  • Transit detection of the long-period volatile-rich super-Earth nu(2) Lupi d with CHEOPS

    • Exoplanets transiting bright nearby stars are key objects for advancing our knowledge of planetary formation and evolution. The wealth of photons from the host star gives detailed access to the atmospheric, interior and orbital properties of the planetary companions. nu(2) Lupi (HD 136352) is a naked-eye (V = 5.78) Sun-like star that was discovered to host three low-mass planets with orbital periods of 11.6, 27.6 and 107.6 d via radial-velocity monitoring(1). The two inner planets (b and c) were recently found to transit(2), prompting a photometric follow-up by the brand new Characterising Exoplanets Satellite (CHEOPS). Here, we report that the outer planet d is also transiting, and measure its radius and mass to be 2.56 +/- 0.09 R-circle plus and 8.82 +/- 0.94 M-circle plus, respectively. With its bright Sun-like star, long period and mild irradiation (similar to 5.7 times the irradiation of Earth), nu(2) Lupi d unlocks a completely new region in the parameter space of exoplanets amenable to detailed characterization. We refine the properties of all three planets: planet b probably has a rocky mostly dry composition, while planets c and d seem to have retained small hydrogen-helium envelopes and a possibly large water fraction. This diversity of planetary compositions makes the nu(2) Lupi system an excellent laboratory for testing formation and evolution models of low-mass planets.

  • Rapid Profiling of Protein Complex Reorganization in Perturbed Systems

    • Protein complexes constitute the primary functional modules of cellular activity. To respond to perturbations, complexes undergo changes in their abundance, subunit composition, or state of modification. Understanding the function of biological systems requires global strategies to capture this contextual state information. Methods based on cofractionation paired with mass spectrometry have demonstrated the capability for deep biological insight, but the scope of studies using this approach has been limited by the large measurement time per biological sample and challenges with data analysis. There has been little uptake of this strategy into the broader life science community despite its rich biological information content. We present a rapid integrated experimental and computational workflow to assess the reorganization of protein complexes across multiple cellular states. The workflow combines short gradient chromatography and DIA/SWATH mass spectrometry with a data analysis toolset to quantify changes in a complex organization. We applied the workflow to study the global protein complex rearrangements of THP-1 cells undergoing monocyte to macrophage differentiation and subsequent stimulation of macrophage cells with lipopolysaccharide. We observed substantial proteome reorganization on differentiation and less pronounced changes in macrophage stimulation. We establish our integrated differential pipeline for rapid and state-specific profiling of protein complex organization.

  • Psychopathy dimensions, Big Five traits, and dispositional aggression in adolescence : Issues of gender consistency

    • In order to evaluate the nomological network of associations between psychopathy and its sub-dimensions, and Big Five domains and dispositional aggression in adolescence, and its consistency across gender-based sub-groups, 1253 Italian high school students (F = 429, M 824) were administered the Italian translations of the Youth Psychopathic Traits Inventory (YPI), Big Five Inventory (BFI) and the Reactive-Proactive Aggression Questionnaire. Males scored on average significantly higher than females on all measures of psychopathy and aggression, whereas females scored significantly higher than males on the BFI O, C, A and N scales. Bivariate correlations between psychopathy scores, and aggression and BFI domains measures were highly similar in females and males. The majority of regression coefficients based on hierarchical regression models were consistently replicated across gender-based groups; effect size estimates for regression models were large, supporting the hypothesis that psychopathy can be described in terms of general personality traits in adolescence and that the relationships between psychopathy, its sub-dimensions and Big Five personality dimensions, and aggression generally are very similar across gender.   • PCprophet : a framework for protein complex prediction and differential analysis using proteomic data

    • Despite the availability of methods for analyzing protein complexes, systematic analysis of complexes under multiple conditions remains challenging. Approaches based on biochemical fractionation of intact, native complexes and correlation of protein profiles have shown promise. However, most approaches for interpreting cofractionation datasets to yield complex composition and rearrangements between samples depend considerably on protein–protein interaction inference. We introduce PCprophet, a toolkit built on size exclusion chromatography–sequential window acquisition of all theoretical mass spectrometry (SEC-SWATH-MS) data to predict protein complexes and characterize their changes across experimental conditions. We demonstrate improved performance of PCprophet over state-of-the-art approaches and introduce a Bayesian approach to analyze altered protein–protein interactions across conditions. We provide both command-line and graphical interfaces to support the application of PCprophet to any cofractionation MS dataset, independent of separation or quantitative liquid chromatography–MS workflow, for the detection and quantitative tracking of protein complexes and their physiological dynamics.

    • To assess the internal consistency, factor structure, and construct validity of the Italian translation of the Youth Psychopathic Traits Inventory-Short Version (YPI-S), both the YPI-S and its full version, the YPI, as well as self-reports of delinquency, aggression, and Big Five domains, were administered to two independent samples (N = 868 and N = 881) of Italian community, nonreferred adolescents. The internal consistency of the YPI-S was adequate, and confirmatory factor analyses showed a good fit of the theoretical three-factor model of the YPI-S in both samples. Hierarchical regression models suggested the same pattern of associations with self-report measures of delinquency and aggression for the YPI-S and YPI, although the YPI was a better predictor of Big Five domains than the YPI-S. The findings support the internal consistency, factor validity, and construct validity of the YPI-S.

    • INTRODUCTION: Endometrial cancer patients with high grade tumours, deep myometrial invasion or advanced stage disease have a poor prognosis. Randomised studies have demonstrated the prevention of loco-regional relapses with radiotherapy (RT) with no effect on overall survival (OS). The possible additive effect of chemotherapy (CT) remains unclear. Two randomised clinical trials (NSGO-EC-9501/EORTC-55991 and MaNGO ILIADE-III) were undertaken to clarify if sequential combination of chemotherapy and radiotherapy improves progression-free survival (PFS) in high-risk endometrial cancer. The two studies were pooled. METHODS: Patients (n=540; 534 evaluable) with operated endometrial cancer International Federation of Obstetrics and Gynaecology (FIGO) stage I-III with no residual tumour and prognostic factors implying high-risk were randomly allocated to adjuvant radiotherapy with or without sequential chemotherapy. RESULTS: In the NSGO/EORTC study, the combined modality treatment was associated with 36% reduction in the risk for relapse or death (hazard ratio (HR) 0.64, 95%confidence interval (CI) 0.41-0.99; P=0.04); two-sided tests were used. The result from the Gynaecologic Oncology group at the Mario Negri Institute (MaNGO)-study pointed in the same direction (HR 0.61), but was not significant. In the combined analysis, the estimate of risk for relapse or death was similar but with narrower confidence limits (HR 0.63, CI 0.44-0.89; P=0.009). Neither study showed significant differences in the overall survival. In the combined analysis, overall survival approached statistical significance (HR 0.69, CI 0.46-1.03; P=0.07) and cancer-specific survival (CSS) was significant (HR 0.55, CI 0.35-0.88; P=0.01). CONCLUSION: Addition of adjuvant chemotherapy to radiation improves progression-free survival in operated endometrial cancer patients with no residual tumour and a high-risk profile. A remaining question for future studies is if addition of radiotherapy to chemotherapy improves the results.

    • The aim of the present study was to investigate if interaction effects among Youth Psychopathic Traits Inventory (YPI) Grandiose-Manipulative (GM), Callous-Unemotional (CU), and Impulsive-Irresponsible (II) may add significant information in predicting self-reports of juvenile delinquent behavior among adolescents. A sample of 558 Italian high school students were administered the YPI and the Self-Reports of Delinquency Scale (SRDS) in order to evaluate interaction effects among YPI GM, CU, and II dimensions. Results showed a significant effect for a three-way interaction among the three YPI dimensions in predicting the SRDS total score, beta = .19, p < .01. In conclusion, the findings seem to underline that a three-factor model of psychopathy may help clinicians and researchers in predicting self-reported delinquency better than the individual factors.

  • The Validity of the Child Problematic Trait Inventory in 6–12 Year Old Italian Children : Further Support and Issues of Consistency Across Different Sources of Information and Different Samples

    • In the present study, teacher’s ratings, mother’s ratings, and father’s ratings on the Child Problematic Trait Inventory (CPTI) were obtained on two independent samples (N = 381 and N = 406, respectively) of Italian school age children (age range: 6–12 years). Cronbach alpha values greater than 0.80 were observed for all CPTI scale scores based on teacher’s ratings, mother’s ratings, and father’s ratings respectively. Although intraclass correlation values suggested poor parent-teacher agreement, robust weighted least square Confirmatory Factor Analyses (WLSMV CFAs) suggested that the three-factor model of the CPTI was consistently replicated as the best fitting model across all informants. Logistic regression analysis conducted in one of the samples showed that teacher’s ratings and mother’s ratings of the CPTI, but not CPTI father’s ratings, significantly predicted child’s problem behavior at school. Multiple regression analyses showed that CPTI scores were significantly predicted by selected temperament dimensions, but with different relationships for different sources of information. Overall, our findings suggest that the CPTI is a reliable and valid measure of problematic traits with a three-factor structure, although this study indicates that multiple sources of information may prove necessary when assessing psychopathy in school-age children with the CPTI.

  • Longevity interventions modulate mechanotransduction and extracellular matrix homeostasis in C. elegans

    • Dysfunctional extracellular matrices (ECM) contribute to aging and disease. Repairing dysfunctional ECM could potentially prevent age-related pathologies. Interventions promoting longevity also impact ECM gene expression. However, the role of ECM composition changes in healthy aging remains unclear. Here we perform proteomics and in-vivo monitoring to systematically investigate ECM composition (matreotype) during aging in C. elegans revealing three distinct collagen dynamics. Longevity interventions slow age-related collagen stiffening and prolong the expression of collagens that are turned over. These prolonged collagen dynamics are mediated by a mechanical feedback loop of hemidesmosome-containing structures that span from the exoskeletal ECM through the hypodermis, basement membrane ECM, to the muscles, coupling mechanical forces to adjust ECM gene expression and longevity via the transcriptional co-activator YAP-1 across tissues. Our results provide in-vivo evidence that coordinated ECM remodeling through mechanotransduction is required and sufficient to promote longevity, offering potential avenues for interventions targeting ECM dynamics.
