  • Thirty new loci for age at menarche identified by a meta-analysis of genome-wide association studies

    • To identify loci for age at menarche, we performed a meta-analysis of 32 genome-wide association studies in 87,802 women of European descent, with replication in up to 14,731 women. In addition to the known loci at LIN28B (P = 5.4 × 10⁻⁶⁰) and 9q31.2 (P = 2.2 × 10⁻³³), we identified 30 new menarche loci (all P < 5 × 10⁻⁸) and found suggestive evidence for a further 10 loci (P < 1.9 × 10⁻⁶). The new loci included four previously associated with body mass index (in or near FTO, SEC16B, TRA2B and TMEM18), three in or near other genes implicated in energy homeostasis (BSX, CRTC1 and MCHR2) and three in or near genes implicated in hormonal regulation (INHBA, PCSK2 and RXRG). Ingenuity and gene-set enrichment pathway analyses identified coenzyme A and fatty acid biosynthesis as biological processes related to menarche timing.

  • Genetic variants associated with subjective well-being, depressive symptoms, and neuroticism identified through genome-wide analyses

    • Very few genetic variants have been associated with depression and neuroticism, likely because of limitations on sample size in previous studies. Subjective well-being, a phenotype that is genetically correlated with both of these traits, has not yet been studied with genome-wide data. We conducted genome-wide association studies of three phenotypes: subjective well-being (n = 298,420), depressive symptoms (n = 161,460), and neuroticism (n = 170,911). We identify 3 variants associated with subjective well-being, 2 variants associated with depressive symptoms, and 11 variants associated with neuroticism, including 2 inversion polymorphisms. The two loci associated with depressive symptoms replicate in an independent depression sample. Joint analyses that exploit the high genetic correlations between the phenotypes (vertical bar(p) over cap vertical bar approximate to 0.8) strengthen the overall credibility of the findings and allow us to identify additional variants. Across our phenotypes, loci regulating expression in central nervous system and adrenal or pancreas tissues are strongly enriched for association.

    • The impact of many unfavorable childhood traits or diseases, such as low birth weight and mental disorders, is not limited to childhood and adolescence, as they are also associated with poor outcomes in adulthood, such as cardiovascular disease. Insight into the genetic etiology of childhood and adolescent traits and disorders may therefore provide new perspectives, not only on how to improve wellbeing during childhood, but also how to prevent later adverse outcomes. To achieve the sample sizes required for genetic research, the Early Growth Genetics (EGG) and EArly Genetics and Lifecourse Epidemiology (EAGLE) consortia were established. The majority of the participating cohorts are longitudinal population-based samples, but other cohorts with data on early childhood phenotypes are also involved. Cohorts often have a broad focus and collect(ed) data on various somatic and psychiatric traits as well as environmental factors. Genetic variants have been successfully identified for multiple traits, for example, birth weight, atopic dermatitis, childhood BMI, allergic sensitization, and pubertal growth. Furthermore, the results have shown that genetic factors also partly underlie the association with adult traits. As sample sizes are still increasing, it is expected that future analyses will identify additional variants. This, in combination with the development of innovative statistical methods, will provide detailed insight on the mechanisms underlying the transition from childhood to adult disorders.   • Implementation of a Discrete Dipole Approximation Scattering Database Into Community Radiative Transfer Model

    • The Community Radiative Transfer Model (CRTM) is a fast model that requires bulk optical properties of hydrometeors in the form of lookup tables to simulate all-sky satellite radiances. Current cloud scattering lookup tables of CRTM were generated using the Mie-Lorenz theory thus assuming spherical shapes for all frozen habits, while actual clouds contain frozen hydrometeors with different shapes. The Discrete Dipole Approximation (DDA) technique is an effective technique for simulating the optical properties of non-spherical hydrometeors in the microwave region. This paper discusses the implementation and validation of a comprehensive DDA cloud scattering database into CRTM for the microwave frequencies. The original DDA database assumes total random orientation in the calculation of single scattering properties. The mass scattering parameters required by CRTM were then computed from single scattering properties and water content dependent particle size distributions. The new lookup tables eliminate the requirement for providing the effective radius as input to CRTM by using the cloud water content for the mass dimension. A collocated dataset of short-term forecasts from Integrated Forecast System of the European Center for Medium-Range Weather Forecasts and satellite microwave data was used for the evaluation of results. The results overall showed that the DDA lookup tables, in comparison with the Mie tables, greatly reduce the differences among simulated and observed values. The Mie lookup tables especially introduce excessive scattering for the channels operating below 90 GHz and low scattering for the channels above 90 GHz.

  • Genome-wide association study identifies 74 loci associated with educational attainment

    • Educational attainment is strongly influenced by social and other environmental factors, but genetic factors are estimated to account for at least 20% of the variation across individuals. Here we report the results of a genome-wide association study (GWAS) for educational attainment that extends our earlier discovery sample of 101,069 individuals to 293,723 individuals, and a replication study in an independent sample of 111,349 individuals from the UK Biobank. We identify 74 genome-wide significant loci associated with the number of years of schooling completed. Single-nucleotide polymorphisms associated with educational attainment are disproportionately found in genomic regions regulating gene expression in the fetal brain. Candidate genes are preferentially expressed in neural tissue, especially during the prenatal period, and enriched for biological pathways involved in neural development. Our findings demonstrate that, even for a behavioural phenotype that is mostly environmentally determined, a well-powered GWAS identifies replicable associated genetic variants that suggest biologically relevant pathways. Because educational attainment is measured in large numbers of individuals, it will continue to be useful as a proxy phenotype in efforts to characterize the genetic influences of related phenotypes, including cognition and neuropsychiatric diseases.

    • To newly identify loci for age at natural menopause, we carried out a meta-analysis of 22 genome-wide association studies (GWAS) in 38,968 women of European descent, with replication in up to 14,435 women. In addition to four known loci, we identified 13 loci newly associated with age at natural menopause (at P < 5 × 10(-8)). Candidate genes located at these newly associated loci include genes implicated in DNA repair (EXO1, HELQ, UIMC1, FAM175A, FANCI, TLK1, POLG and PRIM1) and immune function (IL11, NLRP11 and PRRC2A (also known as BAT2)). Gene-set enrichment pathway analyses using the full GWAS data set identified exoDNase, NF-κB signaling and mitochondrial dysfunction as biological processes related to timing of menopause.

    • Remote sensing of atmospheric state variables typically relies on the inverse solution of the radiative transfer equation. An adequately characterized retrieval provides information on the uncertainties of the estimated state variables as well as on how any constraint or a priori assumption affects the estimate. Reported characterization data should be intercomparable between different instruments, empirically validatable, grid-independent, usable without detailed knowledge of the instrument or retrieval technique, traceable and still have reasonable data volume. The latter may force one to work with representative rather than individual characterization data. Many errors derive from approximations and simplifications used in real-world retrieval schemes, which are reviewed in this paper, along with related error estimation schemes. The main sources of uncertainty are measurement noise, calibration errors, simplifications and idealizations in the radiative transfer model and retrieval scheme, auxiliary data errors, and uncertainties in atmospheric or instrumental parameters. Some of these errors affect the result in a random way, while others chiefly cause a bias or are of mixed character. Beyond this, it is of utmost importance to know the influence of any constraint and prior information on the solution. While different instruments or retrieval schemes may require different error estimation schemes, we provide a list of recommendations which should help to unify retrieval error reporting.

    • BaZr0.9In0.05M0.05O3-d (M ¼ Ga3+ or Yb3+) has been prepared by solid-statesynthesis route.Rietveld analysis of neutron powder diffraction data on as-prepared samples showed thatboth samples crystallise in the cubic space group Pm-3m. Scanning electron microscopyanalysis showed that the grains intheBaZr0.9In0.05Ga0.05O3-d sample were larger than the BaZr0.9In0.05Yb0.05O3-d sample. Dynamic thermogravimetric analysis indicates that the proton concentration was higher inpre-hydrated BaZr0.9In0.05Ga0.05O3-d sample (40%oftheoretical)than that of BaZr0.9In0.05Yb0.05O3-d sample (28%oftheoretical).Proton conductivity was studied on pre-hydrated samples(under both dry and wetAr atmospheres).Thebulk proton conductivities of the heating cycle of pre-hydrated BaZr0.9In0.05M0.05O3-d (M ¼ Ga3+ or Yb3+) samples were 2.1 * 10-5 S cm-1 and 1.9 * 10-4 Scm-1 at 350C, respectively.The effect of co-doping onproton conductivity was investigatedand the results are compared with single doped systems e.g. BaZr0.9M0.1O3-d (M=Ga3+, In3+ orYb3+) samples.

    • Spark-plasma sintering method was used to prepare dense proton conducting perovskite oxide BaZr0.5In0.5O3-delta. Analysis of X-ray powder diffraction data showed that the sample adopt the cubic crystal structure having the space group Pm3m. Thermogravimetric analysis of prehydrated samples showed significant mass losses beyond 300 degrees C due to loss of protons as water vapor. Scanning electron microscope images show that the grain size of the spark-plasma sintered dense sample was smaller than that of solid-state sintered porous sample. The highest total proton conductivity (2 X 10(-3) S cm(-1) at 450 degrees C) was found for dense spark-plasma sintered sample under wet H-2 than the samples prepared by other routes.

    • A wet chemical route was used to prepare the oxygen deficient codoped perovskite oxide BaZr0.5In0.25Yb0.25O3−. Analysis of X-ray powder diffraction data showed that the sample belongs to the cubic crystal system with space group Pmm. Dynamic thermogravimetric (TG) analysis confirmed complete filling of oxygen vacancies (V) by protonic defects (OH) during the hydration process. The proton conductivity was investigated by impedance spectroscopy. The bulk and total conductivities of prehydrated BaZr0.5In0.25Yb0.25O3− were found to be 8.5×10−4 and 2.2×10−5 S cm−1, respectively, at 300°C. The total conductivity in the codoped perovskite oxide was higher compared to that of the respective single doped perovskite oxides with the same doping level. The bulk and grain-boundary mobility and diffusion coefficients of protons were calculated at 200°C using impedance and TG data to obtain the conductivity and proton concentration, respectively. The high bulk diffusivity (2.3×10−7 cm2 s−1) was obtained which indicates that the protons are more free to move in the heavily doped matrix compared to the lightly doped systems where trapping of protons occurs.
