  • A polymorphism in the serotonin receptor 3A (HTR3A) gene and its association with harm avoidance in women.

    • BACKGROUND: The brain neurotransmitter serotonin is known to affect various aspects of human behavior, including personality traits. Serotonin receptor type 3 is a ligand-gated channel encoded by 2 different subunit genes, HTR3A and HTR3B. A polymorphism (C178T) in the 5' region of the HTR3A gene has recently been identified and suggested to be of functional importance. OBJECTIVE: To elucidate the possible association between the C178T polymorphism in the HTR3A gene and personality traits in women. DESIGN: Two independent samples of 35- to 45-year-old Swedish women were recruited using the population register. Sample 1 (n = 195) was assessed via the Karolinska Scales of Personality and the Temperament and Character Inventory; sample 2 (n = 175) was assessed using the latter only. Both samples were genotyped with respect to the C178T polymorphism in the HTR3A gene. The A1596G polymorphism in the same gene was also investigated. RESULTS: A significant association between C178T genotype and the Temperament and Character Inventory factor harm avoidance was observed in sample 1 (corrected for multiple comparisons P =.04); this finding was subsequently replicated in sample 2 (P =.004) (pooled populations: P<.001). In the pooled sample, all harm avoidance subscales were found to be significantly associated with the C178T polymorphism: anticipatory worry (P =.001), fear of uncertainty (P<.001), shyness (P<.001), and fatigability and asthenia (P =.008). In addition, a significant association was found in sample 1 between the C178T polymorphism and the Karolinska Scales of Personality nonconformity factor (corrected P =.002), including the subscales of social desirability (P<.001), indirect aggression (P =.002), verbal aggression (P =.05), and irritability (P<.001). Participants homozygous for the less common T allele (<4%) differed from the remaining women by displaying lower ratings on harm avoidance and nonconformity. CONCLUSION: The C178T polymorphism in the HTR3A gene may affect the personality trait of harm avoidance in women.

  • The programming language GCLA: A definitional approach to logic programming

    • We present a logic programming language, GCLA (Generalized horn Clause LAnguage), that is based on a generalization of Prolog. This generalization is unusual in that it takes a quite different view of the meaning of a logic program--a "definitional" view rather than the traditional logical view. GCLA has a number of noteworthy properties, for instance hypothetical and non-monotonic reasoning. This makes implementation of reasoning in knowledge-based systems more direct in GCLA than in Prolog. GCLA is also general enough to incorporate functional programming as a special case. GCLA and its syntax and semantics are described. The use of various language constructs are illustrated with several examples.

    • Postoperative neurocognitive decline is a frequent complication in adult patients undergoing major surgery with increased risk for morbidity and mortality. The mechanisms behind cognitive decline after anaesthesia and surgery are not known. We studied the association between CSF and blood biomarkers of neuronal injury or brain amyloidosis and long-term changes in neurocognitive function.In patients undergoing major orthopaedic surgery (knee or hip replacement), blood and CSF samples were obtained before surgery and then at 4, 8, 24, 32, and 48 h after skin incision through an indwelling spinal catheter. CSF and blood concentrations of total tau (T-tau), neurofilament light, neurone-specific enolase and amyloid β (Aβ1-42) were measured. Neurocognitive function was assessed using the International Study of Postoperative Cognitive Dysfunction (ISPOCD) test battery 1-2 weeks before surgery, at discharge from the hospital (2-5 days after surgery), and at 3 months after surgery.CSF and blood concentrations of T-tau, neurone-specific enolase, and Aβ1-42 increased after surgery. A similar increase in serum neurofilament light was seen with no overall changes in CSF concentrations. There were no differences between patients having a poor or good late postoperative neurocognitive outcome with respect to these biomarkers of neuronal injury and Aβ1-42.The findings of the present explorative study showed that major orthopaedic surgery causes a release of CSF markers of neural injury and brain amyloidosis, suggesting neuronal damage or stress. We were unable to detect an association between the magnitude of biomarker changes and long-term postoperative neurocognitive dysfunction.

    • Sorting a permutation by block moves is a task that every bridge player has to solve every time she picks up a new hand of cards. It is also a problem for the computational biologist, for block moves are a fundamental type of mutation that can explain why genes common to two species do not occur in the same order in the chromosome, It is not known whether there exists an optimal sorting procedure running in polynomial time. Bafna and Pevzner gave a polynomial time algorithm that sorts any permutation of length n in at most 3n/4 moves. Our new algorithm improves this to [(2n - 2)/3] for n greater than or equal to 9. For the reverse permutation, we give an exact expression for the number of moves needed, namely [(n + 1)/2]. Computations of Bafha and Pevzner up to n = 10 seemed to suggest that this is the worst case; but as it turns out, a first counterexample occurs for n = 13, i.e. the bridge player's case. Professional card players never sort by rank, only by suit. For this case, we give a complete answer to the optimal sorting problem.
