    • The combined detection of a gravitational-wave signal, kilonova, and short gamma-ray burst (sGRB) from GW170817 marked a scientific breakthrough in the field of multimessenger astronomy. But even before GW170817, there have been a number of sGRBs with possible associated kilonova detections. In this work, we re-examine these ` historical' sGRB afterglows with a combination of state-of-the-art afterglow and kilonova models. This allows us to include optical/near-infrared synchrotron emission produced by the sGRB as well as ultraviolet/optical/near-infrared emission powered by the radioactive decay of r-process elements (i.e. the kilonova). Fitting the light curves, we derive the velocity and the mass distribution as well as the composition of the ejected material. The posteriors on kilonova parameters obtained from the fit were turned into distributions for the peak magnitude of the kilonova emission in different bands and the time at which this peak occurs. From the sGRB with an associated kilonova, we found that the peak magnitude in H bands falls in the range [-16.2, -13.1] (95 per cent of confidence) and occurs within 0.8-3.6 d after the sGRB prompt emission. In g band instead we obtain a peak magnitude in range [-16.8, -12.3] occurring within the first 18 h after the sGRB prompt. From the luminosity distributions of GW170817/AT2017gfo, kilonova candidates GRB130603B, GRB050709, and GRB060614 (with the possible inclusion of GRB150101B, GRB050724A, GRB061201, GRB080905A, GRB150424A, and GRB160821B) and the upper limits from all the other sGRBs not associated with any kilonova detection we obtain for the first time a kilonova luminosity distribution in different bands.

    • Smallholder-dominated agricultural mosaic landscapes are highlighted as model production systems that deliver both economic and ecological goods in tropical agricultural landscapes, but trade-offs underlying current land-use dynamics are poorly known. Here, using the most comprehensive quantification of land-use change and associated bundles of ecosystem functions, services and economic benefits to date, we show that Indonesian smallholders predominantly choose farm portfolios with high economic productivity but low ecological value. The more profitable oil palm and rubber monocultures replace forests and agroforests critical for maintaining above- and below-ground ecological functions and the diversity of most taxa. Between the monocultures, the higher economic performance of oil palm over rubber comes with the reliance on fertilizer inputs and with increased nutrient leaching losses. Strategies to achieve an ecological-economic balance and a sustainable management of tropical smallholder landscapes must be prioritized to avoid further environmental degradation.

    • Balanced induction of proinflammatory and type I IFN responses upon activation of Toll-like receptors (TLRs) determines the outcome of microbial infections and the pathogenesis of autoimmune and other inflammatory diseases. Mast cells, key components of the innate immune system, are known for their debilitating role in allergy and autoimmunity. However, their role in antimicrobial host defenses is being acknowledged increasingly. How mast cells interact with microbes and the nature of responses triggered thereby is not well characterized. Here we show that in response to TLR activation by Gram-positive and -negative bacteria or their components, mast cells elicit proinflammatory but not type I IFN responses. We demonstrate that in mast cells, bound bacteria and TLR ligands remain trapped at the cell surface and do not undergo internalization, a prerequisite for type I IFN induction. Such cells, however, can elicit type I IFNs in response to vesicular stomatitis virus which accesses the cytosolic retinoic acid-inducible gene I receptor. Although important for antiviral immunity, a strong I IFN response is known to contribute to pathogenesis of several bacterial pathogens such as Listeria monocytogenes. Interestingly, we observed that the mast cell-dependent neutrophil mobilization upon L. monocytogenes infection is highly impaired by IFN-beta. Thus, the fact that mast cells, although endowed with the capacity to elicit type I IFNs in response to viral infection, elicit only proinflammatory responses upon bacterial infection shows that mast cells, key effector cells of the innate immune system, are well adjusted for optimal antibacterial and antiviral responses.

    • The results indicate that TLR2 activation induces pro-inflammatory and type I interferon responses from distinct subcellular sites: the plasma membrane and endolysosomal compartments respectively. Apart from identifying and characterizing a novel pathway for induction of type I interferons, the present study offers new insights into how TLR signaling discriminates and regulates the nature of responses to be elicited against extracellular and endocytosed microbes. These findings may also have clinical implication. Excessive production of pro-inflammatory cytokines and type I IFNs following activation of TLRs is a central pathologic event in several hyper-inflammatory conditions. The discovery that the induction of pro-inflammatory and type I IFN responses can be uncoupled through pharmacological manipulation of endolysosomal acidification suggests new avenues for potential therapeutic intervention against inflammations and sepsis.

  • The landscape of tolerated genetic variation in humans and primates

    • Personalized genome sequencing has revealed millions of genetic differences between individuals, but our understanding of their clinical relevance remains largely incomplete. To systematically decipher the effects of human genetic variants, we obtained whole-genome sequencing data for 809 individuals from 233 primate species and identified 4.3 million common protein-altering variants with orthologs in humans. We show that these variants can be inferred to have nondeleterious effects in humans based on their presence at high allele frequencies in other primate populations. We use this resource to classify 6% of all possible human protein-altering variants as likely benign and impute the pathogenicity of the remaining 94% of variants with deep learning, achieving state-of-the-art accuracy for diagnosing pathogenic variants in patients with genetic diseases.

    • The results illustrate that signals triggered by LLO contribute to TLR2 redundancy in recognition of L. monocytogenes. Under normal conditions, multiple and, sometimes, redundant pathways cooperate to induce a rapid antimicrobial defense. When one signaling pathway-in this case, TLR2-is removed from the system, the other pathways are still capable of mounting a sufficient response to ensure survival of the host.

    • The effect of the residual oxygen concentration in the process atmosphere during laser-powder bed fusion (L-PBF) of Ti-6Al-4V was investigated, using an external oxygen monitoring system equipped with two types of oxygen sensors typically used in L-PBF hardware: a lambda probe and an electrochemical oxygen sensor. The recordings of the oxygen variations during L-PBF highlighted that the electrochemical sensor is more reliable than the lambda probe, whose signal showed a maximum deviation of about 700 ppm O2 after 7 h, attributed to its sensitivity to hydrogen present in the system. The study revealed that proper monitoring of the oxygen in the laboratory scale L-PBF system used is necessary to limit oxygen and nitrogen pick-ups by the built material. Concentrations as high as 2200 ppm O2 and 500 ppm N2 in the Ti-6Al-4V part built under standard conditions were measured, compared to maximum levels of 1800 ppm O2 and 250 ppm N2 with the external oxygen control. In addition, the findings underline the critical effect of the component design, such as the high aspect ratio columns or the lattice structures, on the heat accumulation in case of Ti-6Al-4V, leading to enhanced oxygen and nitrogen pick-up, as high as 600 ppm O2 and 150 ppm N2 difference between the bottom and top of the cylindrical samples of 70 mm height used in this study. The determination of tensile properties of samples built at different heights put in evidence the detrimental effect of the oxygen increase with build height on the ductility, which decreased from 12% to below 6% between the bottom and top positions. This work highlights that the possible presence of impurities in the L-PBF atmosphere can have harmful impact on the properties of Ti-6Al-4V components, which can be mitigated adjusting the oxygen control system.
