  • Development of hepatic CYP1A and blood vitellogenin in eel (Anguilla anguilla) for use as biomarkers in the Thames Estuary, UK.
  • 2002
  • Ingår i: Marine environmental research. - 0141-1136. ; 50:1-5, s. 367-71
  • Tidskriftsartikel (refereegranskat)abstract
    • The potential of eel (Anguilla anguilla) as a monitoring species for the Thames Estuary, UK, was examined. Hepatic cytochrome P4501A [7-ethoxyresorufin O-deethylase (EROD) activity] and blood vitellogenin (Western analysis) were investigated as biomarkers of exposure to, respectively, organic contaminants and to contaminants showing estrogenic activity. Hepatic microsomal EROD activities in A. anguilla from seven sites in the Thames Estuary in May 1998 varied three-fold (111 +/- 24 to 355 +/- 42 pmol min-1 mg protein-1) (mean +/- S.E.M.) and showed correlation with salinity; however, the latter relationship was not maintained at other times of the year. The range of EROD activities was two- to eight-fold higher than the 37 +/- 8 pmol min-1 mg-1 for A. anguilla from the relatively clean Tamar Estuary. beta-Naphthoflavone treatment (5 mg kg-1 wet wt.; 2 days) of Thames A. anguilla produced a two-fold increase in hepatic microsomal EROD activity. Comparing the Thames EROD data with those for A. anguilla from well-characterised contaminated sites in the Netherlands (Van der Oost, R., Goksøyr, A., Celander, M., Heida, H., & Vermeulen, N. P. E. 1996. Aquatic Toxicology, 36, 189-222), the Thames is suggested to be moderately impacted by polycyclic aromatic hydrocarbons and related contaminants. 17-beta-Estradiol treatment produced the appearance of a plasma protein of 211 Kd app. mol. wt. (recognised by antibodies to vitellogenin of Morone saxatilis), but putative vitellogenin could not be detected in A. anguilla from selected sites in the Thames Estuary.

  • Impact of Polychlorinated Naphthalenes (PCNs) in Juvenile Baltic Salmon, Salmo salar: Evaluation of Estrogenic Effects, Development, and CYP1A Induction
  • 2000
  • Ingår i: Archives of Environmental Contamination and Toxicology. - : Springer Science and Business Media LLC. - 0090-4341 .- 1432-0703. ; 38:2, s. 225-33
  • Tidskriftsartikel (refereegranskat)abstract
    • Juvenile Baltic salmon, Salmo salar, were fed commercial salmon food contaminated with different concentrations of polychlorinated naphthalenes (PCNs; 0.1, 1, 2, or 10 μg PCN/g food). Among other effects, possible estrogenic impact caused by PCNs were evaluated. Fish were therefore fed a diet contaminated with 17β-estradiol (E2; 0.94 or 9.4 μg E2/g food), as a positive control. After 8, 13, 17, and 41 weeks, sampling took place. Growth, liver somatic index (LSI), EROD activity, and vitellogenin content in blood plasma were measured along with morphological studies of gonads and chemical analyses to determine the effects caused by PCNs. Exposure to PCNs did not seem to have any effects on body weight gain, since the group fed the high dose followed the growth in the control group during the entirely experimental period. After 41 weeks of exposure the groups fed 2 and 10 μg PCN/g food had significantly lower LSIs compared with the control, indicating liver toxic effects of PCNs. Furthermore, a dose-dependent induction of EROD activity was found. At week 41, the control group had an activity of 4.9 ± 4.8 pmol/mg prot/min, whereas it was between 69 ± 21 and 720 ± 370 pmol/mg prot/min in the exposed groups, respectively. Examination of gonadal morphology showed that PCNs also had negative effects on ovaries in Baltic salmon, including delayed development. The distribution between females and males, gonadal morphology, and vitellogenin content in blood plasma did, however, indicate that PCNs are not capable of causing effects similar to E2 or xenoestrogens. Exposure to both of the concentrations of E2 resulted in decreased body weight gain, increased LSI, and feminization of the gonads. E2 did, however, not induce any EROD activity.

  • Environmental mutagenesis in different zones of the Baltic Sea
  • 2005
  • Ingår i: Acta Zoologica Lituanica. - 1392-1657. ; 15:2, s. 90-95
  • Tidskriftsartikel (refereegranskat)abstract
    • Abstract. Environmental mutagenesis was assessed in fish from the selected coastal areas of the Baltic SeaKvadofjarden and the Stockholm archipelago (Sweden), the KlaipëdaBûtingë zone (Lithuania), the Gulf of Gdansk (Poland) and the Wismar Bay (Germany). The frequency of micronuclei, as markers of cytogenetic damage, was evaluated in erythrocytes of 911 flounder (Platichthys flesus), 332 perch (Perca fluviatilis) and 458 eelpout (Zoarces viviparus) specimens. Fish was collected during bi-annual sampling campaigns performed in 2001 and 2002. Statistically significant differences (p < 0.0001) regarding season and sampling location were recorded. The highest values of cytogenetic damage were registered in flounder from the Lithuanian coast near Palanga (autumn 2001) and from the Wismar Bay (spring 2001), in perch from the Lithuanian coast near Nemirseta (autumn 2001) and in eelpout from the Wismar Bay (autumn 2001). These values are 510-fold higher than the baseline level of micronucleus incidence. In general, the decrease in environmental mutagenesis was observed in 2002. Nevertheless a significant increase in micronucleus levels was determined in fish after the oil spill in the Bûtingë oil terminal

  • Effects of algal extracts (Polysiphonia fucoides) on rainbow trout (Oncorhynchus mykiss): A biomarker approach
  • 2006
  • Ingår i: Marine Environmental Research. - : Elsevier BV. - 0141-1136. ; 62:Supplement 1
  • Tidskriftsartikel (refereegranskat)abstract
    • The genotoxicity of algal extracts (Polysiphonia fucoides) was investigated in erythrocytes of rainbow trout (Oncorhynchus mykiss). Trout were exposed to 0.5% of the algal extract for 7 days. Comet assay (alkaline and neutral versions) and Micronucleus test were used to assess DNA damage, and Diffusion Assay to detect apoptotic cells. EROD activities and oxidative stress parameters in rainbow trout liver were also measured. A significant induction of DNA single strand breaks comparable to the ones induced by the in vivo exposure to 20 mg/kg B[a]P was observed at the end of the treatment, while increases of double strand breaks and apoptotic cells were not observed. The absence of activation of antioxidant responses seems to underline a mechanism of action of the genotoxic algal extract which does not involve oxidative stress.

  • Adult female European perch (Perca fluviatilis) from the Baltic Sea show no evidence of thiamine deficiency
  • 2021
  • Ingår i: Journal of Sea Research. - : Elsevier BV. - 1385-1101 .- 1873-1414. ; 174
  • Tidskriftsartikel (refereegranskat)abstract
    • Deficiency of thiamine (vitamin B1) has been demonstrated in several species in the northern hemisphere and is suggested as a cause for declining populations. European perch from the Baltic Sea show negative temporal trends for several health biomarkers and poor recruitment of unknown cause. In this study, thiamine status of perch liver from the Baltic Sea was studied with emphasis on seasonal variation. During spring the thiamine concentration increased, reached a higher level during the summer and then decreased again during autumn. Despite this variation the thiamine concentration was always sufficient in the perch liver. These results indicate that direct thiamine deficiency is an unlikely explanation for the health effects observed in adult female perch from the Baltic Sea.

  • Correlating seasonal changes of naturally produced brominated compounds to biomarkers in perch from the Baltic Sea
  • 2021
  • Ingår i: Aquatic Toxicology. - : Elsevier BV. - 0166-445X .- 1879-1514. ; 240
  • Tidskriftsartikel (refereegranskat)abstract
    • Hydroxylated polybrominated diphenyl ethers (OH-PBDEs), naturally produced by algae and cyanobacteria in the Baltic Sea, are potent disrupters of energy metabolism as well as endocrine disruptors and neurotoxins. In this study, European perch (Perca fluviatilis) from the Baltic Sea were sampled from May until October. OH-PBDEs and ten biomarkers were measured in each individual (n = 84 over 18 sampling time points) to study potential correlations between exposure to OH-PBDEs and changes in biomarkers. Several biomarkers showed significant non-linear seasonal variation. In the perch, ethoxyresorufin-O-deethylase (EROD) activity, plasma lactate concentration, and plasma glucose concentration showed a significant positive log-linear correlation with OH-PBDEs, whereas lipid percentage and liver somatic index showed a significant negative log-linear correlation with OH-PBDEs. These results strengthen the concern that OH-PBDEs could cause negative health effects for fish in the Baltic Sea.
