    • A healthy intestinal tract is characterized by controlled homeostasis due to the balanced interaction between commensal bacteria and the host mucosal immune system. Human and animal model studies have supported the hypothesis that breakdown of this homeostasis may underlie the pathogenesis of inflammatory bowel diseases. However, it is not well understood how intestinal microflora stimulate the intestinal mucosal immune system and how such activation is regulated. Using a spontaneous, commensal bacteria-dependent colitis model in IL-10-deficient mice, we investigated the role of TLR and their negative regulation in intestinal homeostasis. In addition to IL-10(-/-) MyD88(-/-) mice, IL-10(-/-) TLR4(-/-) mice exhibited reduced colitis compared to IL-10(-/-) mice, indicating that TLR4 signaling plays an important role in inducing colitis. Interestingly, the expression of IRAK-M, a negative regulator of TLR signaling, is dependent on intestinal commensal flora, as IRAK-M expression was reduced in mice re-derived into a germ-free environment, and introduction of commensal bacteria into germ-free mice induced IRAK-M expression. IL-10(-/-) IRAK-M(-/-) mice exhibited exacerbated colitis with increased inflammatory cytokine gene expression. Therefore, this study indicates that intestinal microflora stimulate the colitogenic immune system through TLR and negative regulation of TLR signaling is essential in maintaining intestinal homeostasis.

    • “Fish eye” is a typical phenomenon of fatigue crack initiation at a subsurface defect such as inclusion during very high cycle fatigue. The formation of a fine grained area and micro-debonding is believed to cause fatigue crack initiation. This paper provides a basic study on the formation of the fine grained area in a martensitic stainless steel during very high cycle fatigue using scanning electron microscopy, SEM, focused ion beam technique, FIB, electron backscatter diffraction, EBSD, and electron channeling contrast imaging, ECCI. The results show that the formation of a fine grained zone is a local behavior. The fine grained zone is very near the fatigue crack initiation origin. In the transversal direction (cross section), the depth of the fine grained zone is only few micrometers. In the longitudinal direction (crack propagation direction), the depth of the fine grain zone is about one micrometer. ECCI analysis shows that in the fine grained area with high retained strain, high plastic deformation can be found. Dislocation slip bands can be observed. They interact with grain boundaries and cause the formation of damage due to impingement cracking. The results indicate that occurrence of plastic deformation in metallic material during very high cycle fatigue is very localized, mainly near the front of the crack tip or a defect.

    • This is an example how of our research and development work regarding a self-developed method to synchronise a video recorder and a personal computer was refined further. Which in turn gave birth to an extensive register of video-registration that still are saved at Chalmers University of Technology (this register’s existence has not been resolved after Engström’s retirement). This method in question has more or less been nicked by other scientists, which who we have cooperated at Lindholmen Utveckling in Gothenburg. Who has constructed a digitalised version of this method, our own equipment comprised usage of VHS-tapes (see other publication registered in Chalmers Public Library CPL). Later on refined further with regard to specific psychological aspects (i.e. the registered operator/subjects were thereby able to self-evaluate the recordings by interactions).
