    • Ionizable pharmaceuticals are a class of emerging contaminants that pose a challenge to analytical chemistry due to their low environmental concentrations. To measure such low concentrations in organism tissue, e.g. fish muscle, specific extraction techniques minimizing co-extraction and interference alongside providing high enrichment of the compounds are needed. In this study we present a technique using hollow fiber liquid phase microextraction which is selective and highly enriching due to a pH gradient across a selective membrane, trapping ions in the extract. Microextraction minimizes the use of organic solvents, thereby making the technique "green". We used high volume pharmaceuticals for method development, specifically, the weak acids ketoprofen, naproxen, diclofenac and ibuprofen, and the weak bases fluoxetine and sertraline. Lyophilized tissue extraction gave higher enrichment than fresh tissue extraction and concentration enrichment factors ranged from 1900 to 3000 times. Method detection limits with the analysis instruments used in this study were for ketoprofen, 0.23 ng g(-1) fish tissue; naproxen, 0.32 ng g(-1) fish tissue; diclofenac, 0.12 ng g(-1) fish tissue; ibuprofen, 0.34 ng g-1 fish tissue; fluoxetine, 13 ng g-1. fish tissue and sertraline, 23 ng g(-1) fish tissue. All analytes were successfully detected in tissue from fish exposed live via spiked water. The resulting extraction parameters shown in this study suggest the developed technique to be a useful work up method for extensive environmental data collection as well as for toxicokinetic studies.

    • Industrial interventions that focus on increased productivity may impair the ergonomics, on a workstation or individual level. This paper presents a method that characterises work time consumption and physical work load of manual work, using video recordings synchronised with physiological measurements of, e.g. muscular activity, and postures. The underlying idea was that it is possible to amalgamate technical and human aspects resulting in a synergetic evaluation. The method was developed through two case studies within the Swedish automotive industry, where manual materials handling was studied. A methodological result was that the synchronising procedure was sufficiently precise to allow work activities to be assigned significantly different levels of physical work load. These different levels may be used to predict physical work load in the design and change of production systems. It was concluded that the method is accurate enough to be a useful tool in industrial interventions. (C) 2002 Elsevier Science Ltd. All rights reserved.

    • B10.RIII mice develop chronic and relapsing experimental autoimmune encephalomyelitis (EAE) after immunization with the myelin basic protein (MBP) peptide 89-101 (VHFFKNIVTPRTP). To investigate the basis for the chronicity of the disease, the subsequent development of an immune responses to other parts of the MBP protein were investigated. Onset of disease occurs 9-25 days after immunization with MBP89-101. T cell responses towards a series of MBP peptides were assessed in an enzyme-linked immunospot assay detecting single cells secreting IFN-gamma. There were responses not only to MBP89-101, but also towards peptides derived from sequences outside of MBP89-101. These peptides were of two kinds: those with sequences completely outside the 89-101 stretch of MBP; and those sharing a short sequence with MBP89-101 depending on alternative splicing of MBP mRNA. Immunization with these peptides also produced chronic EAE and a spreading of the immune response to other MBP peptides. Immunization with stepped peptides around the relevant region (MBP87-110) showed that peptides sharing a 6-amino-acid motif induced EAE after immunization. After MBP89-101 peptide immunization, T cells isolated from lymph nodes did not cross-react in vitro to the other peptides sharing this motif. We suggest that one mechanism for the development of relapses during the disease course is the recruitment of new T cells with specificity for MBP peptides not derived from the peptide used for immunization. This is the first time such a mechanism has been demonstrated in a chronic autoimmune disease model.

  • The Human Eosinophil Proteome. Changes Induced by Birch Pollen Allergy

    • Proteins from human eosinophils were separated bidimensionally and identified by mass spectrometry (336 spots/bands, 98 different proteins). Of these, 24.7% belonged to the cytoskeleton/migration group. Highly basic proteins (11.3%) were concentrated in the granule-containing cell fraction. We detected novel hyperacidic forms of cofilin-1, profilin-1 and adenylyl cyclase-associated protein, and hyperbasic forms of eosinophil-derived neurotoxin/eosinophil protein X and major basic protein homologue. We also found evidence of the triglycosylation of the heavy chain of eosinophil peroxidase. In addition, through comparative 2D image analysis, spot quantification and MS, it was found that hsc70, actin-capping protein and hyperacidic forms of eosinophil peroxidase heavy chain are overexpressed in cells from birch pollen allergic subjects, at the peak of a season. The link between these findings and an increased cellular antigen-presenting capacity and motility are discussed.
