    • The almost unlimited design freedom of the laser-based powder bed fusion of metals (PBF-LB/M) makes this technology very attractive for industry. However, as a developing technology, it still faces some challenges when it comes to productivity and robustness, to name some. Whereas numerous studies covered the impact of laser-based parameters on material properties and robustness, the effect of the processing gas received limited attention. The objective of this study was to evaluate the effect of processing gas composition, containing helium (He) and hydrogen (H2), compared to conventionally used argon (Ar), during PBF-LB/M processing of virgin alloy 718 powder, on printing behavior and part properties. The four gases studied were Ar, Ar +30%He, Ar +30%He +2%H2, and Ar +70%He. Optical Tomography (OT) was used to monitor process stability, which unveiled a significant decrease in process-by products (spatters) between 51 % and 89 % using He and H2-containing gases. It was also found that the process gas decreased the bulk porosity from an average value of 0.08 % when processed with Ar to 0.04 % when using Ar + 70%He. The oxygen pickup by the spatter particles was reduced from 630 ppm (Ar) to 331 ppm (Ar +70%He). EBSD analysis revealed that there were no evident changes in microstructure with the processing gas. The samples processed also had similar tensile properties with yield and ultimate tensile strength of 1180 MPa and 1395 MPa, respectively. However, there was a slight increase in ductility from 16.5 % to 17.2 %, when processed with pure Ar and Ar + 70%He, respectively. This study shows that utilizing standard Ar processing atmosphere with He addition leads to a more stable process with reduced spatter, porosity and a marginal increase in ductility for Alloy 718.

    • We report here genome sequences and comparative analyses of three closely related parasitoid wasps: Nasonia vitripennis, N. giraulti, and N. longicornis. Parasitoids are important regulators of arthropod populations, including major agricultural pests and disease vectors, and Nasonia is an emerging genetic model, particularly for evolutionary and developmental genetics. Key findings include the identification of a functional DNA methylation tool kit; hymenopteran-specific genes including diverse venoms; lateral gene transfers among Pox viruses, Wolbachia, and Nasonia; and the rapid evolution of genes involved in nuclear-mitochondrial interactions that are implicated in speciation. Newly developed genome resources advance Nasonia for genetic research, accelerate mapping and cloning of quantitative trait loci, and will ultimately provide tools and knowledge for further increasing the utility of parasitoids as pest insect-control agents.

Can the process gas play a key role in optimizing the PBF-LB/M process (layer thickness, scan speed, processability of new materials, etc.)? This article provide insights into the current state of research at Linde GmbH regarding he-lium-containing process gases and presenting the novel process gas ADDvance® Laser230. Due to its composition, the gas mixture allows to significantly improve process productivity and stability.

    • The effect of the process gas during laser powder bed fusion (L-PBF) was investigated using high-speed shadowgraphy while melting Ti-6Al-4V powder under high purity argon, helium, and a mixture of both, on a laboratory-scale machine. These recordings reveal that the generation of incandescent spatters can be reduced by at least 60% using pure helium and by ∼30% using addition of helium to argon in comparison to the use of traditional argon. The quantity of colder spatters appeared unaffected by the change of process gas. Different configurations of gas flow versus laser scanning direction were investigated and revealed that fumes and spatters are less accumulated at the laser spot with helium addition. Furthermore, the use of the argon–helium mixture proved to be as efficient as pure argon in the dragging and extraction of the fumes. Shadowgraphs revealed the more rapid expansion of fumes in helium-rich atmospheres, limiting the accumulation of scattering objects close to the laser spot and thus melt pool instability. These results were correlated to process snapshots on an industrial-scale system, confirming the reduction of hot spatter generation. Finally, the findings put in evidence the more rapid cooling of spatters with helium addition to the process gas – a promising aspect to limit powder bed degradation during L-PBF. In addition, the use of mixtures of helium and argon would be economically interesting compared to pure helium, typically more expensive than the traditionally used argon.

    • MicroRNAs (miRNAs) are crucial post-transcriptional regulators that have been extensively studied in Bilateria, a group comprising the majority of extant animals, where more than 30 conserved miRNA families have been identified. By contrast, bilaterian miRNA targets are largely not conserved. Cnidaria is the sister group to Bilateria and thus provides a unique opportunity for comparative studies. Strikingly, like their plant counterparts, cnidarian miRNAs have been shown to predominantly have highly complementary targets leading to transcript cleavage by Argonaute proteins. Here, we assess the conservation of miRNAs and their targets by small RNA sequencing followed by miRNA target prediction in eight species of Anthozoa (sea anemones and corals), the earliest-branching cnidarian class. We uncover dozens of novel miRNAs but only a few conserved ones. Further, given their high complementarity, we were able to computationally identify miRNA targets in each species. Besides evidence for conservation of specific miRNA target sites, which are maintained between sea anemones and stony corals across 500 Myr of evolution, we also find indications for convergent evolution of target regulation by different miRNAs. Our data indicate that cnidarians have only few conserved miRNAs and corresponding targets, despite their high complementarity, suggesting a high evolutionary turnover.

    • Tribolium castaneum is a representative of earth’s most numerous eukaryotic order, a powerful model organism for the study of generalized insect development, and also an important pest of stored agricultural products. We describe its genome sequence here. This omnivorous beetle has evolved an ability to interact with a diverse chemical environment as evidenced by large expansions in odorant and gustatory receptors, as well as p450 and other detoxification enzymes. Developmental patterns in Tribolium are more representative of other arthropods than those found in Drosophila, a fact represented in gene content and function. For one, Tribolium has retained more ancestral genes involved in cell-cell communication than Drosophila, and some are expressed in the growth zone crucial for axial elongation in short germ development. Systemic RNAi in T. castaneum appears to use mechanisms distinct from those found in C. elegans, but nevertheless offers similar power for the elucidation of gene function and identification of targets for selective insect control.
