic uncertainty relation, is a recent addition to the list of these techniques. Here we apply this scheme to quantify the entropy production rate in a class of microscopic heat engine models called Brownian gyrators. In particular, we consider models with anharmonic confining potentials. In these cases, the dynamical equations are indelibly nonlinear, and the exact dependencies of the entropy production rate on the model parameters are unknown. Our results demonstrate that the short-time inference scheme can efficiently determine these dependencies from a moderate amount of trajectory data. Furthermore, the results show that the nonequilibrium properties of the gyrator model with anharmonic confining potentials are considerably different from its harmonic counterpart; especially in setups leading to a nonequilibrium dynamics and the resulting gyration patterns.

    • Designing multifunctional linkers is crucial for tricomponent theranostic targeted nanomedicine development as they are essential to enrich polymeric systems with different functional moieties. Herein, we have obtained a hetero-trifunctional linker from malonic acid and demonstrated its implication as an amphiphilic targeted nanotheranostic system (CB DX UN PG FL). We synthesized it with varying hydrophilic segment to fine-tune the hydrophobic/hydrophilic ratio to optimize its self-assembly. pH-responsive hydrazone-linked doxorubicin was conjugated to the backbone (UN PG FL) containing folate as a targeting ligand. Cobalt carbonyl complex was used for T-2-weighted magnetic resonance imaging (MRI). Electron micrographs of optimized molecule CB DX UN PG((4 kDa)) FL in an aqueous system have demonstrated about 50-60 nm-sized uniform micelles. The relaxivity study and the one-dimensional (1D) imaging experiments dearly revealed the effect of the nanotheranostics system on transverse relaxation (T-2) of water molecules, which validated the system as a T-2-weighted MRI contrast agent. The detailed in vitro biological studies validated the targeted delivery and anticancer potential of CB DX UN PG((4 kDa)) FL. Combining the data on transverse relaxation, folate mediated uptake, and anticancer activity, the designed molecule will have a significant impact on the development of targeted theranostic.

  • Quantitative analysis of non-equilibrium systems from short-time experimental data

    • Estimating entropy production directly from experimental trajectories is of great current interest but often requires a large amount of data or knowledge of the underlying dynamics. In this paper, we propose a minimal strategy using the short-time Thermodynamic Uncertainty Relation (TUR) by means of which we can simultaneously and quantitatively infer the thermodynamic force field acting on the system and the (potentially exact) rate of entropy production from experimental short-time trajectory data. We benchmark this scheme first for an experimental study of a colloidal particle system where exact analytical results are known, prior to studying the case of a colloidal particle in a hydrodynamical flow field, where neither analytical nor numerical results are available. In the latter case, we build an effective model of the system based on our results. In both cases, we also demonstrate that our results match with those obtained from another recently introduced scheme. Thermal fluctuations play a crucial role in non-equilibrium phenomena at microscopic length scales, making it challenging to analyse and interpret experimental data. Here, the authors demonstrate that the short-time thermodynamic uncertainty relation inference scheme can estimate the entropy production rate for a colloidal particle in time-varying potentials and with background flows determined by the presence of a microbubble.

    • In patients with pancreatic cancer (PC), the peritoneal cavity is the second-most common site of metastasis after the liver. Peritoneal macrophages (PMs) have been demonstrated to play a significant role in the peritoneal metastases of different cancers. Gemcitabine (GEM) is known to affect PC-associated immune cells, including macrophages. However, its effect on PMs and its possible clinical implication is yet to be investigated. In this study, mouse-derived PMs were treated with GEM ex vivo to analyze the polarization status. Production of GEM-induced reactive oxygen species (ROS) and reactive nitrogen species was evaluated using DCFH-DA, DAF-FM, and Griess assay. Antitumor effects of PMs on UN-KC-6141and UN-KPC-961 murine PC cells were evaluated in presence and absence of GEM in vitro. Similarly, effect of GEM on human THP-1 macrophage polarization and its tumoricidal effect was studied in vitro. Furthermore, the effect of GEM-treated PMs on peritoneal metastasis of UN-KC-6141 cells was evaluated in a syngeneic mouse model of PC. GEM upregulated M1 phenotype-associated molecular markers (Tnf-α and Inos) in vitro in PMs obtained from naïve mouse. Moreover, IL-4-induced M2-like PMs reverted to M1-like after GEM treatment. Co-culture of UN-KC-6141 and UN-KPC-961 cancer cells with PMs in the presence of GEM increased apoptosis of these cells, whereas cell death was markedly reduced after N-acetyl-l-cysteine treatment. Corroborating these findings co-culture of GEM-treated human THP-1 macrophages also induced cell death in MIAPaCa-2 cancer cells. GEM-treated PMs injected intraperitoneally along with UN-KC-6141 cells into mice extended survival period, but did not stop disease progression and mortality. Together, GEM induced M1-like polarization of PMs from naive and/or M2-polarized PMs in a ROS-dependent manner. GEM-induced M1-like PMs prompted cytotoxicity in PC cells and delayed disease progression in vivo.

    • Reduced transition probabilities have been extracted between excited, yrast states in the N=Z+2 nucleus 94Pd. The transitions of interest were observed following decays of the Iπ=14+, Ex=2129-keV isomeric state, which was populated following the projectile fragmentation of a 124Xe primary beam at the GSI Helmholtzzentrum für Schwerionenforschung accelerator facility as part of FAIR Phase-0. Experimental information regarding the reduced E2 transition strengths for the decays of the yrast 8+ and 6+ states was determined following isomer-delayed Eγ1−Eγ2−△T2,1 coincidence method, using the LaBr3(Ce)-based FATIMA fast-timing coincidence gamma-ray array, which allowed direct determination of lifetimes of states in 94Pd using the Generalized Centroid Difference (GCD) method. The experimental value for the half-life of the yrast 8+ state of 755(106) ps results in a reduced transition probability of B(E2:8→+6+) = 205−25+34 e2 fm4, which enables a precise verification of shell-model calculations for this unique system, lying directly between the N=Z line and the N=50 neutron shell closure. The determined B(E2) value provides an insight into the purity of (g9/2)n configurations in competition with admixtures from excitations between the (lower) N=3pf and (higher) N=4gds orbitals for the first time. The results indicate weak collectivity expected for near-zero quadrupole deformation and an increasing importance of the T=0 proton-neutron interaction at N=48.
