  • Chemoprophylaxis, diagnosis, treatments, and discharge management of COVID-19 : An evidence-based clinical practice guideline (updated version)
  • 2020
  • Ingår i: Military Medical Research. - : Springer Science and Business Media LLC. - 2054-9369. ; 7:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

  • Guidelines for the use and interpretation of assays for monitoring autophagy
  • 2012
  • Ingår i: Autophagy. - : Informa UK Limited. - 1554-8635 .- 1554-8627. ; 8:4, s. 445-544
  • Forskningsöversikt (refereegranskat)abstract
    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Unraveling the unique role of brown graphitic carbon nitride in robust CO2 photoreduction
  • 2023
  • Ingår i: Applied Surface Science. - : ELSEVIER. - 0169-4332 .- 1873-5584. ; 615
  • Tidskriftsartikel (refereegranskat)abstract
    • Photocatalytic CO2 reduction is one of the important means to alleviate the energy crisis. In this work, an oxygen linked and brown graphitic carbon nitride (GACN) was successfully prepared by thermal polymerization after oil bath method. GACN introduced oxygen atoms on surface of BulkCN. Various characterizations of the material show that the prepared GACN has a different structure and higher photoelectronic activity compared to BulkCN. GACN possessed strong photocatalytic CO2 reduction capacity, and the photocatalytic activity was significantly improved compared with BulkCN. In view of density functional theory calculations, it is proved that the oxygen atoms introduced by GACN increase CO2 photoreaction reactivity, enhance electronic activity and reduce the reaction energy barrier. This work can have a positive effect on the photocatalytic application of g-C3N4 with the existence of oxygen atoms.

  • Endothelial cell heterogeneity and microglia regulons revealed by a pig cell landscape at single-cell level
  • 2022
  • Ingår i: Nature Communications. - : Springer Nature. - 2041-1723. ; 13:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Pigs are valuable large animal models for biomedical and genetic research, but insights into the tissue- and cell-type-specific transcriptome and heterogeneity remain limited. By leveraging single-cell RNA sequencing, we generate a multiple-organ single-cell transcriptomic map containing over 200,000 pig cells from 20 tissues/organs. We comprehensively characterize the heterogeneity of cells in tissues and identify 234 cell clusters, representing 58 major cell types. In-depth integrative analysis of endothelial cells reveals a high degree of heterogeneity. We identify several functionally distinct endothelial cell phenotypes, including an endothelial to mesenchymal transition subtype in adipose tissues. Intercellular communication analysis predicts tissue- and cell type-specific crosstalk between endothelial cells and other cell types through the VEGF, PDGF, TGF-beta, and BMP pathways. Regulon analysis of single-cell transcriptome of microglia in pig and 12 other species further identifies MEF2C as an evolutionally conserved regulon in the microglia. Our work describes the landscape of single-cell transcriptomes within diverse pig organs and identifies the heterogeneity of endothelial cells and evolutionally conserved regulon in microglia.

  • Stacking Engineering of Heterojunctions in Half-Metallic Carbon Nitride for Efficient CO2 Photoreduction
  • 2023
  • Ingår i: Advanced Science. - : WILEY. - 2198-3844.
  • Tidskriftsartikel (refereegranskat)abstract
    • Enhancing charge separation in semiconductor photocatalysts is a major challenge for efficient artificial photosynthesis. Herein, a compact heterojunction is designed by embedding half-metallic C(CN)(3) (hm-CN) hydrothermally in BiOBr (BOB) as the backbone. The interface between hm-CN and BOB is seamless and formed by covalent bonding to facilitate the transmission of photoinduced electrons from BOB to hm-CN. The transient photocurrents and electrochemical impedance spectra reveal that the modified composite catalyst exhibits a larger electron transfer rate. The photocatalytic activity of hm-CN/BOB increases significantly as indicated by a CO yield that is about four times higher than that of individual components. Density-functional theory calculations verify that the heterojunction improves electron transport and decreases the reaction energy barrier, thus promoting the overall photocatalytic CO2 conversion efficiency. The half-metal nitride coupled semiconductor heterojunctions might have large potential in artificial photosynthesis and related applications.

  • Control of fluorescence enhancement and directionality upon excitations in a thin-film system
  • 2015
  • Ingår i: Physica status solidi. B, Basic research. - : Wiley. - 0370-1972 .- 1521-3951. ; 252:10, s. 2222-2229
  • Tidskriftsartikel (refereegranskat)abstract
    • Nanostructures with various configurations have been extensively used to engineer the emission properties of embedded fluorophores, but lack the flexibility to dynamically control fluorescence. Here we report a thin-film cavity system, comprising a quarter wavelength thick dye-doped dielectric coating on a reflecting surface, in which the fluorescence enhancement and directionality can be significantly modified by altering the illumination angle. The configuration of the cavity yields absorption properties that are highly dependent on illumination angles, due to the coupling between molecular absorption and Fabry-Perot resonances. Therefore the fluorescence intensity relating to the angle-dependent absorbing efficiency varies with illumination angles. In addition, as a result of synergy between intrinsic absorption of the reflecting surface, Fabry-Perot and surface-plasmon-polariton resonances and illumination-angle dependent excitation efficiencies for differently located molecules, the global emission intensity, including emission from dyes at all locations, can be directionally redistributed by altering the illumination angle.

  • Probing Plasmonic Gap Resonances between Gold Nanorods and a Metallic Surface
  • 2015
  • Ingår i: The Journal of Physical Chemistry C. - : American Chemical Society (ACS). - 1932-7447 .- 1932-7455. ; 119:32, s. 18627-18634
  • Tidskriftsartikel (refereegranskat)abstract
    • The plasmonic resonances in individual gold nanorods nanoscopically coupled to a gold film with different gap spacing have been experimentally and theoretically investigated. The spectral widths, wavelengths, and optical polarizabilities of the maxima in measured single-nanopartide scattering spectra are significantly modified as the gap distance changes in the sub-20 nm domain. Comparing the experimental data with numerical simulations reveals that these modifications arise from the complex hybridization of several dipolar and multipolar plasmon modes that are strongly localized at the gap. These plasmon gap modes have distinct resonant and spatial characteristics as a result of near-field interaction between the elongated nanorods and the gold film. Additionally, the excitation of these gap modes is highly dependent on the gap spacing. Finally, we also discuss influences of these plasmonic modes on absorption properties of the system and propose a potential application of the studied structures in facilitating photothermal conversion.
