    • In this paper, we employ two peroxidases (horseradish peroxidase, HRP and cytochrome c peroxidase, CcP) to demonstrate their ability to retain their redox and biological functions after their immobilisation on mesoporous TiO2 and SnO2 electrodes. We will also demonstrate the use of HRP immobilised on the metal oxide electrodes for the development of reagentless optical and electrochemical biosensors for the detection of hydrogen peroxide (H2O2) with low detection limit of 0.04 and 1 mu M, respectively.     • Photoelectrocatalysis is one of the most favourable techniques that could be used in this remit as it has the potential to utilise natural sunlight to generate oxidants in situ to mediate effective pollutant degradation. This work, therefore, utilises a mesoporous nanoscale WO3/BiVO4 heterostructure photoanode to effectively degrade ibuprofen in wastewater combined with simultaneous green hydrogen generation at the cathode under simulated sunlight. A near complete degradation (>96%) of ibuprofen (starting concentration of 100 mg/L), with no hazardous intermediates (determined via mass spectrometry analysis), along with simultaneous H2 evolution of 114 µmol/cm2 after 145 min was demonstrated in this work. In addition, intermediate product analysis, the role of the type of in situ oxidants on degradation, the mechanistic pathway of degradation, and the material characteristics of mesoporous photoanode were also investigated. First experimental evidence of in situ generated H2O2 contributing to the degradation of ibuprofen is presented.

  • New methods to identify conserved microsatellite loci and develop primer sets of high cross-species utility - as demonstrated for birds

    • We have developed a new approach to create microsatellite primer sets that have high utility across a wide range of species. The success of this method was demonstrated using birds. We selected 35 avian EST microsatellite loci that had a high degree of sequence homology between the zebra finch Taeniopygia guttata and the chicken Gallus gallus and designed primer sets in which the primer bind sites were identical in both species. For 33 conserved primer sets, on average, 100% of loci amplified in each of 17 passerine species and 99% of loci in five non-passerine species. The genotyping of four individuals per species revealed that 24-76% (mean 48%) of loci were polymorphic in the passerines and 18-26% (mean 21%) in the non-passerines. When at least 17 individuals were genotyped per species for four Fringillidae finch species, 71-85% of loci were polymorphic, observed heterozygosity was above 0.50 for most loci and no locus deviated significantly from Hardy-Weinberg proportions. This new set of microsatellite markers is of higher cross-species utility than any set previously designed. The loci described are suitable for a range of applications that require polymorphic avian markers, including paternity and population studies. They will facilitate comparisons of bird genome organization, including genome mapping and studies of recombination, and allow comparisons of genetic variability between species whilst avoiding ascertainment bias. The costs and time to develop new loci can now be avoided for many applications in numerous species. Furthermore, our method can be readily used to develop microsatellite markers of high utility across other taxa.

    • The use of non-intrusive techniques for monitoring nuclear reactors is becoming more vital as western fleets age. As a consequence, the necessity to detect more frequently occurring operational anomalies is of upmost interest. Here, noise diagnostics — the analysis of small stationary deviations of local neutron flux around its time-averaged value — is employed aiming to unfold from detector readings the nature and location of driving perturbations. Given that in-core instrumentation of western-type light-water reactors are scarce in number of detectors, rendering formal inversion of the reactor transfer function impossible, we propose to utilise advancements in Machine Learning and Deep Learning for the task of unfolding. This work presents an approach to such a task doing so in the presence of multiple and simultaneously occurring perturbations or anomalies. A voxel-wise semantic segmentation network is proposed to determine the nature and source location of multiple and simultaneously occurring perturbations in the frequency domain. A diffusion-based core simulation tool has been employed to provide simulated training data for two reactors. Additionally, we work towards the application of the aforementioned approach to real measurements, introducing a self-supervised domain adaptation procedure to align the representation distributions of simulated and real plant measurements.
