Workers’ participation in regional economic change following establishment closure

- This article analyses if and when workers affected by economic destruction in the form of establishment closures move to more productive or newly started establishments in the region, become self-employed, leave the region or become displaced. Results from multinominal probit models show that the majority of these workers face destructive employment outcomes from a Schumpeterian point of view compared to a matched sample of workers not subject to a closure. However, we do find indications of a creative destruction as a small, albeit significant, share become employed in young establishments. Different types of human capital influence the likelihood of triggering positive or negative regional outcomes. While higher education significantly decreases the risk for unemployment, high-income earners more often become engaged in creative outcomes. Firm tenure increases the likelihood of becoming employed in younger establishments. There are significant spatial differences where metropolitan regions excel as loci of creative change, whereas smaller and peripheral regions face far less creative outcomes of economic transformation.

- Collaborative contracting models are often associated with a set of tools and techniques to manage relationships, but the efficiency of such formalization in changing project culture has been doubted. Further, although many projects are successful, collaboration often is more limited than policies and guidelines suggest. In this paper, we view partnering practice as a learning process related to a management innovation and analyse how collaboration practice develops in three major railway projects, all using the same partnering model. We find that partnering is easy to introduce due to the flexibility and adaptability of the concept, but that practitioners prefer to keep collaboration informal and groups small. Also, tangible benefits can often be reached with basic and common-sense approaches. When ambitions and complexity increase, however, more sophisticated relationship management becomes inevitable, calling also for integration with core project processes. Yet, partnering tools and systems do not seem to provide much guidance when it comes to organizing such complex multiparty collaboration. Findings suggest that shortcomings relating to organizational issues are underestimated as causes of conflicts and inefficiencies.

Rough starts and tough times : geographies of workers and firms in transition

- Economic change can lead to multiple and sometimes conflicting outcomes for workers, employers, and regions. At the center of economic change are the dynamic interactions among diverse workers and firms in specific labor market contexts. This thesis approached those spatial interactions from the perspective of labor market matching, where the different opportunities for workers and firms to match were investigated. This adds to the growing body of literature that seeks to understand what factors are behind the growing regional divergence in earnings and employment, and who is particularly impacted by the uneven labor market changes. To this end, the aim of this thesis was to analyze the regional patterns and outcomes for workers and firms in the labor market matching process. Four quantitative studies were conducted using data from Swedish administrative registers from 1995 to 2012. Given that the dynamics of labor market matching are complex, the studies approached the aim from different angles and for different groups of workers. In particular, the outcomes and patterns were investigated in relation to crucial periods for workers and firms, where frictions in labor market matching could potentially have particularly negative effects. The studies investigated earnings, employment, and hiring for young workers in low-paid jobs, for workers displaced by firm closures, and for new firms competing for survival. The empirical results indicate the importance of a flexible labor market that facilitates the matching process where individuals' existing competencies can be applied and developed. This is a particular challenge for workers who lose their jobs to establishment closures outside the large regions, since the results show that worse job matches and less productive re-employment opportunities are more common and can have particularly negative consequences in smaller and more peripheral regions. However, even if there are more job opportunities in larger regions, the results suggest the opportunities are not necessarily accessible to everyone. Results indicate that workers in low-wage jobs do not benefit from a boost in earnings in larger regions, which may put them in a precarious situation considering the higher cost of living in those regions. Lastly, the thesis highlights the importance of not only individual human capital in determining a job match, but also its relational dimension which captures how well different workers’ skills are matched to one another in the workplace. This was found to be associated with individual earnings recovery after plant closure and new firm survival. The findings call for policies that carefully combine supply- and demand-side approaches in economic development.

Organelle movement in melanophores: Effects of Panax ginseng, ginsenosides and quercetin

- Panax ginseng is a traditional herb that has been used for over 2000 years to promote health and longevity. Active components of ginseng include ginsenosides, polysaccharides, flavonoids, polyacetylenes, peptides, vitamins, phenols and enzymes, of which the ginsenosides are considered to be the major bioactive constituents. Although widely used, the exact mechanisms of ginseng and its compounds remain unclear. In this thesis we use melanophores from Xenopus laevis to investigate the effects of Panax ginseng extract G115 and its constituents on organelle transport and signalling. Due to coordinated bidirectional movement of their pigmented granules (melanosomes), in response to defined chemical signals, melanophores are capable of fast colour changes and provide a great model for the study of intracellular transport. The movement is regulated by alterations in cyclic adenosine 3’:5’-monophosphate (cAMP) concentration, where a high or low level induce anterograde (dispersion) or retrograde (aggregation) transport respectively, resulting in a dark or light cell. Here we demonstrate that Panax ginseng and its constituents ginsenoside Rc and Rd and flavonoid quercetin induce a concentration-dependent anterograde transport of melanosomes. The effect of ginseng is shown to be independent of cAMP changes and protein kinase A activation. Upon incubation of melanophores with a combination of Rc or Rd and quercetin, a synergistic increase in anterograde movement was seen, indicating cooperation between the ginsenoside and flavonoid parts of ginseng. Protein kinase C (PKC) inhibitor Myristoylated EGF-R Fragment 651-658 decreased the anterograde movement stimulated by ginseng and ginsenoside Rc and Rd. Moreover, ginseng, but not ginsenosides or quercetin, stimulated an activation of 44/42-mitogen activated protein kinase (MAPK), previously shown to be involved in both aggregation and dispersion of melanosomes. PKC-inhibition did not affect the MAPK-activation, suggesting a role for PKC in the ginseng- and ginsenoside-induced dispersion but not as an upstream activator of MAPK.

- Melanophores from Xenopus laevis are pigmented cells, capable of quick colour changes through cyclic adenosine 3':5'-monophosphate (cAMP) coordinated transport of their intracellular pigment granules, melanosomes. In this study we use the melanophore cell line to evaluate the effects of Panax ginseng extract G115 on organelle transport. Absorbance readings of melanophore-coated microplates, Correlate-EIA direct cAMP enzyme immunoassay kit, and western blot were used to measure the melanosome movement and changes in intracellular signalling. We show that Panax ginseng induces a fast concentration-dependent anterograde transport of the melanosomes. No significant increase in the cAMP level was seen and pre-incubation of melanophores with the protein kinase C (PKC) inhibitor EGF-R Fragment 651-658 (M-EGF) only partly decreased the ginseng-induced dispersion. We also demonstrate that Panax ginseng, endothelin-3 (ET-3) and alpha-melanocyte stimulating hormone (MSH) stimulate an activation of mitogen activated protein kinase (MAPK). Pre-incubation with M-EGF decreased the MAPK activity induced by ET-3 and MSH, but again only marginally affected the response of Panax ginseng. Thus, in melanophores we suggest that Panax ginseng stimulates an anterograde transport of pigment organelles via a non-cAMP and mainly PKC-independent pathway.

Active components of ginseng include ginsenosides, polysaccharides, flavonoids, polyacetylenes, peptides, vitamins, phenols and enzymes, of which the ginsenosides are considered to be the major bioactive constituents. In spite of extensive use, the exact mechanisms of ginseng and its components are still unknown. In the present study we use melanophores from Xenopus laevis to investigate the effects of Panax ginseng extract G115, ginsenosides and the flavonoid quercetin on pigment organelle transport and signalling. Through coordinated transport of their black pigmented granules (melanosomes), melanophores are capable of fast colour changes.
