  • Whole grain-rich diet reduces body weight and systemic low-grade inflammation without inducing major changes of the gut microbiome: A randomised cross-over trial

    • Objective T o investigate whether a whole grain diet alters the gut microbiome and insulin sensitivity, as well as biomarkers of metabolic health and gut functionality. Design 60 Danish adults at risk of developing metabolic syndrome were included in a randomised cross-over trial with two 8-week dietary intervention periods comprising whole grain diet and refined grain diet, separated by a washout period of =6 weeks. The response to the interventions on the gut microbiome composition and insulin sensitivity as well on measures of glucose and lipid metabolism, gut functionality, inflammatory markers, anthropometry and urine metabolomics were assessed. Results 50 participants completed both periods with a whole grain intake of 179±50 g/day and 13±10 g/day in the whole grain and refined grain period, respectively. Compliance was confirmed by a difference in plasma alkylresorcinols (p<0.0001). Compared with refined grain, whole grain did not significantly alter glucose homeostasis and did not induce major changes in the faecal microbiome. Also, breath hydrogen levels, plasma short-chain fatty acids, intestinal integrity and intestinal transit time were not affected. The whole grain diet did, however, compared with the refined grain diet, decrease body weight (p<0.0001), serum inflammatory markers, interleukin (IL)-6 (p=0.009) and C-reactive protein (p=0.003). The reduction in body weight was consistent with a reduction in energy intake, and IL-6 reduction was associated with the amount of whole grain consumed, in particular with intake of rye. Conclusion C ompared with refined grain diet, whole grain diet did not alter insulin sensitivity and gut microbiome but reduced body weight and systemic lowgrade inflammation.

    • © 2018, The Author(s). Adherence to a low-gluten diet has become increasingly common in parts of the general population. However, the effects of reducing gluten-rich food items including wheat, barley and rye cereals in healthy adults are unclear. Here, we undertook a randomised, controlled, cross-over trial involving 60 middle-aged Danish adults without known disorders with two 8-week interventions comparing a low-gluten diet (2 g gluten per day) and a high-gluten diet (18 g gluten per day), separated by a washout period of at least six weeks with habitual diet (12 g gluten per day). We find that, in comparison with a high-gluten diet, a low-gluten diet induces moderate changes in the intestinal microbiome, reduces fasting and postprandial hydrogen exhalation, and leads to improvements in self-reported bloating. These observations suggest that most of the effects of a low-gluten diet in non-coeliac adults may be driven by qualitative changes in dietary fibres.

  • Local and long-range atomic/magnetic structure of non-stoichiometric spinel iron oxide nanocrystallites

    • Spinel iron oxide nanoparticles of different mean sizes in the range 10-25 nm have been prepared by surfactant-free up-scalable near- and super-critical hydrothermal synthesis pathways and characterized using a wide range of advanced structural characterization methods to provide a highly detailed structural description. The atomic structure is examined by combined Rietveld analysis of synchrotron powder X-ray diffraction (PXRD) data and time-of-flight neutron powder-diffraction (NPD) data. The local atomic ordering is further analysed by pair distribution function (PDF) analysis of both X-ray and neutron total-scattering data. It is observed that a non-stoichiometric structural model based on a tetragonal γ-Fe2O3 phase with vacancy ordering in the structure (space group P43212) yields the best fit to the PXRD and total-scattering data. Detailed peak-profile analysis reveals a shorter coherence length for the superstructure, which may be attributed to the vacancy-ordered domains being smaller than the size of the crystallites and/or the presence of anti-phase boundaries, faulting or other disorder effects. The intermediate stoichiometry between that of γ-Fe2O3 and Fe3O4 is confirmed by refinement of the Fe/O stoichiometry in the scattering data and quantitative analysis of Mössbauer spectra. The structural characterization is complemented by nano/micro-structural analysis using transmission electron microscopy (TEM), elemental mapping using scanning TEM, energy-dispersive X-ray spectroscopy and the measurement of macroscopic magnetic properties using vibrating sample magnetometry. Notably, no evidence is found of a Fe3O4/γ-Fe2O3 core-shell nanostructure being present, which had previously been suggested for non-stoichiometric spinel iron oxide nanoparticles. Finally, the study is concluded using the magnetic PDF (mPDF) method to model the neutron total-scattering data and determine the local magnetic ordering and magnetic domain sizes in the iron oxide nanoparticles. The mPDF data analysis reveals ferrimagnetic collinear ordering of the spins in the structure and the magnetic domain sizes to be ∼60-70% of the total nanoparticle sizes. The present study is the first in which mPDF analysis has been applied to magnetic nanoparticles, establishing a successful precedent for future studies of magnetic nanoparticles using this technique.

  • Bifidobacterium species associated with breastfeeding produce aromatic lactic acids in the infant gut

    • Breastfeeding profoundly shapes the infant gut microbiota, which is critical for early life immune development, and the gut microbiota can impact host physiology in various ways, such as through the production of metabolites. However, few breastmilk-dependent microbial metabolites mediating host–microbiota interactions are currently known. Here, we demonstrate that breastmilk-promoted Bifidobacterium species convert aromatic amino acids (tryptophan, phenylalanine and tyrosine) into their respective aromatic lactic acids (indolelactic acid, phenyllactic acid and 4-hydroxyphenyllactic acid) via a previously unrecognized aromatic lactate dehydrogenase (ALDH). The ability of Bifidobacterium species to convert aromatic amino acids to their lactic acid derivatives was confirmed using monocolonized mice. Longitudinal profiling of the faecal microbiota composition and metabolome of Danish infants (n = 25), from birth until 6 months of age, showed that faecal concentrations of aromatic lactic acids are correlated positively with the abundance of human milk oligosaccharide-degrading Bifidobacterium species containing the ALDH, including Bifidobacterium longum, B. breve and B. bifidum. We further demonstrate that faecal concentrations of Bifidobacterium-derived indolelactic acid are associated with the capacity of these samples to activate in vitro the aryl hydrocarbon receptor (AhR), a receptor important for controlling intestinal homoeostasis and immune responses. Finally, we show that indolelactic acid modulates ex vivo immune responses of human CD4+ T cells and monocytes in a dose-dependent manner by acting as an agonist of both the AhR and hydroxycarboxylic acid receptor 3 (HCA3). Our findings reveal that breastmilk-promoted Bifidobacterium species produce aromatic lactic acids in the gut of infants and suggest that these microbial metabolites may impact immune function in early life.

    • Solid oxide cells are an exciting technology for energy conversion. Fuel cells, based on solid oxide technology, convert hydrogen or hydrogen-rich fuels into electrical energy, with potential applications in stationary power generation. Conversely, solid oxide electrolysers convert electricity into chemical energy, thereby offering the potential to store energy from transient resources, such as wind turbines and other renewable technologies. For solid oxide cells to displace conventional energy conversion devices in the marketplace, reliability must be improved, product lifecycles extended, and unit costs reduced. Mathematical models can provide qualitative and quantitative insight into physical phenomena and performance, over a range of length and time scales. The purpose of this paper is to provide the reader with a summary of the state-of-the art of solid oxide cell models. These range from: simple methods based on lumped parameters with little or no kinetics to detailed, time-dependent, three-dimensional solutions for electric field potentials, complex chemical kinetics and fully-comprehensive equations of motion based on effective transport properties. Many mathematical models have, in the past, been based on inaccurate property values obtained from the literature, as well as over-simplistic schemes to compute effective values. It is important to be aware of the underlying experimental methods available to parameterise mathematical models, as well as validate results. In this article, state-of-the-art techniques for measuring kinetic, electric and transport properties are also described. Methods such as electrochemical impedance spectroscopy allow for fundamental physicochemical parameters to be obtained. In addition, effective properties may be obtained using micro-scale computer simulations based on digital reconstruction obtained from X-ray tomography/focussed ion beam scanning electron microscopy, as well as percolation theory. The cornerstone of model validation, namely the polarisation or current-voltage diagram, provides necessary, but insufficient information to substantiate the reliability of detailed model calculations. The results of physical experiments which precisely mimic the details of model conditions are scarce, and it is fair to say there is a gap between the two activities. The purpose of this review is to introduce the reader to the current state-of-the art of solid oxide analysis techniques, in a tutorial fashion, not only numerical and but also experimental, and to emphasise the cross-linkages between techniques.

  • Dense sampling of bird diversity increases power of comparative genomics

    • Whole-genome sequencing projects are increasingly populating the tree of life and characterizing biodiversity(1-4). Sparse taxon sampling has previously been proposed to confound phylogenetic inference(5), and captures only a fraction of the genomic diversity. Here we report a substantial step towards the dense representation of avian phylogenetic and molecular diversity, by analysing 363 genomes from 92.4% of bird families-including 267 newly sequenced genomes produced for phase II of the Bird 10,000 Genomes (B10K) Project. We use this comparative genome dataset in combination with a pipeline that leverages a reference-free whole-genome alignment to identify orthologous regions in greater numbers than has previously been possible and to recognize genomic novelties in particular bird lineages. The densely sampled alignment provides a single-base-pair map of selection, has more than doubled the fraction of bases that are confidently predicted to be under conservation and reveals extensive patterns of weak selection in predominantly non-coding DNA. Our results demonstrate that increasing the diversity of genomes used in comparative studies can reveal more shared and lineage-specific variation, and improve the investigation of genomic characteristics. We anticipate that this genomic resource will offer new perspectives on evolutionary processes in cross-species comparative analyses and assist in efforts to conserve species. A dataset of the genomes of 363 species from the Bird 10,000 Genomes Project shows increased power to detect shared and lineage-specific variation, demonstrating the importance of phylogenetically diverse taxon sampling in whole-genome sequencing.

  • Effect of stress on NiO reduction in solid oxide fuel cells: a new application of energy-resolved neutron imaging

    • Recently, two new phenomena linking stress field and reduction rates in anode-supported solid oxide fuel cells (SOFCs) have been demonstrated, so-called accelerated creep during reduction and reduction rate enhancement and nucleation due to stress (Frandsen et al., 2014). These complex phenomena are difficult to study and it is demonstrated here that energy-resolved neutron imaging is a feasible technique for combined mechanics-chemical composition studies of SOFC components, including commercially produced ones. Cermet anode supports, which prior to the measurements were reduced under varying conditions such as different temperatures, various times and different values of applied stress, have been measured. Thus, samples with different contents (and gradients) of Ni and NiO phases were investigated. The first Bragg edge transmission neutron measurements applied for the studies of the reduction progress in these samples were performed at two neutron beamline facilities (ISIS in the UK, Helmholtz Zentrum Berlin in Germany). The obtained results demonstrate the possibility to image and distinguish NiO and Ni phases within SOFC anode supports by energy-resolved neutron imaging and the potential of the neutron imaging method for in situ studies of reduction processes.

    • This article reports on in situ macroscopic scale imaging of NiO-YSZ (YSZ is yttria-stabilized zirconia) reduction under applied stress - a phase transition taking place in solid oxide electrochemical cells in a reducing atmosphere of a hydrogen/nitrogen mixture and at operation temperatures of up to 1073K. This process is critical for the performance and lifetime of the cells. Energy-resolved neutron imaging was applied to observe the phase transition directly with time and spatial resolution. Two different approaches are presented for using this imaging technique for the investigation of chemical and physical processes requiring controlled atmosphere and elevated temperature. The first type of measurement is based on alternating stages of short-term partial chemical reaction and longer neutron image acquisition, and the second type is a real in situ neutron imaging experiment. Results of applying energy-resolved neutron imaging with both approaches to the NiO-YSZ reduction investigation indicate enhancement of the reduction rate due to applied stress, which is consistent with the results of the authors' previous research.Results of the first in situ energy-resolved neutron imaging of NiO-YSZ (YSZ is yttria-stabilized zirconia) reduction under applied stress are presented. Neutron experiments were performed at the pulsed neutron source ISIS (UK) using a time-of-flight approach.
