    • Human tumor-derived cell lines are indispensable tools for basic and translational oncology. They have an infinite life span and are easy to handle and scalable, and results can be obtained with high reproducibility. However, a tumor-derived cell line may not be authentic to the tumor of origin. Two major questions emerge: Have the identity of the donor and the actual tumor origin of the cell line been accurately determined? To what extent does the cell line reflect the phenotype of the tumor type of origin? The importance of these questions is greatest in translational research. We have examined these questions using genetic profiling and transcriptome analysis in human glioma cell lines. We find that the DNA profile of the widely used glioma cell line U87MG is different from that of the original cells and that it is likely to be a bona fide human glioblastoma cell line of unknown origin.

    • Kungl. Gustav Adolfs Akademien för svensk folkkultur är en riksakademi med säte i Uppsala. Sin uppgift att främja forskning rörande svensk folklig kultur, begreppet taget i vid mening, skall Akademien enligt sina stadgar bland annat fullgöra genom att i sina skriftserier offentliggöra forskningsrön inom ämnesområden som den har till uppgift att vårda sig om. Huvudserien är Acta Academiae Regiae Gustavi Adolphi, vars första nummerutkom 1933. Bland övriga serier kan nämnas Folklivsskildringar och bygdestudier, Kungl. Gustav Adolfs Akademiens småskrifter och Svenska sagor och sägner.På senare år har intresset för ordspråksforskning eller - med en internationell term - paremiologi varit på uppgång i flera av de nordiska länderna. Ett tecken på detta är symposiet Aspekter på ordspråk som i april 2011 anordnades i Uppsala av Kungl. Gustav Adolfs Akademien för svensk folkkultur. Syftet var att skapa kontakter mellan nordiska ordspråksforskare inom skilda discipliner. Studiet av ordspråk är nämligen ett tvärvetenskapligt forskningsfält i skärningspunkten mellan folkloristik, språkvetenskap och pedagogik.I föreliggande volym ingår elva bidrag som presenterades vid symposiet. Både geografiskt och tidsmässigt uppvisar artiklarna stor spännvidd: från Island i väster till Ryssland i öster, från medeltida samlingar till dagens ordspråkstravestier. En del av författarna anlägger språkliga perspektiv, andra betraktar ordspråken utifrån en genreteoretisk eller kulturhistorisk synvinkel, och ytterligare några fokuserar på insamlingen av ordspråk eller enskilda betydelsefulla samlingar. Tillsammans ger artiklarna en bild av mångfalden av angreppspunkter i dagens ordspråksforskning.

  • Distinct germline genetic susceptibility profiles identified for common non-Hodgkin lymphoma subtypes

    • Lymphoma risk is elevated for relatives with common non-Hodgkin lymphoma (NHL) subtypes, suggesting shared genetic susceptibility across subtypes. To evaluate the extent of mutual heritability among NHL subtypes and discover novel loci shared among subtypes, we analyzed data from eight genome-wide association studies within the InterLymph Consortium, including 10,629 cases and 9505 controls. We utilized Association analysis based on SubSETs (ASSET) to discover loci for subsets of NHL subtypes and evaluated shared heritability across the genome using Genome-wide Complex Trait Analysis (GCTA) and polygenic risk scores. We discovered 17 genome-wide significant loci (P < 5 × 10−8) for subsets of NHL subtypes, including a novel locus at 10q23.33 (HHEX) (P = 3.27 × 10−9). Most subset associations were driven primarily by only one subtype. Genome-wide genetic correlations between pairs of subtypes varied broadly from 0.20 to 0.86, suggesting substantial heterogeneity in the extent of shared heritability among subtypes. Polygenic risk score analyses of established loci for different lymphoid malignancies identified strong associations with some NHL subtypes (P < 5 × 10−8), but weak or null associations with others. Although our analyses suggest partially shared heritability and biological pathways, they reveal substantial heterogeneity among NHL subtypes with each having its own distinct germline genetic architecture.

  • Microsatellite instability and mutations in BRAF and KRAS are significant predictors of disseminated disease in colon cancer

    • Background: Molecular alterations are well studied in colon cancer, however there is still need for an improved understanding of their prognostic impact. This study aims to characterize colon cancer with regard to KRAS, BRAF, and PIK3CA mutations, microsatellite instability (MSI), and average DNA copy number, in connection with tumour dissemination and recurrence in patients with colon cancer. Methods: Disease stage II-IV colon cancer patients (n = 121) were selected. KRAS, BRAF, and PIK3CA mutation status was assessed by pyrosequencing and MSI was determined by analysis of mononucleotide repeat markers. Genome-wide average DNA copy number and allelic imbalance was evaluated by SNP array analysis. Results: Patients with mutated KRAS were more likely to experience disease dissemination (OR 2.75; 95% CI 1.28-6.04), whereas the opposite was observed for patients with BRAF mutation (OR 0.34; 95% 0.14-0.81) or MSI (OR 0.24; 95% 0.09-0.64). Also in the subset of patients with stage II-III disease, both MSI (OR 0.29; 95% 0.10-0.86) and BRAF mutation (OR 0.32; 95% 0.16-0.91) were related to lower risk of distant recurrence. However, average DNA copy number and PIK3CA mutations were not associated with disease dissemination. Conclusions: The present study revealed that tumour dissemination is less likely to occur in colon cancer patients with MSI and BRAF mutation, whereas the presence of a KRAS mutation increases the likelihood of disseminated disease.

  • A meta-analysis of Hodgkin lymphoma reveals 19p13.3 TCF3 as a novel susceptibility locus

    • Recent genome-wide association studies (GWAS) of Hodgkin lymphoma (HL) have identified associations with genetic variation at both HLA and non-HLA loci; however, much of heritable HL susceptibility remains unexplained. Here we perform a meta-analysis of three HL GWAS totaling 1,816 cases and 7,877 controls followed by replication in an independent set of 1,281 cases and 3,218 controls to find novel risk loci. We identify a novel variant at 19p13.3 associated with HL (rs1860661; odds ratio (OR) = 0.81, 95% confidence interval (95% CI) = 0.76-0.86, P-combined 3.5 x 10(-10)), located in intron 2 of TCF3 (also known as E2A), a regulator of B-and T-cell lineage commitment known to be involved in HL pathogenesis. This meta-analysis also notes associations between previously published loci at 2p16, 5q31, 6p31, 8q24 and 10p14 and HL subtypes. We conclude that our data suggest a link between the 19p13.3 locus, including TCF3, and HL risk.

    • This book gives a critical account of various methods used in music analysis. In the first chapter, a number of current approaches such as semiotics, musical implications, Schenkerian analysis, and generative theory are demonstrated on Mozart’s K. 331 theme. Five essays deal with important concepts in music analysis: ambiguity, formal proportions, and similarity within and between works. A further chapter provides a discussion of probability, kinship, and influence – decisive criteria when judging musical plagiarism. The last essay, studying a piece by Schubert, sifts the prospects of deciphering a composer’s sexual leanings from his music.

  • Chopin : The Preludes and Beyond

    • The first study of this volume looks for reminiscences of Dies Irae in Chopin’s works. A great number of allusions and affinities are found in the preludes as well as in Chopin’s output. The study also yields insights into Chopin’s composition method. These intertextual findings are used in an attempt to establish the extra-musical content of the Second Ballade. Five preludes – A minor, E minor, B minor, A major and C minor – are closely examined, using diverse analytical approaches. A primary concern is to critically assess previous readings, and Schenkerian ones in particular. An analysis of the initial right-hand passage of the F-minor étude from Méthode brings up matters of idiomatic and ontology.

  • Musical conception of abstract film - The case of Viking Eggeling's Diagonal Symphony

    • The idea of the Gesamtkunstwerk and the belief in the kinship of the arts were passed on from late romanticism to many of the modernist movements in the first decades of the twentieth century. Thus artists and film-makers wished to accomplish a fusion that transferred the achievements of non-figurative art to the new film medium, swayed by the constraints of realist story-telling. But abstract film needed principles that could organize its visual flow, and some theorists thought that music, the most abstract of the arts, could be of avail. This paper will present an analysis in musical terms of Viking Eggeling's abstract film the Diagonal Symphony, and it will be shown both that he relied on the most widely used formal pattern of classical music, the sonata form, to mould his only film, and that he employed a number of other musical devices to give interest and coherence to its details. The account will start, however, with a short biographical sketch and with some pertinent general information on the film.
