  • Frequency and typing of Propionibacterium acnes in prostate tissue obtained from men with and without prostate cancer

    • Background: Prostate cancer is the most common cancer among men in Western countries but the exact pathogenic mechanism of the disease is still largely unknown. An infectious etiology and infection-induced inflammation has been suggested to play a role in prostate carcinogenesis and Propionibacterium acnes has been reported as the most prevalent microorganism in prostatic tissue. We investigated the frequency and types of P. acnes isolated from prostate tissue samples from men with prostate cancer and from control patients without the disease.Methods: We included 100 cases and 50 controls in this study. Cases were men diagnosed with prostate cancer undergoing radical prostatectomy and controls were men undergoing surgery for bladder cancer without any histological findings of prostate cancer. Six biopsies taken from each patient's prostate gland at the time of surgery were used for cultivation and further characterization of P. acnes.Results: The results revealed that P. acnes was more common in men with prostate carcinoma than in controls, with the bacteria cultured in 60 % of the cases vs. 26 % of the controls (p = 0.001). In multivariable analyses, men with P. acnes had a 4-fold increase in odds of a prostate cancer diagnosis after adjustment for age, calendar year of surgery and smoking status (OR: 4.46; 95 % CI: 1.93-11.26). To further support the biologic plausibility for a P. acnes infection as a contributing factor in prostate cancer development, we subsequently conducted cell-based experiments. P. acnes- isolates were co-cultured with the prostate cell line PNT1A. An increased cell proliferation and cytokine/chemokine secretion in infected cells was observed.Conclusion: The present study provides further evidence for a role of P. acnes in prostate cancer development.

  • Multilocus sequence typing and repetitive-sequence-based PCR (DiversiLab) for molecular epidemiological characterization of Propionibacterium acnes isolates of heterogeneous origin

    • Propionibacterium acnes is a gram-positive bacillus predominantly found on the skin. Although it is considered an opportunistic pathogen it is also been associated with severe infections. Some specific P. acnes subtypes are hypothesized to be more prone to cause infection than others. Thus, the aim of the present study was to investigate the ability to discriminate between P. acnes isolates of a refined multilocus sequence typing (MLST) method and a genotyping method, DiversiLab, based on repetitive-sequence-PCR technology.The MLST and DiversiLab analysis were performed on 29 P. acnes isolates of diverse origins; orthopedic implant infections, deep infections following cardiothoracic surgery, skin, and isolates from perioperative tissue samples from prostate cancer. Subtyping was based on recA, tly, and Tc12S sequences.The MLST analysis identified 23 sequence types and displayed a superior ability to discriminate P. acnes isolates compared to DiversiLab and the subtyping. The highest discriminatory index was found when using seven genes. DiversiLab was better able to differentiate the isolates compared to the MLST clonal complexes of sequence types.Our results suggest that DiversiLab can be useful as a rapid typing tool for initial discrimination of P. acnes isolates. When better discrimination is required, such as for investigations of the heterogeneity of P. acnes isolates and its involvement in different pathogenic processes, the present MLST protocol is valuable.

    • Detection and identification of pathogens in environmental samples for biosecurity applications are challenging due to the strict requirements on specificity, sensitivity and time. We have developed a concept for quick, specific and sensitive pathogen identification in environmental samples. Target identification is realized by padlock- and proximity probing, and reacted probes are amplified by RCA (rolling-circle amplification). The individual RCA products are labeled by fluorescence and enumerated by an instrument, developed for sensitive and rapid digital analysis. The concept is demonstrated by identification of simili biowarfare agents for bacteria (Escherichia coli and Pantoea agglomerans) and spores (Bacillus atrophaeus) released in field.

    • Globalisation, competitive markets and increasing sustainabilityrequirements are demanding companies to focus on visibility to improve theirsupply chains and ultimately their businesses. This paper aims to identifyperspectives, principles and prospects of production logistics visibility (PLV). Thestudy is based on a literature review of articles presenting definitions, developmenttrends and the future role of PLV. It is concluded that visibility is generally definedby availability, quality, accessibility and usefulness of information. PLV could referto the extent to which actors within the production systems have access to timelyand accurate information considered useful to their operations. According to thefindings, antecedents of PLV include digitalisation, IoT and connectivity. Thesesteps are required to turn data into meaningful information that can be used fordecision making in production and logistics setting to improve operational andbusiness performance. Furthermore, to fully benefit from PLV, there has to be anintegration of external and internal perspectives. Concluding, the paper definesfuture research efforts including four lines of exploration and development: (1)Intra-site visibility for material management including dynamic synchronisation,takt and resource planning. (2) Supply-oriented visibility for dynamic status andprediction of supply network status. These two initial lines of enquiry should includethe perspectives of stakeholders, parameter, enabling technologies and potentialimpact. (3) A synthesised framework for Production Logistics Visibility, relying onutilizing antecedents and enabling multi-criteria decision in production logisticsbased on visibility, where performance in terms of efficiency, sustainability andflexibility is ensured. (4) To specifically detail and exploit the potential inproduction logistics visibility in the aspect of environmental sustainability andclosed material and product loops

    • The aim of this study was to determine antibiotic susceptibility of Propionibacterium acnes isolates from prostate. Prostate-derived P. acnes isolates (n = 24, Umeå & Örebro, Sweden, 2007-2010) and a panel of control strains (n = 25, Sweden) collected from skin and deep infections were assessed for resistance to penicillin G, piperacillin-tazobactam, imipenem, gentamicin, azithromycin, erythromycin, vancomycin, ciprofloxacin, moxifloxacin, tetracycline, tigecycline, fusidic acid, clindamycin, rifampicin, linezolid, daptomycin, trimethoprim-sulfamethoxazole, and metronidazole. In addition, the isolates were tested for inducible clindamycin resistance. All prostate derived P. acnes isolates displayed wild-type distribution of MIC-values, without evidence of acquired resistance. In the reference panel, 5 of 25 isolates had acquired macrolide resistance with cross-resistance to azithromycin, clindamycin, and erythromycin. In addition, one of these isolates was resistant to tetracycline.

    • The disposal of soil material after ex-situ treatment of contaminated soil is an issue of growing concern. The handling and use of this material are surrounded by numerous regulatory, economic, technical and societal aspects that complicate or hinder recycling. As a consequence, the lack of means of recovery can in the long term bias the whole remedial process. In addition, it can affect the competition between various treatment options such as ex-situ, and in-situ techniques and landfilling. At the same time the materials must not have any negative environmental impacts, and their usage must be compatible with existing risk assessment and management frameworks regarding contaminated land. Other concerns such as a possible distinction against “lightly” contaminated materials, waste status and public acceptance add to the complexity. This paper focuses on Swedish conditions, but does also provide an outlook concerning EU regulation. A summary of leaching and batch tests employed for re-use of soil and waste is presented as well as an overview of the eco-toxicological aspects of treated materials. The main conclusion is that re-cycling of treated soil is desirable from numerous aspects, but has to go along an adequate risk assessment.

  • An Improved Quotation Process within Manufacturing SMEs as Part of a Strategy for Growth

    • This paper is generally focusing on the customer and supplier relation in a product creation perspective. The work has specifically been performed together with a metal working SME, and a main customer to this company. Four criterias for a long term succesful relationship and business success was stated from the beginning of the project: In house efficiency; Openness in the chain of suppliers; Co-operative product development and Precision of quotation. Those four areas are all covered in case studies, in close relations to the SME. The problems to combine close relationships and competitive business between customer´s and suppliers are discussed in the paper. A specific task in the project has been to create a demonstrator, a prototype quotation generator for the SME. The goal for the demonstrator was to increase precision and decrease lead times in the quotation process. An evaluation prooves that this is possible, and the demonstrator will stepwise be implemented in the SME. During the project, the SME has had a significant growth. Investments in automation and 3D CAD are made. The project is funded by Vinnova and the companies involved.

    • Background: The current SARS-CoV-2 pandemic has highlighted a need for easy and safe blood sampling in combination with accurate serological methodology. Venipuncture for testing is usually performed by trained staff at healthcare centres. Long travel distances to healthcare centres in rural regions may introduce a bias of testing towards relatively large communities with closer access. Rural regions are therefore often not represented in population-based data.Aim: The aim of this retrospective cohort study was to develop and implement a strategy for at-home testing in a rural region of Sweden during spring 2021, and to evaluate its role to provide equal health care for its inhabitants.Methods: We developed a sensitive method to measure antibodies to the S-protein of SARS-CoV-2 and optimised this assay for clinical use together with a strategy of at-home capillary blood sampling.Results: We demonstrated that our ELISA gave comparable results after analysis of capillary blood or serum from SARS-CoV-2-experienced individuals. We demonstrated stability of the assay under conditions that reflected temperature and humidity during winter or summer. By assessment of capillary blood samples from 4,122 individuals, we could show both feasibility of the strategy and that implementation shifted the geographical spread of testing in favour of rural areas.Conclusion: Implementation of at-home sampling enabled citizens living in remote rural areas access to centralised and sensitive laboratory antibody tests. The strategy for testing used here could therefore enable disease control authorities to get rapid access to information concerning immunity to infectious diseases, even across vast geographical distance.
