    • The focus of this paper is to show that the E-C-A paradigm offers an excellent approach for specifying the behavior of context-aware information push services. Such a service enables its operator to provide the users with tailored messages related to their current situation (context). The paper introduces CAIPS, an implementation of such a service for the tourism domain. The underlying E-C-A rules are presented and the design of the associated rule-engine is described. The engine’s rule-interpreter is based on event-notification services and the object-oriented query-language HQL. The paper further presents a graphical high-level editor which supports business-experts in “writing” the CAIPS E-C-A rules. The presented approach enables the rapid development of new tailored messages (related to the user’s context) without the need to modify the underlying application, i.e. without the trouble of writing new code for new message types.

  • Modification of a High-Throughput Automatic Microbial Cell Enumeration System for Shipboard Analyses

    • In the age of ever-increasing "-omics" studies, the accurate and statistically robust determination of microbial cell numbers within often-complex samples remains a key task in microbial ecology. Microscopic quantification is still the only method to enumerate specific subgroups of microbial clades within complex communities by, for example, fluorescence in situ hybridization (FISH). In this study, we improved an existing automatic image acquisition and cell enumeration system and adapted it for usage at high seas on board an oceanographic research ship. The system was evaluated by testing settings such as minimal pixel area and image exposure times ashore under stable laboratory conditions before being brought on board and tested under various wind and wave conditions. The system was robust enough to produce high-quality images even with ship heaves of up to 3m and pitch and roll angles of up to 6.3 degrees. On board the research ship, on average, 25% of the images acquired from plankton samples on filter membranes could be used for cell enumeration. Automated enumeration was highly correlated with manual counts (r(2)>0.9). Even the smallest of microbial cells in the open ocean, members of the alphaproteobacterial SAR11 clade, could be confidently detected and enumerated. The automated image acquisition and cell enumeration system developed here enables an accurate and reproducible determination of microbial cell counts in planktonic samples and allows insight into the abundance and distribution of specific microorganisms already on board within a few hours.IMPORTANCE In this research article, we report on a new system and software pipeline, which allows for an easy and quick image acquisition and the subsequent enumeration of cells in the acquired images. We put this pipeline through vigorous testing and compared it to manual microscopy counts of microbial cells on membrane filters. Furthermore, we tested this system at sea on board a marine research vessel and counted bacteria on board within a few hours after the retrieval of water samples. The imaging and counting system described here has been successfully applied to a number of laboratory-based studies and allowed the quantification of thousands of samples and FISH preparations (see, e.g., H. Amann, Science 336:608-611, 2012, http://dx.doi.org/10.1126/science.1218344). We adjusted the standard image acquisition software to withstand ship movements. This system will allow for more targeted sampling of the microbial community, leading to a better understanding of the role of microorganisms in the global oceans.

    • BACKGROUND: Response (tumour volume reduction) to induction chemotherapy has been used to stratify secondary local and systemic treatment of Intergroup Rhabdomyosarcoma Study Group III (IRSG-III) embryonal rhabdomyosarcoma (RME) in consecutive CWS-trials. To evaluate its actual impact we studied response-related treatment and outcomes.PROCEDURE: Patients with IRSG-III RME <21 years and non-response (NR, <33% volume reduction) in five consecutive CWS-trials were analysed and compared with partial responders (PAR, ≥33% reduction). The NR was reviewed and sub-classified as Objective Response (OR, <0%-33% reduction) or Stable/Progressive Disease (SPD).RESULTS: Fifty-nine of 529 patients had NR (n = 34 OR, n = 25 SPD). Primary risk-factors including age, tumour size, and TN-classification did not differ between NR and PAR groups but NR had more patients with unfavourable sites comparatively (P = 0.04). There were no differences in primary risk-factors between OR and SPD. Significant factors associated with poor outcome in multivariate analysis were NR, TN-classification, age >10 years, tumour size >5 cm and therapy in older trials. After response assessment n = 24 NR continued to receive induction chemotherapy, n = 32 received other combinations and n = 3 no further chemotherapy. Forty-two non-responders were irradiated, and the tumours were completely resected in n = 20. After a median follow-up of 8 years, 34 NR are alive. Seventeen of 21 failures leading to disease-related deaths were locoregional. The five-year overall survival rate (OS) was 76 ± 4% for PAR, 79 ± 14% for OR, but only 40 ± 19% for SPD (P < 0.001).CONCLUSION: Response to induction chemotherapy appears to be an important surrogate marker of poor outcome in patients with SPD largely due to ineffective local control.

    • Mobile information services show great potential both as an on-site information source for customers and as a communication and distribution channel for tourism providers. Although in nontourism domains the adoption rate of using mobile technologies has been growing exponentially, comparable figures are only moderate in travel and tourism. Moreover, relatively little research has been conducted to understand travelers' behavioral intention to use mobile technologies. thus, the article proposes a technology acceptance model which is especially designed for mobile information services in the tourism domain. the model is an extension of the Unified theory of Acceptance and Use of technology, and is empirically tested through laboratory-based usage data obtained from users evaluating a mobile tourist guide developed for the ski resort DolomitiSuperski. Structural equation modeling (SeM) revealed that hedonic quality and social influence are main drivers of the behavioral intention to use mobile technologies in tourism, while monetary transparency and price fairness emerged as major usage barriers.

    • Masked trimethyl lock (TML) systems as molecular moieties enabling the bioresponsive release of compounds or dyes in a controlled temporal and spatial manner have been widely applied for the development of drug conjugates, prodrugs or molecular imaging tools. Herein, we report the development of a novel amino trimethyl lock (H2N-TML) system as an auto-immolative molecular entity for the release of fluorophores. We designed Cou-TML-N3 and MURh-TML-N3, two azide-masked turn-on fluorophores. The latter was demonstrated to selectively release fluorescent MURh in the presence of physiological concentrations of the redox-signaling molecule H2S in vitro and was successfully applied to image H2S in human cells.

    • Mobile information services are gradually supplementing and may in the future even replace traditional information sources to support tourists during on-site stay. The scope of the paper is to identify already in the pre-prototype phase types and functionalities of mobile information services which are most relevant to tourists during their urban destination stay (Davis & Venkatesh, 2004). For this aim a user-centred approach using, both, focus groups and expert surveys is proposed. In addition, multiple regression based on acceptance data gathered from 705 tourists allow the deduction of potentially accepted mobile information services targeted to the specific destination use. Finally, by drawing on these results, both, the architectural framework as well as the various building and implementation steps of a mobile tourist guide called innsbruck.mobile are discussed

    • As mobile tourist guides show huge potentials to support tourists’ information search, executives need to know which mobile systems characterized by which specific functionalities should be provided. The paper proposes an empirical approach combining qualitative and quantitative methods to deduce both, product definition and product marketing information related to mobile tourist guides, respectively. First, a methodology-mix combining qualitative (i.e. explorative survey, focus group interviews) and quantitative techniques (i.e. pilot study, expert interviews) responds to the question ‘Which services should be developed?’ Secondly, again in the pre-prototype phase and based on survey data (N = 705) ideal functionality compositions for promising tourist guides are deduced with regression analysis. Finally, based on technology acceptance theory (Venkatesh et al., 2003a) customer prejudices towards mobile tourist guides are identified. The obtained results support hypotheses indicating strong performance expectations and pronounced effort expectancies towards the deduced concepts designed to support information search by means of mobile tourist guides

    • Background: Low-grade fibromyxoid sarcoma (LGFMS) is a rare soft-tissue tumor with benign histologic appearance, though fully malignant behavior is possible.Methods: Patients with LGFMS<21 years registered in Cooperative Weichteilsarkom Studiengruppe trials until 2017 were analyzed. Firstline treatment consisted of complete surgical resection whenever possible.Results: Median age of 31 patients was 10.9 years (first month to 17.1 years). Twenty-six tumors were confirmed to the tissue of origin (T1), four invaded contiguous structures (T2), one was TX. Eight were >5 cm. The best surgical result was resection with free margins (R0) in 24 and microscopic residuals (R1) in seven. Five-year event-free (EFS), 5-year local-relapse-free (LRFS), and 5-year overall-survival were 71 +/- 18.6% confidence interval (CI) 95%, 76 +/- 17.6% CI 95%, and 100%, respectively. Six patients suffered local relapse in a median of 1 year, one combined within 1.3 year and one metastatic relapse with lesions in the lung, back muscles, and thigh discovered in whole-body imaging 6 years after the first diagnosis. In univariate analysis, T status correlated with EFS (T1 79.6 +/- 18.6%, T2 50.0 +/- 49.0%, P = .038). Resection with free margins tends to be associated with better LRFS (R0 82.4 +/- 18.6%, R1 53.6 +/- 39.4%, P = .053). Among 24 patients with R0 resection, five (21%) suffered relapse, thereof three local, one metastatic, and one combined. Among seven patients with R1-resection, three (43%) suffered local relapse.Conclusion Special caution is advisable in T2 tumors. The metastatic potential with lesions in unusual sites indicates that affected patients need to be informed. If long-term follow-up with whole-body imaging is beneficial, it may be addressed in larger intergroup analyses. Further research in disease biology is essential for optimal treatment and follow-up care.
