  • On Reduction of Substrate Noise in Mixed-Signal Circuits

    • Microelectronics is heading towards larger and larger systems implemented on a single chip. In wireless communication equipment, e.g., cellular phones, handheld computers etc., both analog and digital circuits are required. If several integrated circuits (ICs) are used in a system, a large amount of the power is consumed by the communication between the ICs. Furthermore, the communication between ICs is slow compared with on-chip communication. Therefore, it is favorable to integrate the whole system on a single chip, which is the objective in the system-on-chip (SoC) approach.In a mixed-signal SoC, analog and digital circuits share the same chip. When digital circuits are switching, they produce noise that is spread through the silicon substrate to other circuits. This noise is known as substrate noise. The performance of sensitive analog circuits is degraded by the substrate noise in terms of, e.g., lower signal-to-noise ratio and lower spurious-free dynamic range. Another problem is the design of the clock distribution net, which is challenging in terms of obtaining low power consumption, sharp clock edges, and low simultaneous switching noise.In this thesis, a noise reduction strategy that focus on reducing the amount of noise produced in digital clock buffers, is presented. The strategy is to use a clock with long rise and fall times. It is also used to relax the constraints on the clock distribution net, which also reduce the design effort. Measurements on a test chip show that the strategy can be implemented in an IC with low cost in terms of speed and power consumption. Comparisons between substrate coupling in silicon-on-insulator (SOI) and conventional bulk technology are made using simple models. The objective here is to get an understanding of how the substrate coupling differs in SOI from the bulk technology. The results show that the SOI has less substrate coupling when no guard band is used, up to a certain frequency that is highly dependent of the chip structure. When a guard band is introduced in one of the analyzed test structures, the bulk resulted in much higher attenuation compared with SOI. An on-chip measurement circuit aiming at measuring simultaneous switching noise has also been designed in a 0.13 µ SOI process.

    • To significantly increase the sampling rate of an A/D converter (ADC), a time interleaved ADC system is a good option. The drawback of a time interleaved ADC system is that the ADCs are not exactly identical due to errors in the manufacturing process. This means that time, gain and offset mismatch errors are introduced in the ADC system. These errors cause non harmonic distortion in the sampled signal. One way to decrease the impact of the mismatch errors is to spread the distortion over a wider frequency range by randomizing the order in which the ADCs are used in the interleaved structure. In this paper we analyze how the spectrum is affected by mismatch errors in a randomly interleaved ADC system. We also discuss how the mismatch errors can be estimated.

  • De novo DHDDS variants cause a neurodevelopmental and neurodegenerative disorder with myoclonus

    • Subcellular membrane systems are highly enriched in dolichol, whose role in organelle homeostasis and endosomal-lysosomal pathway remains largely unclear besides being involved in protein glycosylation. DHDDS encodes for the catalytic subunit (DHDDS) of the enzyme cis-prenyltransferase (cis-PTase), involved in dolichol biosynthesis and dolichol-dependent protein glycosylation in the endoplasmic reticulum. An autosomal recessive form of retinitis pigmentosa (retinitis pigmentosa 59) has been associated with a recurrent DHDDS variant. Moreover, two recurring de novo substitutions were detected in a few cases presenting with neurodevelopmental disorder, epilepsy, and movement disorder. We evaluated a large cohort of patients (n=25) with de novo pathogenic variants in DHDDS and provided the first systematic description of the clinical features and long-term outcome of this new neurodevelopmental and neurodegenerative disorder. The functional impact of the identified variants was explored by yeast complementation system and enzymatic assay. Patients presented during infancy or childhood with a variable association of neurodevelopmental disorder, generalized epilepsy, action myoclonus/cortical tremor, and ataxia. Later in the disease course they experienced a slow neurological decline with the emergence of hyperkinetic and/or hypokinetic movement disorder, cognitive deterioration, and psychiatric disturbances. Storage of lipidic material and altered lysosomes were detected in myelinated fibers and fibroblasts, suggesting a dysfunction of the lysosomal enzymatic scavenger machinery. Serum glycoprotein hypoglycosylation was not detected and, in contrast to retinitis pigmentosa and other congenital disorders of glycosylation involving dolichol metabolism, the urinary dolichol D18/D19 ratio was normal. Mapping the disease-causing variants into the protein structure revealed that most of them clustered around the active site of the DHDDS subunit. Functional studies using yeast complementation assay and in vitro activity measurements confirmed that these changes affected the catalytic activity of the cis-PTase and showed growth defect in yeast complementation system as compared with the wild-type enzyme and retinitis pigmentosa-associated protein. In conclusion, we characterized a distinctive neurodegenerative disorder due to de novo DHDDS variants, which clinically belongs to the spectrum of genetic progressive encephalopathies with myoclonus. Clinical and biochemical data from this cohort depicted a condition at the intersection of congenital disorders of glycosylation and inherited storage diseases with several features akin to of progressive myoclonus epilepsy such as neuronal ceroid lipofuscinosis and other lysosomal disorders.

  • Genetic studies of body mass index yield new insights for obesity biology.

    • Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

    • Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms.

  • Structural Dynamics of Light-Driven Proton Pumps

    • Bacteriorhodopsin and proteorhodopsin are simple heptahelical proton pumps containing a retinal chromophore covalently bound to helix G via a protonated Schiff base. Following the absorption of a photon, all-trans retinal is isomerized to a 13-cis conformation, initiating a sequence of conformational changes driving vectorial proton transport. In this study we apply time-resolved wide-angle X-ray scattering to visualize in real time the helical motions associated with proton pumping by bacteriorhodopsin and proteorhodopsin. Our results establish that three conformational states are required to describe their photocycles. Significant motions of the cytoplasmic half of helix F and the extracellular half of helix C are observed prior to the primary proton transfer event, which increase in amplitude following proton transfer. These results both simplify the structural description to emerge from intermediate trapping studies of bacteriorhodopsin and reveal shared dynamical principles for proton pumping.

    • Approaches exploiting trait distribution extremes may be used to identify loci associated with common traits, but it is unknown whether these loci are generalizable to the broader population. In a genome-wide search for loci associated with the upper versus the lower 5th percentiles of body mass index, height and waist-to-hip ratio, as well as clinical classes of obesity, including up to 263,407 individuals of European ancestry, we identified 4 new loci (IGFBP4, H6PD, RSRC1 and PPP2R2A) influencing height detected in the distribution tails and 7 new loci (HNF4G, RPTOR, GNAT2, MRPS33P4, ADCY9, HS6ST3 and ZZZ3) for clinical classes of obesity. Further, we find a large overlap in genetic structure and the distribution of variants between traits based on extremes and the general population and little etiological heterogeneity between obesity subgroups.

  • Getting to grips with automated prepreg handling

    • Automated manufacturing of composite parts based on prepreg material is receiving increased interest with the rising use of composite materials for high-performance applications. The two main automation alternatives for prepreg layup, automated tape layup and fiber placement, are not cost-effective for all types of products, and manual labor is common for the manufacturing of complex parts in low manufacturing volumes. Alternatives to the two dominant automation solutions have been pursued, but so far these have had a limited impact in industrial applications. This paper presents four different solutions for automated layup of prepreg plies to flat laminates that can be formed in a subsequent forming process, which decrease the system complexity. The solutions target products where the layup of flat laminates today is done manually due to technical limitations or cost considerations. The layup solutions must manage challenges caused by material properties such as low material rigidity and tack, and be able to handle a high number of ply shapes. All four solutions are designed for prepreg that is covered with a stiff backing paper. The aim of the paper is to analyze and compare the four solutions. It can be concluded that the most versatile solution is a dual-arm robot equipped with simple end effectors. The dual-arm solution presents a possibility to control the pick-up and lay-down motions that make peeling motions possible, which is advantageous when picking material that has tacked to the pickup surface.
