- Hypoglycemic sulfonylureas represent a group of clinically useful antidiabetic compounds that stimulate insulin secretion from pancreatic beta cells. The molecular mechanisms involved are not fully understood but are believed to involve inhibition of potassium channels sensitive to adenosine triphosphate (KATP channels) in the beta cell membrane, causing membrane depolarization, calcium influx, and activation of the secretory machinery. In addition to these effects, sulfonylureas also promoted exocytosis by direct interaction with the secretory machinery not involving closure of the plasma membrane KATP channels. This effect was dependent on protein kinase C (PKC) and was observed at therapeutic concentrations of sulfonylureas, which suggests that it contributes to their hypoglycemic action in diabetics.

- Det område som idag kallas Strövområdet Arriesjön har bytt skepnad flera gånger under de senaste tre hundra åren. De processer och den historia som åstadkommit dessa förändringar speglas i de två historiska kapitlen. Men i det här kapitlet ger vi en översikt över områdets rent fysiska förändringar med hjälp av de flesta kartor och flygfoton som finns tillgängliga, från den första kartbilden 1702 till Region Skånes karta över dagens strövområde. På det sättet framträder inte bara förändringarna utan även det som består – Risebjärs plats i landskapet, topografin, de fuktiga områdena i öster och söder som idag är öppna vattenspeglar, och de bestående gränserna.

Den fjärde kvadrantens dilemma : Kunskapsbedömning i en föränderlig historiekultur

- The purpose of this dissertation is to explore how history teachers in Swedish upper-secondary school perceive voluntary external tests in history. The study focuses on history teachers' perceptions of 1) the design of the tests, 2) the implementation of the tests, 3) the use of the tests and 4) the impact of the tests on their assessment practice. The overall aim is to investigate how historical culture, as it is expressed in the school subject of history, changes over time. The analytical focus for the study has been to explore the significance of knowledge assessment in general, and voluntary external tests in particular, for this change. The temporal focus for the study has been the 2011 Swedish school policy reforms manifested in the curriculum for upper-secondary school (Gy11).The theoretical framework combines theory of history didactics and theory of assessment. Historical culture and assessment culture are the central concepts of the study. The overall hypothesis of the study is that the relationship between the school's historical-cultural dimension and the assessment-cultural dimension contains a latent tension referred to as the dilemma of the fourth quadrant.The empirical material was collected from qualitative interviews with eight history teachers. The first sub-study included four teachers during the year 2009 who had used a voluntary external test named the History Teachers' Test (HLP). The second sub study included four teachers during the year 2016 who had used a voluntary external test named the Course Test in History (KP).The results show that history teachers perceive and handle the external tests in different ways. One possible interpretation of this difference is that the tests respond to different needs or problems. In the study, two such problem areas have emerged, the equivalence problem and the alignment problem. A conclusion based on the empirical results is that the HLP-teachers use the test results for a summative purpose to deal with an equivalence problem, while the KP-teachers use the test tools for a formative purpose to deal with an alignment problem. The results also show that there is strong a connection between the school's historical-cultural dimension and assessment-cultural dimension, manifested in the history teachers' different ways of perceiving and using voluntary external tests.

- Both stearic acid and methyl stearate chemisorbs onto an oxide surface of aluminum with an asymmetric coordination of the carboxylate group as concluded from infrared (IR) spectroscopy data. Similarities in the IR spectra of the films from the two compounds suggest that the ester is bonded in the same way as the acid, and that the ester therefore undergoes hydrolysis during the surface reaction. X-Ray photoelectron spectroscopy (XPS) and IR data are interpreted in terms of self-assembled monolayer formation and a more dense film from the carboxylic acid in comparison with that from the ester. Large-scale occurrence patterns of red-listed lichens and fungi on old oaks are influenced both by current and historical habitat density

- Current occurrence patterns of species associated with ancient trees may reflect higher historical habitat densities, because the dynamics of the habitat and the colonisation-extinction processes for many inhabiting species are expected to be slow. We tested this hypothesis in southeast Sweden by analysing species occurrence per parish for twelve redlisted lichen species and nine redlisted fungus species in relation with current density of big oaks, the density of oaks in the 1830s and connectivity with parishes with the species present. For most species, the occurrence was positively related with current density of habitat (for 18 species out of 21) and parish area (for 16 species). Historical habitat density was positively related with occurrence for 11 species, while connectivity with current occurrences in the surroundings was positive for the occurrence of 12 species and negative for the occurrence of two. For lichen species the connectivity measure that best explained the variation was at a larger spatial scale as compared to fungus species. Even if the density of old oaks remains in the future, inhabiting species will most likely decline because their distribution patterns are not in equilibrium with the current habitat density. Therefore, to allow long-term persistence of inhabiting species the number of old oaks should be increased. Areas where such an increase is most urgent could be identified based on species occurrence data and current habitat density, but because species data will always be incomplete data on the historical habitat distribution is valuable.

DNA methylation profiling improves routine diagnosis of paediatric central nervous system tumours: A prospective population-based study

- Aims: Paediatric brain tumours are rare, and establishing a precise diagnosis can be challenging. Analysis of DNA methylation profiles has been shown to be a reliable method to classify central nervous system (CNS) tumours with high accuracy. We aimed to prospectively analyse CNS tumours diagnosed in Sweden, to assess the clinical impact of adding DNA methylation-based classification to standard paediatric brain tumour diagnostics in an unselected cohort. Methods: All CNS tumours diagnosed in children (0-18 years) during 2017-2020 were eligible for inclusion provided sufficient tumour material was available. Tumours were analysed using genome-wide DNA methylation profiling and classified by the MNP brain tumour classifier. The initial histopathological diagnosis was compared with the DNA methylation-based classification. For incongruent results, a blinded re-evaluation was performed by an experienced neuropathologist. Results: Two hundred forty tumours with a histopathology-based diagnosis were profiled. A high-confidence methylation score of 0.84 or more was reached in 78% of the cases. In 69%, the histopathological diagnosis was confirmed, and for some of these also refined, 6% were incongruent, and the re-evaluation favoured the methylation-based classification. In the remaining 3% of cases, the methylation class was non-contributory. The change in diagnosis would have had a direct impact on the clinical management in 5% of all patients. Conclusions: Integrating DNA methylation-based tumour classification into routine clinical analysis improves diagnostics and provides molecular information that is important for treatment decisions. The results from methylation profiling should be interpreted in the context of clinical and histopathological information.
