The process of opting for female permanent contraception : A qualitative study of women's experiences in Sweden

- Objectives: We aimed to explore Swedish women's decision-making experiences regarding permanent contraception. Study design: In this study, we included 17 women aged 30–48 who were scheduled to undergo female permanent contraceptive procedures. We conducted semistructured interviews using two broad open-ended questions. We analyzed these data using systematic text condensation based on the principles of psychological phenomenological analysis. Results: The interviewees experienced no counseling or support from health care workers regarding permanent contraception until they specifically asked for it. Participants reported that they themselves place the responsibility of permanent contraception solely on women. Consequently, our participants described feeling hesitancy and ambivalence in the process of deciding to have the procedure. Once the decision was made and the women were on the waiting lists for surgery, they experienced relief and empowerment. Conclusions: Our findings suggest that health care providers in Sweden miss opportunities to support patient-centered decision-making regarding permanent contraception. This study indicates that women make deliberate and considered decisions regarding permanent contraception and are best positioned to know when the procedure should take place in their reproductive lives. Implication statements: Health care professionals should discuss permanent contraception as an option with all women desiring contraception to allow them to decide if that method is right for them. © 2020 Elsevier Inc.

Escherichia coli single-stranded DNA-binding a molecular tool for improved sequence protein quality in pyrosequencing

- Pyrosequencing is a four-enzyme bioluminometric DNA sequencing technique based on a DNA sequencing by synthesis principle. Currently, the technique is limited to analysis of short DNA sequences exemplified by single-nucleotide polymorphism analysis. In order to expand the field for pyrosequencing, the read length needs to be improved and efforts have been made to purify reaction components as well as add single-stranded DNA-binding protein (SSB) to the pyrosequencing reaction. In this study, we have performed a systematic effort to analyze the effects of SSB by comparing the pyrosequencing result of 103 independent complementary DNA (cDNA) clones. More detailed information about the cause of low quality sequences on templates with different characteristics was achieved by thorough analysis of the pyrograms. Also, real-time biosensor analysis was performed on individual cDNA clones for investigation of primer annealing and SSB binding on these templates. Results from these studies indicate that templates with high performance in pyrosequencing without SSB possess efficient primer annealing and low SSB affinity. Alternative strategies to improve the performance in pyrosequencing by increasing the primer-annealing efficiency have also been evaluated.

- Escherichia coli (E. coli) single-stranded binding protein (SSB) is a valuable protein for various biotechnical applications, such as PCR and DNA sequencing, Here we describe an efficient expression and purification scheme where the tendency of SSB to aggregate at low salt concentration and high protein concentration is avoided. The method contains fewer steps of purification and results in high protein yield, compared to previous published protocols. In our protocol, cells are harvested after cultivation overnight and SSB is isolated by ammonium sulfate precipitation followed by anion-exchange chromatography. The yield from a 2-liter fed-batch fermenter is 2 g protein, which is higher than all production methods for SSB earlier reported, Moreover, the two classical isolation steps combined in the purification scheme are robust, cost-efficient, and suitable for scaling up. The resulting SSB is pure and a correctly folded tetramer with an apparent binding to single-stranded DNA with a K-D of 10(-8) M, as determined by surface plasmon resonance.

- The overall theme for this dissertation is the position of higher Swedish civil servants in the borderland between politics and administration and between flexibility, loyalty, impartiality, and autonomy. An important assumption is that the attitudes and beliefs are especially significant in a system such as the Swedish, where civil servants may act under conditions of substantial autonomy. The dissertation is based on interviews with higher civil servants and a sample of members of the Swedish parliament. Among the civil servants the study is limited to the higher echelons in the national public administration - undersecretaries of state, assistant undersecretaries of state, and director-generals of administrative agencies. The study is comparative - comparisons are made between 1971/73 and 1990. The central instruments of analysis consist of three role conceptions of civil servants. They are constructed from three fundamental coordinating principles and are designated the judicial, the political and the market-oriented civil servant roles. In a second step, the theoretical role typology is adapted to "the practice" which is manifested in the Swedish public administration's development in a longer perspective.How the role of the civil servants in the political system was perceived, as well as the political positions, was found to be dependent on the positions of the interviewees in the politico-administrative system. This meant that the undersecretaries, irrespective of role perception, found themselves farthest to the left among the studied groups, then followed by the assistant undersecretaries and farthest right the director-generals. An exception to this rule was the politically-oriented director-generals, who were left of all the assistant undersecretaries irrespective of role perception and even to the left of the judicially-oriented undersecretaries. An important observation was that the market-oriented role of civil servants had strengthened its position over time. It could also be observed in 1990 that the undersecretaries were clearly different from their colleagues in 1971 through applying more of a political perspective of the civil servant role. The analyses showed that among the non-political civil servants - the assistant undersecretaries - the political role model had a strong position at both interview occasions. Furthermore, both politicians and civil servants showed a more negative attitude to politics and the administration. The higher civil servants had begun to waiver somewhat in their belief in themselves and the capacity of the state. The analyses of the role perception of the MPs showed that their opinions were closely connected with their political affiliation. Among social democrats and members from the Left Party (the former Communist party), the political role conception was dominating at both times and among non-socialists the judicial. An important observation when dealing with the views of the members of parliament was how surprisingly unfamiliar politicians were with the work of the public administation.

- I samband med Donald Schöns bortgång hölls ett minnesseminarium i Malmö i april 1998. De medverkande talade om Schöns arbete och vad det betytt för dem. I den här filmen har vi samlat delar av tre inlägg. Erik Stolterman karakteriserar Schöns sätt att arbeta i relation till mer vedertagna uppfattningar om vad god forskning är. Bertil Rolf kritiserar Schön för att ge upp anspråken på sanning. Jerker Lundequist pekar på att Schöns teori saknar estetiska begrepp. Filmen vänder sig till den som studerar Schöns arbete och vill ta del av andra perspektiv på det.

- Recent advances in single-photon-counting detectors are enabling the development of novel approaches to reach micrometer-scale resolution in x-ray imaging. One example of such a technology are the MEDIPIX3RX-based detectors, such as the LAMBDA which can be operated with a small pixel size in combination with real-time on-chip charge-sharing correction. This characteristic results in a close to ideal, box-like point spread function which we made use of in this study. The proposed method is based on raster-scanning the sample with sub-pixel sized steps in front of the detector. Subsequently, a deconvolution algorithm is employed to compensate for blurring introduced by the overlap of pixels with a well defined point spread function during the raster-scanning. The presented approach utilizes standard laboratory x-ray equipment while we report resolutions close to 10 mu m. The achieved resolution is shown to follow the relationship p/n with the pixel-size p of the detector and the number of raster-scanning steps n. (C) 2016 Optical Society of America
